Abstract:As one of the blood-rich malignancies, the growth and metastasis of osteosarcoma both depend on its angiogenesis, a procedure in which vascular endothelial growth factor (VEGF) acts essentially. Although with the advent of neoadjuvant chemotherapy, more aggressive surgical excision and logical therapy strategy, the 5-year survival rate remains relatively stable at 70%, at best. However, antiangiogenic therapeutics, through gene silencing and targeting key sequences, probably brings an outlook to the conventional algorithm. In our current research, human-specific VEGF-siRNA (small interfering RNA) mediated by adenovirus was constructed and a cell line of MG63 was cultured and used to make an osteosarcoma-bearing nude mice model. The recombined adenovirus vector of Ad-VEGF-siRNA could successfully suppress VEGF expression and slow down the multiplication of MG63 cells in vivo; likewise, the down regulation of VEGF could be detected in vitro of the animal model. Inhibitory effects on osteosarcoma growth and blockage of pulmonary metastasis could be observed in the following oncotherapy procedure. The study demonstrates potent growth and pulmonary metastasis inhibitory effects of VEGF-siRNA on osteosarcoma in vivo and in vitro, which could potentially be applicable to the treatment of cancers as an antiangiogenic therapeutic in the near future.
Effects of adenovirus-mediated VEGF-siRNA on the growth of xenografted human osteosarcoma in nude mice

Abstract:Objective: Tumor growth and metastasis depend on angiogenesis. VEGF is the key gene in angiogenesis. The angiogenesis could be inhibited by blockade of VEGF expression thus suppressing the growth and metastasis of tumor. This study aimed to design a short hairpin RNA targeting VEGF, construct its adenovirus eukaryotic expression vector (Ad-VEGF-siRNA), and observe the effects of Ad-VEGF-siRNA transfection on growth of xenografted osteosarcoma MG 63 in nude mice. Methods: The Ad-VEGF-siRNA was transfected into cultured human osteosarcoma cell line MG 63. In vitro cell growth activities were tested by MTT assay. The expression of VEGF was determined using RT-PCR method. The xenograft model of nude mice was established. The Ad-VEGF-siRNA was injected intotumor body regularly. The tumor formation rate, tumor volume, tumor inhibiting rate, and the expression of apoptosis and proliferation-related genes were measured. Results: Transfection of Ad-VEGF-siRNA significantly inhibited the growth of MG 63 cells. The weight and volume of xenografted osteosarcoma significantly decreased compared with the control group after intratumor injection of Ad-VEGF-siRNA (P < 0.01). TUNEL staining showed that the apoptosis of MG 63 cells increased. Immunohistochemistry suggested that the expression of bcl-2 significantly decreased in osteosarcoma tissues (P < 0.01). Conclusion: Ad-VEGF-siRNA specifically and efficiently blocked the expression of VEGF with high efficiency, significantly inhibited the growth of xenografted osteosarcoma in nude mice, and induced the apoptosis of osteosarcoma MG 63 cells.
Research on the Inhibition on the Expression of VEGF in Tumor Tissues by Adenovirus Mediated VEGF-siRNA

Abstract:Objective To study the inhibition on the expression of VEGF in tumor tissues of nude mouse model carried with human osteosarcoma by adenovirus mediated VEGF-siRNA,and provide basis for the study of osteosarcoma vessels targeting therapy.Methods After establishment of nude mouse model carried with human osteosarcoma by subcutaneous inoculationg of MG63 cells suspension(1×106/ml),30 mice were randomly divided into 3 groups,A,B and C.Agent was administered to each group through vena caudalis 7 days after inoculation.Ad-VEGF-siRNA 100 μl was administered to group A every other day,3 times a week and 9 times in total with the total amount of 2×109 pfu.Ad-EGFP 100 μl was administered to group B every other day,3 times a week and 9 times in total with the total amount of 2×109 pfu.PBS was administered to group C every other day,3 times a week and 9 times in total.Tumor samples were taken on the 14th and 26th day from the mice of each group,the amount of VEGF-mRNA in the tumor samples were measured by the means of real time PCR.Results The average RQ value of VEGF-mRNA in tumor tissue of nude mice of each group on 14th and 26th day were:(0.088±0.094),(0.075±0.086) in group A,(0.179±0.103) and(0.167±(0.265)),in group B and(0.185±0.085),(0.183±0.128) in group C respectively.Average RQ value of group A had significant difference with the other two groups(P0.05).There was no significant difference between group B and C(P0.05).Conclusion Ad-VEGF-siRNA can inhibit the expression of VEGF in the osteosarcoma tissues,and this provides theoretical basis for the osteosarcoma vessels targeting therapy with RNA interference technology.
[Morphological Changes in Osteosarcoma Xenografts in Nude Mice after Inhibiting Angiogenesis by Ad-VEGF-siRNA].

Abstract:BACKGROUND AND OBJECTIVE:Vascular endothelial growth factor (VEGF) is a critical stimulator for angiogenesis of osteosarcoma. This study was to investigate the morphological changes in osteosarcoma xenografts in nude mice after inhibiting angiogenesis by Ad-VEGF-siRNA.METHODS:The animal model of osteosarcoma xenografts was constructed by subcutaneous inoculation of osteosarcoma MG63 cells into nude mice. The mice were randomly divided into three groups, 15 mice in each group: group A and control groups B and C. The tumor nodules were injected with Ad-VEGF-siRNA in group A, an equal dose of Ad-EGFP and PBS, respectively, in groups B and C. The volume and weight of tumors were measured every two days after injection and the growth curves were drawn. The tumor samples were observed and analyzed using HE staining. Angiogenesis was assessed by immunohistochemical staining using anti-VEGF and CD31 antibodies. The ultrastructure of vasculogenic mimicry (VM) was observed using transmission electron microscopy (TEM).RESULTS:The mean volume, weight, microvessel density (MVD) and VEGF expression of tumors in Ad-VEGF-siRNA-treated group were significantly lower than those in the control groups. MVD was positively correlated to VEGF expression, with a correlation coefficient of 0.9989. The number of VM was 1.40+/-0.55 in the Ad-VEGF-siRNA group, which was significantly less than those in the two control groups.CONCLUSION:Ad-VEGF-siRNA could effectively block angiogenesis in osteosarcoma xenografts in nude mice.
Effect and Mechanism Analysis of Sirna in Inhibiting VEGF and Its Anti-Angiogenesis Effects in Human Osteosarcoma Bearing Rats.

Abstract:OBJECTIVE:To explore the effect of siRNA in inhibiting vascular endothelial growth factor (VEGF) expression in tumor cells from human osteosarcoma bearing rats and its anti-angiogenesis effect, to further study the reliability, effectiveness and safety of VEGF as a therapeutic target in treating osteosarcoma.MATERIALS AND METHODS:After treatment, the long diameter and short diameter of tumor lesion were detected by Vernier caliper, and the tumor volume and tumor inhibition rate were calculated. The whole-body fluorescence imaging was used to detect the general morphology and volume change of tumor lesion before and after treatment. The rats were killed after treatment, RT-PCR and Western blot were used to detect VEGF expression. MTT was used to detect the proliferative ability of tumor cells in vitro.RESULTS:Three chemotherapies could inhibit the growth of tumor lesion, the decrease of tumor volume was significant (p < 0.05), the therapeutic effect in Ad-VEGF-siRNA + neoadjuvant chemotherapy group was better than the other two groups, the differences were statistically significant (p < 0.05). Furthermore, the three chemotherapies could inhibit the invasiveness of tumor cells, which was most significant in Ad-VEGF-siRNA + neoadjuvant chemotherapy group (p < 0.05).CONCLUSIONS:The growth of tumor tissue in osteosarcoma bearing rats is inhibited in Ad-VEGF-siRNA group, Ad-VEGF-siRNA + neoadjuvant chemotherapy group and Ad-VEGF-siRNA + anti-angiogenesis chemotherapy group. The effect in Ad-VEGF-siRNA + neoadjuvant chemotherapy is more significant than simple biological therapy or Ad-VEGF-siRNA + anti-angiogenesis chemotherapy.

Abstract:Objective: To investigate the inhibitory effect of adenovirus-mediated VEGF-siRNA on transplanted osteo- sarcoma in nude mice.Methods: VEGF-siRNA gene was cloned into the genome of replication-deficient adenovirus to construct Ad-VEGF-siRNA;the latter was then used to infect osteosarcoma MG63 cell line in vitro;and the expression of VEGF gene was detected by RT-PCR.Osteosarcoma transplantation model was established in nude mice;VEGF expression in tumor tissue was analyzed and the inhibitory effect on tumor growth and lung metastasis were also observed.Results: The recombinant adenovirus vector Ad-VEGF-siRNA was successfully constructed.In vivo and in vitro experiment both showed that Ad-VEGF-siRNA significantly downregulated VEGF expression in MG63 cells and transplanted tumor tissue. It was found that Ad-VEGF-siRNA significantly inhibited transplanted osteosarcoma growth(P0.05)and lung metastasis (P0.05)in tumor-bearing nude mice.Conclusion: The constructed Ad-VEGF-siRNA can effectively inhibit the expression of VEGF,depress transplanted tumor growth,and inhibit lung metastasis in nude mice.

Abstract:Objective To investigate the effects of RNA silencing via adenovirus-mediated vascular endothelial growth factor (VEGF) siRNA on proliferation and apoptosis of osteosarcoma cells.MethodsThe small hairpin RNA targeting VEGF gene carried by adenovirus were transfected into cultured human osteosarcoma MG63 cells.The in vitro cellular growth activities were assayed by MTT colorimetry.The cell apoptosis was studied by Hoechst staining and flow cytometry.The models of transplanted osteosarcoma in nude mice were successfully induced by intratumoral injection of humanosteosarcoma MG63 cells with with 0.2 ml Ad-VEGF-siRNA.Meanwhile,the in vivo cellular growth activities and apoptosis were studied by immunohistochemistry.Results Compared with control group,after the siRNA expression vetor was transfected into the MG63 cells,the cellular growth activities were decreased obviously at 24th,48th and 72nd h,with the apoptosis rate of 7.27%,12.01% and 20.09% respectively.The tumorigenesis of Ad-VEGF-siRNA group was greatly reduced.TUNEL revealed the sum of apoptosis MG63 cells was increased after therapy,however,immunohistochemistry demonstrated the expression of PCNA and bcl-2 was decreased obviously (P<0.01).Conclusion Ad-VEGF-siRNA could effectively and specifically the VEGF expression,tumor growth and angiogenesis,and promote apoptosis of the tumor cells.
Effect of bevacizumab in the RFP-tagged osteosarcoma nude mice model of human osteosarcoma in vivo

Abstract:Objective To explore the influence of bevacizumab ( Avastin) on the growth and angiogenesis of RFP-tagged os-teosarcoma orthotopic nude mice model of human osteosarcoma. Methods Twenty-one nude mice inoculated with human osteosarcoma cell line 143-B-RFP were randomly divided into three groups:control group, low dose Avastin group ( Avastin-L) and high dose Avas-tin group ( Avastin-H) . Avastin-L group and Avastin-H group received continuous 0. 2ml injection of 2. 0mg/kg or 5. 0mg/kg Avastin twice a week for 3 weeks, while control group was given an equal volume of saline. The body weight was recorded every 3 days. The in situ tumor size was measured to calculate the volume and the tumor inhibition rate every 7 days by fluorescence imaging system. Immu-nohistochemical En Vision method was used to detect the expression of CD34 in tumor tissue to evaluate the microvessel density ( MVD) . The vascular endothelial growth factor ( VEGF) levels of plasma and tumor tissue were measured by ELISA method. Results No significant difference was observed on body weight and lung metastasis rate among 3 groups ( P>0. 05) . Compared with control group, there were higher tumor inhibition rate, and lower tumor volume, MVD and VEGF levels of plasma and tumor tissue in both Avastin-L group and Avastin-H group (P<0. 05).The improvement effect of the above indexes were stronger in Avastin-H group versus Avastin-L group ( P<0. 05) . Conclusion Avastin can inhibit the growth and angiogenesis of human osteosarcoma, and reduce the VEGF level without influences on lung metastasis.
Hvegf Sirna Inhibits Initial Growth of A549 Tumor Cells in Nude Mice and Angiogenesis on CAM Assay

Abstract:OBJECTIVE:To investigate the effect of small interfering RNA (siRNA) targeting human vascular endothelial growth factor (hVEGF) on A549 cell growth in nude mice and angiogenesis on chorioallantoic membrane (CAM) assay.METHODS:Three pairs of hVEGF siRNA-plasmid and non-silencing-plasmid were constructed, and transfected into A549 cells through lipofectamine 2000, respectively. The most effective pair of hVEGF siRNA-plasmid was selected by ELISA and real-time RT-PCR. A549 cells transfected with selected hVEGF siRNA- plasmid, A549 cells transfected with non-silencing-plasmid and A549 cells without transfection were inoculated into nude mice, respectively. Chick embryos were randomly divided into four groups and CAM was treated by different solutions for 48 h: culture media DMEM as negative control group,un-transfected A549 cell culture supernatants as positive control group, hVEGF siRNA A549 cell culture supernatants as hVEGF siRNA group and nonsilencing siRNA A549 cell culture supernatants as non-silencing siRNA group. The CAMs were harvested on d12 for microscopic assays.RESULT:Compared with control group, hVEGF siRNA-plasmid induced 48% reduction in hVEGF secretion by A549 cells accompanied by 70% reduction in hVEGF mRNA. Compared with non-silencing siRNA group, the mean tumor volume of murine xenograft was reduced by 58% in hVEGF siRNA group; time for xenografts growing to 50 mm(3)was delayed by 5.4 d. hVEGF contents in xenograft were reduced by 54%; but mean doubling time of tumors and the growth rate of tumors were not significantly reduced. In CAM assays, hVEGF content was zero in negative group, and in hVEGF siRNA group that was 40%-44% of non-silencing siRNA group or positive group; vessels branch points of CAM in hVEGF siRNA group or non-silencing siRNA group or positive group were increased by 45%-55% compared with negative group; total vessel length of CAM in hVEGF siRNA group was increased by 53% compared with negative group, while in non-silencing siRNA group or positive group that was increased by 97% or 99%. Compared with negative control group, the proliferation of microvessels was increased when cell culture supernatant with hVEGF was added in hVEGF siRNA group, significant proliferated vessels were observed in non-silencing siRNA group or positive group.CONCLUSION:A plasmid-mediated hVEGF siRNA has been constructed and verified, which can effectively downregulate the expression of hVEGF in human A549 cells, resulting in the inhibition of angiogenesis. hVEGF siRNA can delay initial growth of A549 tumor xenograft but not reduce the growth rate.
[The Influence of Ad-AVEGF165 on Human Malignant Melanoma Growth in Nude Mice].

Abstract:OBJECTIVE:To investigate the effect of antisense VEGF165 infection on the growth of A375 cells in nude mice.METHODS:A375 cells were injected s.c into the axilla of the nude mouse. After the tumor formed, we cut it into 16 pieces equally, then transplanted into another 15 nude mice. There were three groups: Group PBS, Group Ad-GFP, and Group Ad-aVEGF. Four weeks after interfere, the mice were sacrificed and their tumors were excised for naked eye and histological observation. The VEGF expression was checked with ISH and immunohistochemistry staining. The micro-vessel density (MVD) in tumor mass was counted by VIII factor immunohistochemistry staining.RESULTS:The visible and palpable nodules had developed at all the injected sites. Tumor growth speed was more slowly in Group Ad-aVEGF than that in other groups. GFP gene could express effectively in tumor mass. Ad-aVEGF infection could suppress the growth of tumors, and there were no obvious side effects. Ad-aVEGF resulted more tissue necrosis, but it had no obvious effect on cell apoptosis. VEGF expression was inhibited significantly in Group Ad-aVEGF, and MVD was decreased accordingly.CONCLUSIONS:Ad-aVEGF interfere may be a new method against human malignant melanoma, whose main mechanism is to induce ischemia, but not apoptosis.
Effects of Vascular Endothelial Growth Factor-Sirna on Apoptosis and Vasculo-Genic Mimicry of Osteosarcoma Cells

Abstract:Objective: To study the role of vascular endothelial growth factor (VEGF)-siRNA in the proliferation, apoptosis, and vasculogenic mimicry of osteosarcoma cell line MG63. Methods: pSilencer vector was used to construct pSilencer-VEGF siRNA and the latter was used to transfect MG63 cells (experimental group). MG63 cells were also transfected with blank SiRNA to serve as control (control group). Western blotting was used to examine the expression of VEGF protein in MG63 cells in the 2 groups. The proliferation, apoptosis, and vasculogenic mimicry of MG63 cells were investigated using MTT assay, light microscope, and flow cytometry combined with Annexin V-FITC/PI and H-E staining. Results: The pSilencer-VEGF plasmid was confirmed by DNA sequencing. Sequence-specific siRNA targeting VEGF significantly decreased the expression of VEGF protein expression in MG63 cells. 48, 72 and 96 hours after pSilencer-VEGF-siRNA transfection, the inhibitory rate of MG63 cells proliferation were 41.67%, 43.92%, and 35.32%, respectively. Early apoptosis was also noticed in MG63 cells, with an apoptosis rate of 28.27%. Vasculogenic mimicry of MG63 cells was not found in experimental group, but was noticed in control group. Conclusion: Sequence-specific siRNA targeting VEGF can inhibit the proliferation, promote early apoptosis, and interfere with the vasculogenic mimicry of osteosarcoma cells, which casts new lights on clinical treatment of osteosarcoma.
Effect of anti-vascular endothelial growth factor antibody on the progression of human serous ovarian carcinoma xenotransplanted into nude mice

Abstract:Objective To study the inhibiting effect of the anti-VEGF121 monoclonal antibody on the progression of serous ovarian carcinoma in subcutaneous xenotransplanted nude mouse model. Methods The tissues of human serous ovarian carcinoma were directly transplanted into nude mice to construct the model for studying the angiogenesis-inhibiting effect.Nude mice were randomly divided into two groups: control group with PBS,and treated group with anti-VEGF antibody.After xenotransplantation,they were given i.p,twice a week for 9 times.The tumor volume and weight were evaluated after 1-week xenotrasnsplantation.The microvessel density(MVD) in tumor was immunohistochemically evaluated after treatment. Results There was significant differences in volume and weight of tumors between the two groups.The volume was smaller and weight was lighter in treated group than in the control group.MVD was significantly lower in treated groups than in control group. Conclusion Anti-VEGF121 antibody can inhibit angiogenesis of subcutaneously xenotransplanted tumor of human serous ovarian carcinoma into nude mice to inhibit its growth.

Abstract:OBJECTIVE: This study was to explore the effects of RNA interference mediated vascular endothelial growth factor (VEGF) gene silencing on biological behavior of renal cell carcinoma (RCC), transplanted renal tumor and angiogenesis in nude mice. METHODS: The specific siRNA sequence targeting VEGF were designed and synthesized to construct hVEGF-siRNA plasmid which was transfected into RCC 786-O cells. Reverse transcriptase-polymerase chain reaction (RT-PCR) was used for the detection of VEGF gene expression and western blot was adopted for the examination of VEGF protein expression. The 3-(4,5-Dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was used to detect cell growth as well as cell migration and invasion. The transplanted renal tumor models in nude mice were established, and the growth condition of nude mice, and VEGF protein expression in transplanted tumor slices and the microvessel density (MVD) were detected. RESULTS: The expression level of VEGF mRNA in VEGF-siRNA group was significant lower than that in the control group and negative group, suggesting that establishment of plasmid specifically inhibited the expression of VEGF gene The expression level of VEGF protein in VEGF-siRNA group was significant lower than that in the control group and negative group. VEGF gene silencing has the significant inhibition effects on proliferation, migration and invasion of RCC 786-O cells. The tumor weight, VEGF protein positive rate and MVD in VEGF-siRNA group were significant lower than those in negative group and blank group. CONCLUSION: The VEGF gene silencing could inhibit the cell proliferation, migration and invasion of RCC 786-O cells; inhibition of VEGF protein expression could prevent transplanted RCC growth and tumor angiogenesis.

Abstract:Objective: To construct an adenovirus-mediated mouse endostatin vector (Ad-mEndo) and to observe its inhibitory effect on the pulmonary inetastasis of osteosarcoma in nude mice, so as to discuss the relationship between ES expression and the pulmonary metastasis of osteosarcoma. Methods: Recombinant adenovirus plasmid pDC315-mEndo was constructed and used to prepare recombinant Ad-mEndo. Osteosarcoma MG-63 cells were subcutaneously injected into the right fore limbs to establish nude mouse model of osteosarcoma; and the models were randomly divided into 3 groups: AdmEndo group, Ad-EGFP group and PBS group; animals receiving no transplantation served as blank control. The corresponding agents were injected (20 μl per time) for a consecutive of 5 times on a weekly basis. The tumor volumes, histopathological characteristics were observed; ELISA was employed to examine the serum ES level. Animals were sacrificed 7 weeks later and the pulmonary metastasis was observed. Results: Sixteen days later, the tumor volume was (1.53 ± 0.05) cm3 in Ad-EGFP group, (1.56 ± 0.07) cm3 in the PBS group, and (0.91 ± 0.03) cm3 in the Ad-mEndo group, with the tumor inhibitory rate being 40.7% in the last group. The serum ES level in the Ad-mEndo, group was significantly higher than that of the other groups (P <0.05). No pulmonary metastasis was noticed in the Ad-mEndo group, but extensive metastases were seen in the lungs of the other 2 groups, with the metastasis rates being 80% and 90%. Mice without metastasis had significantly higher levels of ES compared with those with metastasis (P < 0.05). Conclusion: Adenovirus-mediated mouse endostatin can significantly inhibit the pulmonary metastasis of osteosarcoma in nude mice, indicating a directly link between expression of ES and pulmonary metastasis.
Effects of Sirna-Mediated HIF-1α Gene Silencing on Angiogenesis in Osteosarcoma.

Abstract:Objective: To explore angiogenesis in osteosarcoma under the condition of hypoxia-inducible factor (HIF)-1 alpha gene silenced by small interference RNA (siRNA).Methods: The SaOS-2 osteosarcoma cells, transfected with the recombinant plasmid pSilencer2.1-HIF-1 alpha or pSilencer2.1-SCR, were classified as HIF-1 alpha/siRNA group or SCR/siRNA group, respectively. In which, vascular endothelial growth factor (VEGF) immunohistochemistry were performed. HIF-1 alpha and VEGF protein contents were detected by western blot. Gene expressions of HIF-l alpha and VEGF were quantified by qPCR. Then the transfected SaOS-2 cells were inoculated in nude mice and transplantation tumor were checked via HE staining, VEGF and CD34 immunohistochemistry, and calculation of microvascular density (MVD).Results: In vitro, VEGF immunohistochemistry stains, HIF-1 alpha and VEGF protein contents, and the relative expressions of HIF-1 alpha mRNA and VEGF mRNA in HIF-1 alpha/siRNA group were obviously reduced. In vivo, morphological observation illustrated that heteromorphism were not obvious in the cells of HIF-1 alpha/siRNA group and vascular systems were sparse in its transplantation tumor tissue, and immunohistochemistry revealed that both VEGF and CD34 stains were significantly decreased in HIF-1 alpha/siRNA group, and MVD in HIF-1 alpha/siRNA group (7.3 +/- 1.1) were obviously less than that in SCR/siRNA group (17.2 +/- 3.2) (P<0.05).Conclusion: Angiogenesis in osteosarcoma can be inhibited by siRNA-mediated HIF-1 alpha gene silencing, which is expected to provide a novel and attractive target of therapeutic strategies of osteosarcoma.

Abstract:OBJECTIVE:To investigate the effects of vascular endothelial growth factor (VEGF) antisense oligodeoxynucleotide (ASODN) on the mRNA and protein expression of VEGF, Flt-1, and kinase insert domain containing receptor (KDR) and VEGF excretion in human gallbladder carcinoma cells.METHODS:Human gallbladder carcinoma cells of the line GBC-SD were cultured and transfected with VEGF ASODN and sense oligodeoxynucleotide (SODN) mediated by Oligofectamine. The toxicity of SODN and Oligofectamine to the GBC-SD cells was examined by MTT method. RT-PCR was used to detect the mRNA and expression of VEGF, Flt-1, and KDR, and ELISA was used to detect the protein expression of VEGF.RESULTS:MTT method showed that SODN and Oligofectamine were not toxic to the GBC-SD cells. The mRNA expression levels of VEGF, Flt-1, and KDR of the ASODN and ASODN + Oligofectamine groups were all significantly lower than those of the control group (all P < 0.05), and were the lowest 72 hours after transfection, and then gradually increased. ELISA showed that there were not significant differences in the VEGF protein concentration in the supernatant of the GBC-SD cells among the SODN, SODN + Oligofectamine, and control groups (all P < 0.05), however, the VEGF protein concentration in the supernatant of the GBC-SD cells of the ASDN and ASDN + Oligofectamine groups were significantly lower than that of the control group (both P < 0.05).CONCLUSION:VEGF ASODN inhibits the mRNA and protein expression of VEGF, Flt-1, and KDR and VEGF excretion in human gallbladder carcinoma cells. Oligofectamine strengthens the effect of ASODN.

Abstract:Objective To construct recombinant adenovirns containing small interfering RNA (siRNA) targeting human VEGF-C mRNA,then to study the inhibitory effects of adenovirus-mediated VEGF-C siRNA on growth and lymphangiogenesis of human colon cancer in BABL/c nude mice model.Methods In vitro:the specific siRNA sequence targeting human VEGF-C mRNA was selected.The homologous double-strand DNA was designed and synthesized.After such DNA was inserted into pDC316-EGFP-U6 by BamHI and HindⅢ,pDC316-VEGF-C siRNA-EGFP-U6 was obtained,then it was co-transfected into 293 cells with the hone plasmid pBHGF35.After generated by homologous recombination,Ad5F35-VEGF-C siRNA-EGFP-U6 was obtained,and it was packaged and amplified in 293 cells.The human colon cancer model was established in nude mice by hypo-injection LoVo cells.They were divided randomly into four groups (n=6):adenovirus,virus without target gene,single siRNA and PBS control group.Injecting intervention intra-tumorly in each groups,calculating the tumors volume and drawing the growth curve,calculating micro-vascular density (MVD) and micro-lymphatic density (LVD) by staining respectively of the lymphatic and vascular with anti-LYVE-1 monoclonal antibody and anti-CD<,34> monoclonal antibody were completed.Results Adenovirus group tumor sizes were smaller than other groups.The LVD were 8.47±2.1 and 17.35±4.7 (P<0.05) in adenovirns group and the PBS control group.The MVD were 22.65±6.04 and 23.19±7.63 (P>0.05) in adenovims group and the PBS control group respectively.Conclusion The adenovirus-mediated VEGF-C siRNA can significantly inhibit the growth and lymphangiogenesis of human colon cancer in nude mice model,hut have no effect on microangium vessels growth.

Abstract:Purpose Effect of antisense vascular endothelial growth factor was studied in prostate cancer LNCaP C42 cells biological features such as,proliferation and invasion in vitro and in vivo. Methods Tumor cells proliferation with antisense VEGF were detected by MTT analysis in vitro.In vivo,the tumor cells transfected with antisense VEGF were injected sc into nude mice.The tumor volume (mm3) wasmeasured by using a caliper,applying the formula [volume=0.52×(width)2×(length)] for approximating the volume of a spheroid.Intratumoral microvessel density (MVD) were detected by immunohistochemistry using mouse anti-human CD31 monoclonal antibody,and lung metastases nodule are evaluated using HE stai- ning. Results Prostate cancer LNCaP C42 proliferation was inhibited by antisense VEGF in vitro.In compared with the controls,growth of the implanted tumor was significantly reduced in sizes in mice transfected with antisense VEGF.The MVD decreased significantly in the mice transfected with antisense VEGF [(31±3.25) versus (14.25±3.40),P0.05],and the lung metastases were also significantly inhibited. Conclusions Antisense VEGF has a strong inhibitory effect both on tumor growth and metastases of human prostate cancer in vitro and in vivo by inhibiting tumor angiogenesis.It was suggested that inhibition of VEGF is a valuable therapeutic way in prostate cancer.
Effects of Antisense Oligonucleotides of VEGF-C on Anti-Lymphangiogenesis in Nude Mice with Orthotopic Implantation of Lung Cancer

Abstract:Objective To investigate the effects of antisense oligonucleotides of vascular endothelial growth factor C (VEGF-C) on anti-lymphangiogenesis in nude mice with orthotopic implantation of lung cancer. Methods After establishing the model of nude mice with orthotopic implantation of the human lung cancer cell line A549,30 nude mice were randomized into three groups with 10 mice each. The mice were treated with phosphate buffer solution (group PBS), sense oligonucleotides(group SODN) or antisense oligonucleotides (group AODN) within 24 h after A549 injection. The VEGF-C expression and lymphatic microvessel density(LMVD) of lung cancer were detected by RT-PCR,Western blot and immunohistochemistry. Results The expressions of VEGF-C and LMVD in the orthotopic implantation of lung cancer were less in group AODN than those in groups of PBS and SODN (P< 0. 05 or P< 0. 01). Conclusion The antisense oligonucleotides of VEGF-C can inhibit the expression of VEGF-C and lymphangiogenesis in nude mice of orthotopic implantation of lung cancer.

Abstract:Objective The purpose of the experiment is to study the effect of angiostatin gene on growth and metastasis of pancreatic carcinoma strain PC-3 orthotopic implantation model in mice establish.Methods The orthotopic implantation model was established by subcapsular implantation of human pancreatic carcinoma strain PC-3(2×106 cells) into pancrean of nude mice using microsyringe.Two weeks after implatation and gene therapy,the mice were sacrificed and the tumor sizes measured and the presence of metastasis recorded.The microvasular density(MVD)was examined by immunohistochemical staning.Result Compared with the untreated controls,growth of the orthotopic implantated tumor was significantly reduced in size in mice with angiostatin gene therapy,tumor metastasis was markedly inhibited,and the MVD was also decreased.Conclusion Angiostatin gene therapy has strong inhibitory effect both on tumor growth and metastasis of pancreatic carcinoma in nude mice.
