Transplantation of Labeled Neural Stem Cells Results in Functional Recovery after Traumatic Brain Injury in Rats

Abstract:Objective To label neural stem cells (NSCs) by different methods and to investigate the proliferation and differentiation of NSCs in rat model of traumatic brain injury.Methods A controlled cortical impact device was used to cause focal cortical contusion in rats with Feeney's method.NSCs labeled with green fluorescent protein (GFP) and superparamagnetic iron oxide (SPIO) were grafted stereotactically into the region nearby the contusion site.All animals were randomly divided into sham group,TBI group,PBS group,GFP labeled group and SPIO labeled group.The neurological severity score (NSS) and the rotarod test were used to evaluate neurological function recovery of rats at definite time following the transplantation.The transplanted NSCs were analyzed in recipient rat brains by immunohistochemical method.Otherwise the biodistribution of grafted NSCs was monitored noninvasively by magnetic resonance (MR) imaging.Results Significant improvement in NSS and the rotarod test were observed in rats receiving NSCs transplantation compared with control groups and PBS group (P 0.05). The transplanted NSCs preferentially engrafted into the region near the injured brain differentiated and expressed the neuronal marker β-tubilin III.MR imagining showed remarkable hypointense signal in the region of transplantation especially in T2GRE sequence.Conclusion Neurological and functional improvement was observed in rats with NSCs transplantation following traumatic brain injury.The new generated neurons may promote the functional recovery following traumatic brain injuries.And GFP labeling combined with MRI tracking of SPIO labeled NSCs could evaluate the therapeutic effect following transplantation.
