The mRNA expressions and the clinical significance of thymidylate synthase and dihydropyrimidine dehydrogenase in colorectal carcinoma

Abstract:Objective To explore the mRNA expressions of thymidylate synthase (TS) and dihydropyrimidine dehydrogenase (DPD) in colorectal carcinoma, and to evaluate their relationship with clinicopathological features. Methods The mRNA expressions of TS and DPD in the tumor tissues and the corresponding normal mucosa of patients with colo-rectal carcinoma were detected by semi-quantitative reverse-transcription polymerase chain reaction (RT-PCR), and then the relationship between the TS/DPD expressions and the clinicopathological features of colorectal carcinoma was analyzed. In addition, the protein expression localization was studied by immunohistochemistry in 10 cases. Results In colorectal carcinoma, the TS mRNA level in the tumor tissues was significantly higher than that in the adjacent normal tissues (1.37±0.72 vs. 0.97±0.59, P<0.001), and the DPD mRNA level in the tumor tissues was lower than that in the normal tissues (0.75±0.38 vs. 0.91±0.47, P=0.055). The TS mRNA level significantly correlated with lymph node metastasis (P<0.001) and Duke's stages (P=0.003). The DPD mRNA was highly expressed in the male patients (P=0.005) or the patients with rectal carcinoma (P=0.034). Immunohistochemistry showed the TS was expressed in both cytoplasm and nucleus, and DPD was only expressed in cytoplasm. Conclusions In colorectal cancinoma, the TS mRNA level in the tumor tissues is significantly higher than that in the corresponding normal tissues, but no statistical difference of the DPD mRNA level exists between the tumor and the normal tissues. The TS mRNA expression correlates very well with the clinical stage, while the DPD mRNA in the female patients or the patients with colon carcinoma has low expression.
