Abstract:Objective:To detect the expression of heat shock protein 70 (HSP70) in human renal carcinoma tissues and cultured ACHN cells by using quantum dots-tagged fluorescence technology and its significance. Methods:Using the fluorescence property of quantum dots, indirect immunofluorescence method and immunocytochemical method were used to detect the expression of HSP70 tagged by quantum dots in renal carcinoma tissues and ACHN cells cultured in vitro. Results:Confocal fluorescence microscopy showed that HSP70 were significantly expressed in human renal cancer tissues and ACHN cells cultured in vitro characterized by homogeneous distribution of intensive salmon pink fluorescence. Compared with FITC tagging, quantum dots tagged fluorescence had good specificity and signal to background. There was no notable quenching after excitation by quantum dots for 30 min. Conclusion:Quantum dots can be used to label subcellular proteins and have obvious advantages compared with the traditional fluorescence methods. The quantum dots-tagged fluorescence could be applied as a new method for clinical labeling detection.

Abstract:To investing the applying value of the novel fluorescent probe semiconductor quantum dot in immunofluorescence assay.We detected the expression of HSP70 in renal cell carcinoma tissure sections with semiconductor quantum dot(QD605)or conventional organic dye FITC as fluorescent labels,compared image difference and photostability.All labeling signals of quantum dot were found to be more specific and brighter than those of FITC.Moreover,the quantum dot exhibited exceptional photostability during continuous illumination for 1 h by a 488 nm laser.semiconductor quantum dot as fluorescent probe can offer substantial advantages over exsiting fluorophores in IFA for detecting tumor biomarkers.

Abstract:A new Quantum Dots(Qdots) nanocrystal composed of semiconductor core and zinc sulfide shell, and its feasibility as labels in immunofluorescence analysis for the imaging of tumor biomarkers by laser scanning confocal microscope(LSCM) was investigated. Qdots taged by mercaptoacetic acid were conjugated with second antibody, then imaging differences of Heat Shock Proteins 70(HSP70) in renal carcinoma tissure sections with immunofluorescence analysis method using Qdots bioconjugates and conventional organic dye FITC were observed by LSCM to assess the brightness and opticalstability of Qdots. The experimental results showed Qdots bioconjugates achieved the better results in demonstrating HSP70 with more brighter color and more clear picture than FITC labels. Moreover, the label signals of Qdots did not fade clearly after continued exposure to a 488 nm laser for 1 h. The Qdots bioconjugates have good feasibility in immunofluorescence analysis for the bioimaging by LSCM.

Abstract:Quantum dots (QDs) probes were used to tag and trace cancer biomarkers in cancer tissues based on the system of home-made Hadamard transform (HT) spectral microscopic imaging, which can be applied to provide high-resolution fluorescence spectrum and image of single cells and tissues. In situ fluorescence imaging for cancer marker proteins, such as estrogen receptor (ER), human epidermal growth factor receptor 2 (HER2), proliferating cell nuclear antigen (PCNA) and cytokeratin 20 (CK20) in tumor tissues, were realized by using the HT system to capture quantitative information for these proteins when tumor tissues were immunostained with QDs probes. A method to evaluate tumor malignancy of the specimens based on in situ analysis of distribution of marker proteins was proposed based on the comparative study of positive samples and negative controls. The investigation of ER contents of the cores in breast cancer tissue microarrays (TMAs) shows that the technique of QDs-immunohistochemistry (IHC)/HT spectral imaging is more sensitive than conventional IHC method. The results also demonstrate that the QDs-IHC/HT spectral imaging technique can be applied to visualize and quantitatively measure the subcellular molecules inside tumor tissues, and the coupling of HT spectral imaging to the probing of subcellular molecules with QDs has great potential in biology and medical diagnosis.

Abstract:Objective To explore the different expressi on of protein between human kidney cancer and normal kidney tissues and to ident ify specific biomarkers. Methods The protein spectra of 12 samples from human kidney cancer patients and another 12 samples taken from t he normal renal tissues which were distal to the renal cancer were analyzed by S ELDI(Surfaced enhanced laser desorption/ionization)technology. Resul ts Compared with normal kidney tissues,two specific protein peaks wi th molecular weight of 5750.4 Da and 5861.7 Da respectively of the protein spect ra in human kidney cancer on IMAC3 protein chip elevated significantly with high specificity and sensitivity. Conclusions Detection of s pecific protein in human kidney cancer is significant both for study on cancer d evelopment mechanism and determination of clinical specific biomarkers.
Expression of Heat Shock Factor in Primary Renal Cell Carcinoma and Its Relationship with Multidrug Resistance

Abstract:Objective To investigate the expression of heat shock factor 1 in human primary renal cell carcinoma (RCC) and its relationship with P-glycoprotein (P-gp).Methods Fifty-four samples of RCC tissue and 10 samples of normal renal tissue were detected preoperatively for the expression of HSF1 and P-gp by using immunohistochemistry.Results The expression of HSF1 and P-gp was detectable in 100% samples of normal renal tissue,but in the samples of RCC tissue the expression of P-gp was detectable in 70.37% and nearly no expression of HSF1 was detectable.Conclusion Although indirect proofs have suggested that HSF1 could participate in the MDR-1 gene expression, based on our preliminary findings, it is necessary to further investigate the relationship between HSF1 and MDR in RCC.
Detection of Prostate Stem Cell Antigen Expression in Human Prostate Cancer Using Quantum-Dot-Based Technology

Abstract:Quantum dots (QDs) are a new class of fluorescent labeling for biological and biomedical applications. In this study, we detected prostate stem cell antigen (PSCA) expression correlated with tumor grade and stage in human prostate cancer by QDs-based immunolabeling and conventional immunohistochemistry (IHC), and evaluated the sensitivity and stability of QDs-based immunolabeling in comparison with IHC. Our data revealed that increasing levels of PSCA expression accompanied advanced tumor grade (QDs labeling, r = 0.732, p < 0.001; IHC, r = 0.683, p < 0.001) and stage (QDs labeling, r = 0.514, p = 0.001; IHC, r = 0.432, p = 0.005), and the similar tendency was detected by the two methods. In addition, by comparison between the two methods, QDs labeling was consistent with IHC in detecting the expression of PSCA in human prostate tissue correlated with different pathological types (K = 0.845, p < 0.001). During the observation time, QDs exhibited superior stability. The intensity of QDs fluorescence remained stable for two weeks (p = 0.083) after conjugation to the PSCA protein, and nearly 93% of positive expression with their fluorescence still could be seen after four weeks.

Abstract:Hsp70 proteins are implicated in resistance to chemotherapy in cancers, the detection of which is important for cancer treatment and prognosis. In this work, we report the study on the detection of specific intracellular target protein in fixed cells using GlcNAc-conjugated CdSeTe QDs. The QDs were coupled with Con A via a carbodiimide reaction and then were further assembled with GlcNAc by lectin-carbohydrate interaction between Con A and GlcNAc. The obtained QDs-Con A-GlcNAc conjugates have an emission wavelength at 650 nm that is close to the near-infrared (NIR) regions and a specific recognition for Hsp70. These results show that the QDs-Con A-GlcNAc probe can be a promising tool for direct localization of the Hsp70 protein.

Abstract:Background/Aims: In the present study, we describe a novel and straightforward approach to produce a cyclic- arginine-glycine-aspartic (RGD)-peptide-conjugated quantum dot (QD) probe as an ideal target tumor biomarker. Due to its specific structure, the probe can be used for targeted imaging of pancreatic carcinoma cells. Methods: Pancreatic carcinoma cells were routinely cultured and marked with QD-RGD probe. The QD-RGD probe on the fluorescence-labeled cancer cell was observed by fluorescence microscopy and laser confocal microscopy. Cancer cell viability was detected by MTT assay after culturing with QD-RGD probe. Results: Fluorescence microscopy and laser confocal microscopy displayed that 10nmol/L QD-RGD probe was able to effectively mark pancreatic carcinoma cells. In comparison with organic dyes and fluorescent proteins, the quantum dot-RGD probe had unique optical and electronic properties. Conclusion: QD-RGD probe has a low cytotoxicity with an excellent optical property and biocompatibility. These findings support further evaluation of QD-RGD probes for the early detection of pancreatic cancer.

Abstract:Background:Pancreatic carcinoma is obscure in onset and progresses rapidly with poor prognosis and low five-year survival rate.Currently,targeted therapy of cancer has become a hot spot of research.Quantum dots-RGD has been used as a fluorescent probe targeting marker of pancreatic carcinoma.Aims:To study the target marking and biocompatibility of quantum dots-RGD probe in human pancreatic carcinoma cell line SW1990.Methods:Pancreatic carcinoma SW1990 cells were conventionally cultured and marked by quantum dots-RGD probe.Fluorescence microscopy and laser confocal microscopy were used to observe the marking state of 10 nmol/L quantum dots-RGD probe in pancreatic carcinoma SW1990 cells.Activities of pancreatic carcinoma SW1990 cells were assessed by MTT assay after culturing with various concentrations of quantum dots-RGD probe(0,10,15,20,40,70 nmol/L) for 12,24,48 hours,respectively. Results:Fluorescence microscopy and laser confocal microscopy showed that 10 nmol/L quantum dots-RGD probe could effectively target mark pancreatic carcinoma SW1990 cells.Activities of SW1990 cells in 10,15,20 nmol/L quantum dotsRGD probe groups were not statistically different from that of blank control group.Conclusions:The quantum dots-RGD probe can target mark pancreatic carcinoma SW1990 cells with excellent optical specificity and biocompatibility,and would be helpful for studies on targeted therapy of pancreatic carcinoma.

Abstract:Objective To study the expression and significance of HSP70 and HSP90 in renal cell carcinoma.Methods Immunohistochemical S-P method was used to detect the expression of HSP70 and HSP90 in 32 cases of renal cell carcinoma.Results HSP70 and HSP90 were overexpressed in renal cell carcinoma,and the positivity rate was 90.6% and 84.4% respectively(P<0.05).No correlation was found between the degree of these expression and clinical stage,the histologic grade and the his-tologic type(P>0.05).Conclusion HSP70 and HSP90 are promotional to the occurrence and deve-lopment of renal cell carcinoma.

Abstract:Quantum dots, as a new kind of biological fluorescence material, have properties superior to the traditional organic dyes. In the recent decade, researches about the application of quantum dots in biomedicine have made great progress. Tumor markers have vital importance in the diagnosis and treatment of carcinoma; however, they are not widely used for lack of specificity and sensitivity. Researches about applications of quantum dots in the detection of tumor markers are centralized in three aspects, namely the detection of serum/plasma tumor markers, the detection of tissue and cell tumor markers (such as molecular imaging of cell and tissue), and the detection of tumor in vivo animals. A combination of quantum dots and flow cytometry technique for developing a new technique to detect multiple kinds of tumor markers at the same time with greater sensitivity, specificity, rapidity and convenience may have the potential for clinical use of quantum dots.

Abstract:We describe here a novel approach for detection of cancer markers using quantum dot protein microarrays. Both relatively new technologies; quantum dots and protein microarrays, offer very unique features that together allow detection of cancer markers in biological specimens (serum, plasma, body fluids) at pg/ml concentration. Quantum dots offer remarkable photostability and brightness. They do not exhibit photobleaching common to organic fluorophores. Moreover, the high emission amplitude for QDs results in a marked improvement in the signal to noise ratio of the final image. Protein microarrays allow highly parallel quantitation of specific proteins in a rapid, low-cost and low sample volume format. Furthermore the multiplexed assay enables detection of many proteins at once in one sample, making it a powerful tool for biomarker analysis and early cancer diagnostics.

Abstract:Objective To investigate the expression and significance of HSPD1 in renal cell carcinoma and adjacent normal tissues.Methods Two-dimensional chromatography and mass spectrometry method were applied to identifying the difference of HSPD1 expressed in renal cell carcinoma and adjacent normal tissues of 10 patients with renal cell car-cinoma.Results Identifications by mass spectrometer showed that HSPD1 expressed significantly lower in renal cell carcinoma than that in the tissue adjacent to carcinoma,and then confirmed the accuracy by western blot.Conclusion the decrease of HSPD1 expression may have a closely relationship with renal cell carcinoma.

Abstract:This study explored the 605 nm carboxyl of water-soluble quantum dots to assess the practicability of the nasopharyngeal carcinoma marker EBNA1 antibody. We used 605 nm carboxyl water-soluble quantum dots and nasopharyngeal carcinoma EBNA1 antigen in 1-(3-dimethylaminopropyl-3-2-ethylcarbonimine hydrochloride to generate quantum dot-labeled antigen. Gel imaging system showed that serum group and 60 patients with nasopharyngeal carcinoma had significant differences in 30 cases of normal adult serogroup brightness, Levene test in the two groups ELISA absorbance value was P<0.001, namely the nasopharyngeal carcinoma group and the normal adult serum group measured absorbance value difference was obvious. The statistical significance, and the detection technology were of high specificity and sensitivity. In conclusion, this study adopted double antigen clip combining immune chromatography test EBNA1 antibody in serum, compared with the traditional enzyme-linked immunoassays this method is more rapid, with simpler operation, rapid detection for developing quantum dot immune chromatography technology the EBNA1 antibody kit provides a theoretical basis.

Abstract:DNER, Delta/Notch-like epidermal growth factor (EGF)-related receptor, is a neuron-specific transmembrane protein carrying extracellular EGF-like repeats. The prognostic value of DNER in prostate cancer has not been evaluated. Here we showed that the up-regulation of DNER protein was observed in prostate cancer detected by immunohistochemistry (IHC) and quantum dot-based immunofluorescent imaging and quantitative analytical system (QD-IIQAS). However, a higher accuracy of measurements of DNER expression in prostate cancer was found by QD-IIQAS than by IHC (AUC = 0.817 and 0.617, respectively). DNER was significantly higher in patients undergoing bone metastasis (P = 0.045, RR = 3.624). In addition, DNER overexpression was associated with poor overall survival (OS) (P = 0.028, adjusted HR = 8.564) and recurrence-free survival (RFS) (P = 0.042, adjusted HR = 3.474) in patients suffering prostate cancer. Thus, QD-IIQAS is an easy and accurate method for assessing DNER and the DNER expression was an independent prognostic factor in prostate cancer.
Ratiometric Fluorescence Sensor Arrays Based on Quantum Dots for Detection of Proteins

Abstract:Optical cross-reactive sensor arrays have recently been demonstrated as a powerful tool for high-throughput protein analysis. Nevertheless, applying this technology to protein detection is complicated by many external factors, such as the interfering substances, the background noise, and sample environmental changes in the biological matrix. Herein we demonstrate that a ratiometric fluorescence sensor array based on quantum dots can be employed to circumvent these limitations. Several intrinsic dual-emitting Mn-doped quantum dots capped with different organic functional groups were designed as sensing elements. Distinct and reproducible response patterns against the ratiometric sensor array were obtained from ten proteins in a buffer of different pH (pH 5.7, 7.4, and 8.3) and spiked into human urine. Linear discrimination analysis of the response patterns showed successful differentiation of the analytes at concentrations as low as 50 nM with high identification accuracy. Furthermore, this sensor system also enables the detection of these eight proteins (at 500 nM) in human urine without any treatment. The ratiometric fluorescence change from quantum dots for analysis of proteins can eliminate effectively the signal interference from the pH value change and the fluorescent background in human urine. The present study will open a new avenue to improve the discrimination ability of sensor arrays.

Abstract:The use of highly specific and highly sensitive immunofluorescent probes is a promising approach for biomedical imaging in living tissue. We focus on immunofluorescence with quantum dot bioconjugates for hepatoma detection in vivo. We synthesized specific immunofluorescent probes by linking quantum dots to AFP (alpha-fetoprotein) antibody for specific binding AFP-an important marker for hepatocellular carcinoma cell lines. In in vivo studies, the characteristic quantum dot (QD) fluorescent property is exhibited by the QDs-Anti-AFP probes in tumor and they demonstrate active tumor targeting and spectroscopic hepatoma imaging with an integrated fluorescence imaging system. We investigate the inhomogeneous distribution of the QDs-Anti-AFP probes in tumor by using a site-by-site measurement method to test their ability for distribution studies of cancer cells. These results demonstrate the practicality of QD bioconjugates as attractive fluorescent probes for biomedical detection.

Abstract:Objective To explore the marking method of 605 nm carboxyl water-soluble quantum dots in naso-pharyngeal carcinoma marker EBNA1 .Methods The 605 nm carboxyl water-soluble quantum dots and nasopharyn-geal carcinoma EBNA1 antigen were performed the covalent cross-linking under the action of 1-(3-dimethyl amino propyl)-3-2 ethyl carbon imine hydrochloride ,the quantum dots labeled antigen was generated ,then the labeled anti-gen was detected by UV-Vis absorption spectroscopy ,fluorescence spectrometry and agarose gel electrophoresis anal-ysis .Results The stable covalent bond formation of carboxyl water-soluble quantum dots with nasopharyngeal carci-noma EBNA1 antigen by the surface amino and carboxyl condensation on was found ,the carboxyl water-soluble quan-tum dots achieved the successful connection with EBNA1 nasopharyngeal carcinoma antigen ,the labeled fluorescence emission spectrum detection results showed that the emission peak position of the labeled quantum dots EBNA1 solu-tion was near 380 nm ,keeping good fluorescence properties .Conclusion The 605 nm carboxyl water-soluble quantum dots can stably label nasopharyngeal carcinoma marker EBNA1 by adopting the covalent cross-linking method to pro-vide reliable basis for subsequent research .

Abstract:AIM:To explore the expression and significance of heat shock protein 70 (Hsp70), glucose regulated protein 94 (Grp94) and immunoglobulin G (IgG) in human lung carcinoma.METHODS:The expression of Hsp70, Grp94 and IgG in 40 human lung carcinomas was studied using immunohistochemical technique and image analysis. The localization among Hsp70, Grp94 and IgG was analyzed by double labeling immunofluorescent staining and laser scanning confocal microscopy.RESULTS:Hsp70, Grp94 and IgG in Human lung carcinomas showed high expression. The positive rate of Hsp70, Grp94 and IgG was 65% (26/40), 45% (18/40), and 82.5% (33/40), respectively. The average value of optical density was 5.10 +/- 0.32, 3.52 +/- 0.35, and 8.12 +/- 0.31, respectively. Hsp70 was localized in nucleus and cellular cytoplasm while Grp94 and IgG were mainly localized in cellular cytoplasm. Ten cases showed Hsp70 was co-localized with IgG and Eighteen cases showed Grp94 was co-localized with IgG.CONCLUSION:High expression of Hsp70, Grp94 and IgG in Human lung carcinomas suggested that Hsp70, Grp94 and IgG might play an important role in the development of human lung carcinoma. IgG is co-localized with Hsp70 or Grp94. The study will lay a theorical basis for further research on anti-tumor immunotherapy.
