Abstract:BSTRACT The integration pattern and adjacent host sequences of the inserted pMThGH-transgene in the F4 hGH -transgenic common carp were extensively studied. Here we show that each F4 transgenic fish contained about 200 copies of the pMThGH-transgene and the transgenes were integrated into the host genome generally with concatemers in a head-to-tail arrangement at 4-5 insertion sites. By using a method of plasmid rescue, four hundred copies of transgenes from two individuals of F4 transgenic fish, A and B, were recovered and clarified into 6 classes. All classes of recovered transgenes contained either complete or partial pMThGH sequences. The class I, which comprised 83% and 84.5% respectively of the recovered transgene copies from fish A and B, had maintained the original configuration, indicating that most transgenes were faithfully inherited during the four generations of reproduction. The other five classes were different from the original configuration in both molecular weight and restriction map, indicating that a few transgenes had undergone mutation, rearrangement or deletion during integration and germline transmission. In the five types of aberrant transgenes, three flanking sequences of the host genome were analyzed. These sequences were common carp β -actin gene, common carp DNA sequences homologous to mouse phosphoglycerate kinase-1 and human epidermal keratin 14 , respectively.

Abstract:To gain information on the integration pattern of pMThGH-transgene, 50 transgenes were recovered from F 4 generation of pMThGH transgenic common carp ( Cyprinus carpio L.) and 33 recovered genes were analyzed. The restriction maps of these recovered genes were constructed by digestion with five kinds of enzymes. These transgenes can be classified into 4 types according to their restriction maps. Only one type of transgenes maintains its original molecular form, whereas the other three types are very different from the original one and vary each other on both molecular weight and restriction maps. This implies that the sequences of most transgenes have been deleted and/or rearranged during integration and inheritance. The results of PCR amplification and Southern blot hybridization indicate that MThGH in Type I transgene keeps intact but most of its sequence has been deleted in other three types. All these results suggest that transgenes in F 4 generation of transgenic carp are highly polymorphic. Two DNA fragments concerning integration site of transgenes were cloned from recovered transgenes, and found to be homologous to the 5′UTR of β-actin gene of common carp and mouse mRNA for receptor tyrosine kinase ( RTK ), respectively.

Abstract:The tetraploid fish has been developed by assortative breeding the hybrids of Carassius auratus red var. (Female) x cyprinus carpio (Male), which has the stable genetic characters and can reproduce themselves. An "all-fish" recombinant DNA construct (pCAgcGHc) containing common carp beta-actin gene promoter and cDNA for grass carp (Ctenopharyngodon idella) growth hormone gene was introduced into fertilized eggs of the allotetroploid fish through microinjection as soon as artificial insemination was done. Artificial insemination was carried out between the female and the male transgenic allotetraploid fish which contain the "all-fish" recombinant DNA construct (pCAgcGHc) and are the biggest in the size. Fifty F1 samples of transgenic allotetraploid fish of 150 days and 50 allotetraploid fish (regarded as the control) were chosen, and the weight and the body length of each were measured, the results showed that F1 of transgenic allotetraploid fish of 150 days had obvious growth dominance compared with the control. Genomic DNA of tail fin was extracted from 20 F1 of transgenic allotetraploid fish of 150 days and the control. Proper primers were introduced to check whether the sample had the transgene. Pa, the upstream primer, is located in beta-actin promoter, and Pg, the downstream primer, is located in growth hormone cDNA for grass carp (gcGHc). The transgene was detected in 90% F1 of transgenic allotetraploid fish in tail fin DNA by polymerase chain reaction (PCR) amplification. Sperm could be squeezed out from a few F1 of transgenic allotetraploid fish of 150 days, however, this phenomenon did not exist in the controls. The importance of forming the pure line of transgenic allotetraploid was elucidate in the paper.
The onset of foreign gene transcription in nuclear-transferred embryos of fish

Abstract:The transcriptional onset of hGH -transgene in fish was studied in the following three cases: the first is in MThGH -transgenic F 4 common carp ( Cyprinus carpio ) embryos, the second is in nuclear-transferred embryos supported by the transgenic F 4 embryonic nuclei, and the third is in nuclear-transferred embryos supported by the transgenic F 4 tail-fin nuclei. RT-PCR results show that the hGH -transgene initiates its transcriptional activity from early-gastrula stage, the early blastula stage and even 16-cell stage in the first, second and third cases, respectively. It looks like that fish egg cytoplasm could just offer a very restricted reprogramming on transcriptional activity of specific gene in differentiated cell nuclei by nuclear transplantation.

Abstract:In order to study the feasibility of piggyBac transposon application in the fish,the plasmid consisting of the piggyBac inverted terminal repeats flanking a fusion of the Bombyx mori cytoplasmic actin gene BmA3 promoter and the enhanced green fluorescent protein(EGFP) gene and a nonautonomous helper plasmid encoding the piggyBac transposes was introduced into the zygote of the Macropodus opercularis through microinjection.The PCR identification indicated that the EGFP gene mediated by piggyBac transposon existed in the genome of the M.opercularis.Transgene was stably transferred to the next generation through normal Mendelian inheritance.The foreign DNA integration rate,i.e.,the rate of number of G1 broods with EGFP positive fish to the number of fertile fish was 12.30%.These results prove that piggyBac plasmid can be a new vector for the transgenic experiment in fish.The system constructed in our experiment is feasible.

Abstract:Recombinant “all-fish” growth hormone gene (GH) was microinjected into the fertilized eggs of carp. A comparison between the growth traits of transgenics and non-transgenics was carried out, and the transgenic individuals with significant “fast-growing” effect were successfully gained. A comparison on the reproductivities was also given out between the transgenics and their non-transgenic siblings, and showed that the reproductive capacity of transgenics was substantially equivalent to those of the non-transgenics. On the other hand, the genetic separation and the characteristic distribution of the F1 generation were genetically analyzed, which gave solid evidence for the hypothesis that 2–3 chromosomes are integrated with transgene. In addition, the distinct biological effects for multisite-integrated transgenes were further discussed. The present study opens a door for the breeding of “fast-growing” transgenic fish.
The Relationship Between Internal Domain Sequences of Piggybac and Its Transposition Efficiency in BmN Cells and Bombyx Mori.

Abstract:The piggyBac transposon, which includes terminal inverted repeat sequences and internal domain (ID) sequences, is widely used as a tool for insect transformation. To optimize this system for transgenic research on Bombyx mori, we examined the effects of the amount of the transposase plasmid and its ID sequences on the expression of green fluorescent protein (GFP). Four kinds of transposon plasmids, pB[A3GFP]-1 with the full length of ID sequences, pB[A3GFP]-2 having only the 3' ID sequence, pB[A3GFP]-3 without ID sequences, and pB[A3GFP]-4 containing 333 bp of the 5' ID sequence, and 179 bp of the 3' ID sequence were constructed with GFP as the marker. After transfecting these four plasmids into BmN cells, we analyzed the transfecting efficiency by comparing the GFP positive to negative cell ratio. Our results indicated that plasmid pB-4 got the highest ratio on the 22nd day. Moreover, the GFP positive to negative cell ratio increased with higher amount of transposase plasmid without overproduction inhibition. Furthermore, we injected three piggyBac transposon plasmids, pB[A3GFP]-1, pB[3xP3GFP]-3, and pB[3xP3RFP]-4 harboring different markers into preblastoderm stage eggs of B. mori, and found that the transformation efficiency of pB[3xP3RFP]-4 was 3.8 folds higher than pB[A3GFP]-1, whereas pB[3xP3GFP]-3 failed to produce transformants. Our results suggested that pB-4 may be one of the best piggyBac transposon plasmids currently available for germline transformation in B. mori.

Abstract:Some members of hairy/Enhancer-of-split -related gene (HES) family have important effects on axial mesoderm segmentation and the establishment and maintenance of the somite fringe. In fishes, the her6 gene, a member of the HES family, is the homologue of hes1 in mammals and chicken. In this study, the her6 gene and its full-length cDNA from the common carp ( Cyprinus carpio ) were isolated and characterized. The genomic sequence of common carp her6 is approximately 1.7 kb, with four exons and three introns, and the full-length cDNA of 1314 bp encodes a putative polypeptide of 271 amino acids. To analyse the promoter sequence of common carp her6 , sequences of various lengths upstream from the transcription initiation site of her6 were fused to enhanced green fluorescent protein gene ( eGFP ) and introduced into zebrafish embryos by microinjection to generate transgenic embryos. Our results show that the upstream sequence of 500 bp can direct highly efficient and tissue-specific expression of eGFP in zebrafish embryos, whereas a fragment of 200 bp containing the TATA box and a partial suppressor of hairless paired site sequence (SPS) is not sufficient to drive eGFP expression in zebrafish embryos.

Abstract:We demonstrate a novel method for identification of the insertion site of transgenic DNA by construction a circular DNA containing part of the transgenic DNA (tDNA) and its flanking sequence, which would be different from those in the native genomic DNA (gDNA). Using follistatin (FST) as a model of tDNA, we cleaved the sequence of FST gene and the adjacent sequence with a proper combination of restriction endonuclease and nicking endonuclease and isolated the resultant fragments containing the target FST gene from the gDNA background. Two types of previously unknown ligation property of T7 DNA ligase were disclosed in our experiments. One is non-templated single-stranded overhang ligation capability; the other one is the successful ligation of 3’-overhang and 5’-recessed ends in the presence of two mismatched base pairs. By virtue of these efficient ligation reactions, we successfully construct the circular DNA with high cyclization yield. Further coupling with nested inverse PCR, the method enabled identification of transgenic follistatin a (fsta) cDNA in zebrafish embryonic DNA. The established method provides a practical tool to determine the insertion site of tDNA with a sensitivity of 103 copies in 1.0 μg of gDNA and high specificity. It may be further extended for gene doping detection and assessment of the safety of gene therapy and genetically modified organisms.

Abstract:The genetic stability and expression efficiency of exogenous genes in transgenic animals are closely related to integration site and copy number. In our laboratory, by transgenic manipulation and subsequent test crosses, we established an ‘‘all-fish’’ growth hormone(GH)transgenic common carp family that exhibits fast growth.In this present study, genome walking, real-time quantitative polymerase chain reaction, and fluorescence in situ hybridization techniques were applied to identify the integration characteristics of the exogenous grass carp GH gene in the transgenic common carp. The exogenous GH genes, in the form of two complete and one incomplete tandem repeats, were found to have integrated into an ATrich region near the end of a chromosome pair. We hypothesize that the high efficiency of exogenous GH gene expression might be due to the low copy number in the genome and the AT-rich integration site.
Nuclear Transplantation with Early-Embryonic Cells of Transgenic Fish

Abstract:Like other transgenic animals, transgenic fishes produced by microinjection are transgenic mosaics. In order to produce homogenous transgenic fish, the transgenic blastula or gastrula cells were dissociated from Carassius auratus, Pengze var, and Cyprinus carpio, Huanghe var., and the nuclei were transferred into the mature eggs of the same species via microinjection or electro-fusion. Five nuclear-transferred Carassius auratus, Pengze var. and one Cyprinus carpio, Huanghe var. were obtained and the existence of the transgene was detected. The possibility of generating homogenous strain of transgenic fish by nuclear transplantation with transgenic early-embryonic cells is discussed.

Abstract:Distant hybridization refers to the combination of different genomes from different species and further leads to genetic structure changes in hybrids. For next-generation sequencing limitations, the genetic changes with respect to hybrids or polyploids cannot be fully explained. Here, we obtained 117,526 full-length isoforms from a triploid hybrid grass carp (3nGT, 3n = 72) by the crossing of female grass carp (GC, 2n = 48) and male topmouth culter (TC, 2n = 48) using single-molecule long reads sequencing technology. Further, the gene fusion, alternative splicing (AS), transcription of homologous genes, and novel genes were analyzed. The results showed that 1563 fusion genes were detected in 3nGT including 872 heterogenous fusion genes derived from GC and TC genomes. Meanwhile, 175,644 AS events were identified and divided into five models, of which IntronR was the dominant model. In addition, 8964 novel genes were discovered, of which 2802 novel genes were annotated. Finally, scans results of Hox gene family revealed that both GC and TC Hox genes were transcribed in 3nGT, but presented asymmetric transcription patterns with a bias towards the maternal parent GC genome. This study not only provides a rich resource of transcript isoforms for the superior triploid grass carp, but also provides an important insight into rapid genetic changes in hybrid fish.

Abstract:The purpose of this study was to obtain the full-length sequence of common carp interleukin 10(IL-10) genomic DNA.The sequence of common carp IL-10 full-length cDNA(GenBank:JX 524550) was used.In this study,we designed two pairs of primers in its untranslated regions.Using the carp spleen genomic DNA as the template,we carried out PCR and obtained the IL-10 full-length genomic DNA sequence.The sequence was 2176 bp in length,and the GenBank accession number was JX524551.We compared the gained sequence with the other IL-10 and found their similarity of all harboring 5 exons and 4 introns.Moreover,all exons were comparatively conserved.The exons and introns splice sites were in accordance with gt......ag rules.Our results indicated that it was the sizes of introns that were crucial in the process of evolution.

Abstract:The Chinese sturgeon, Acipenser sinensis, is a large-sized panadromous cartilagous fish and requires more than ten years to reach sexual maturity. An understanding of the reproductive organs and fertilization process is a precondition for controlled reproduction of this endangered species. The developing oocyte is surrounded by an extracellular matrix composed of the zona pellucide (ZP) protein in several isoforms, all of which play important roles in fertilization and embryo protection. The present study reports the identification of two ZP genes, AsZPAX and AsZPB, in this fish. The full-length cDNAs of AsZPAX and AsZPB were 2950 base pairs (bp) and 1832bp, predicting proteins of 946 and 592 amino acid residues, respectively. Both proteins have a conserved ZP domain. In a gene tree, AsZPAX and AsZPB were respectively clustered with their counterparts in other vertebrates. Tissue distribution analysis demonstrated that the AsZPAX RNA was transcribed in both the ovary and the liver, while AsZPB was transcribed exclusively in the ovary. RNA in situ hybridization revealed apparent RNA expression of both AsZPAX and AsZPB in growing oocytes of the 4-year-old ovary.

Abstract:Abstract How two subgenomes in allo-tetraploids adapt to coexistence and coordinate through structure and expression evolution requires extensive studies. In the present study, we report an improved genome assembly of allo-tetraploid common carp, an updated genome annotation of allo-tetraploid goldfish and the chromosome-scale assemblies of a progenitor-like diploid Puntius tetrazona and an outgroup diploid Paracanthobrama guichenoti . Parallel subgenome structure evolution in the allo-tetraploids was featured with equivalent chromosome components, higher protein identities, similar transposon divergence and contents, homoeologous exchanges, better synteny level, strong sequence compensation and symmetric purifying selection. Furthermore, we observed subgenome expression divergence processes in the allo-tetraploids, including inter-/intrasubgenome trans- splicing events, expression dominance, decreased expression levels, dosage compensation, stronger expression correlation, dynamic functionalization and balancing of differential expression. The potential disorders introduced by different progenitors in the allo-tetraploids were hypothesized to be alleviated by increasing structural homogeneity and performing versatile expression processes. Resequencing three common carp strains revealed two major ecotypes and uncovered candidate genes relevant to growth and survival rate.

Nuclear Transplantation of Somatic Cells of Transgenic Red Carp (cyprinus Carpio Haematopterus).

Abstract:Nuclear transplantation was performed using kidney cells, tail fin cells and cultured tail fin cells of 18th passage of the F4 hGH gene transferred red carp (Cyprinus carpio haematopterus) as donors and the eggs of loach (Misgurnus anguillicaudatus) of Huanghe carp (Cyprinus carpio haematopterus) as recipients. Using loach unfertilized eggs as recipients 0.33% of reconstructed embryos developed in to neural plate from kidney cells, and 0.1% of reconstructed embryos developed in to muscular reaction stage from tail fin cells of F4hGH gene transferred red carp. A tatal of 0.4% reconstructed embryos from cultured tail fin cells of 18th passage and unfertilized eggs of Huanghe carp developed in to myotome stage. All the nuclear transfer embryos were positive for hGH gene by PCR analysis. The results suggest that exogenous hGH gene is stable in the F4hGH gene transferred red carp, hGH gene exists in all the cells of the F4 transgenic red carp. Somatic cell nuclear transfer is an effective pathway for producing homogenous transgenic fish, but with low efficiency. The reasons of low efficiency of somatic nuclear transplantation may be incompatibility of nuclear and ooplasm, incongruity of cell cycle, aberrance of chromosomes, etc.
The expression of salmon insulin-like growth factor I cDNA in mirror carp

Abstract:The fertilized eggs of mirror carp (Cyprinus carpio var. specularis.) were microinjected with an "all-fish" recombinant gene pCAsIGFc, a salmon insulin-like growth factor-I cDNA(sIGFc) capped with a carp β-actin gene promoter and the upstream region in pUC118. The polymerase chain reaction (PCR) assay showed that the transgene was detectable in all recipient embryos and in 80% of the 5 months old founders' caudal fin. For the caudal fin-transgenic founders, however, the transgene was only found in part of other tissues. The reverse transcription-polymerase chain reaction (RT-PCR) assay showed that the transgene expression occurred in 2/3 of gene-transferred embryos and in part of tissues of the 5 months old transgenic founders. The results indicate that existence and expression of the CAsIGFc in tissues of the transgenic mirror carp is heterogeneous.

Many research results showed that outside genes were inserted into the plant genome with recombinant type, and the integration was related to border T-DNA sequence in the course of Agrobaterium mediated transformation. SAR structure was also found in the integration location of transformants with direct DNA transformation method. The pollen-tube pathway transformation, one of direct DNA transformation method, was very successfully used in Bt transgenic cotton in China. But until now there has been no report about Bt gene integration. The aim of this research was to investigate flanking DNA structure in order to explain the mechanism of direct transformation method in the future. The structure of flanking DNA fragments of Bt integration in four different transgenic Bt cotton varieties including Simian-3, 161 resistant breeding line, 161 sensitive breeding line and Guokangmian-1 was analyzed with TAIL-PCR and nested PCR. The flanking DNA sequences of different self-crossed progenies from one plant are the same. In contrary, their DNA sequences are diverse for different breeding lines. Upstream flanking fragments in some transgenic cotton contained short transformation plasmid sequences, downstream flanking fragments in all the transgenic cotton varieties were composed of high percentage AT. The percentage of AT in Simian-3 transgenic Bt variety was as high as 92%. All the flanking fragments were of multi-copies and similar to repetitive sequences. No recognition sites of TOP enzyme were found in these fragments.

Abstract:Using a nuclear transplantation approach, the integration and expression of the green fluorescent protein ( GFP ) gene in the embryogenesis of transgenic loach ( Misgurnus anguillicaudatus Cantor) have been studied. The GFP gene expression is first observed at the gastrula stage, which is consistent with the initiation of cell differentiation of fish embryos. The time course of the foreign gene expression is correlated with the regulatory sequences. The expression efficiency also depends on the gene configuration: the expression of pre-integrating circular plasmid at early embryos is higher than that of the linear plasmid. The integration of the GFP gene is first detected at the blastula stage and lasts for quite a long period. When two types of different plasmids are co-injected into fertilized eggs, the behavior of their integration and expression is not identical.
