Expression of Large Conductance Calcium-Activated Potassium Channels Decreases Cellular Protein Tyrosine Phosphorylation

Abstract:Objective To study the regulation of cellular protein tyrosine phosphorylation by large conductance calcium-activated potassium (BKCa) channels. Methods The change of protein tyrosine phosphorylation was detected after transfection of BKCa, channels or cotransfection of BKCa channels and v-Src in HEK293 cells. The effects of BKCa channel agonist NS1619 and antagonist paxilline were compared on the protein tyrosine phosphorylation of hippocampal neurons in vitro. Results Protein tyrosine phosphorylation by endogenous and expressed v-Src tyrosine kinase was reduced markedly by the expression of BKCa channels in HEK293 cells. The cellular protein tyrosine phosphorylation of hippocampal neurons was decreased by BKq, channels agonist NS1619, but increased by BKq, channel antagonist paxilline. Conclusion Expression of BKCa channels decreases cellular protein tyrosine phosphorylation.
