Effects of Shugan Tongluo Formula on Angiogenesis in Rats with Mammary Gland Hyperplasia

Abstract:Objective To observe effects of Shugan Tongluo Formula,a traditional Chinese prescription for soothing liver-qi and expelling the obstruction from collaterals,on angiogenesis in rats with mammary gland hyperplasia.Methods Eight-four unpregnancy female rats were selected and averagely randomized into seven groups: normal group,model group,tamoxifen group,Rupixiao(a patent traditional Chinese medicine for nodules of the breast) group,and low-,middle-and high-dosage Shugan Tongluo Formula treated groups.The rat model of mammary gland hyperplasia was induced by intramuscular injection of benzestrofol and progesterone.The model rats were treated with corresponding drugs for 30 days,respectively.The breast tissues were dyed by immunohistochemical technique.The positive rates of vascular endothelial growth factor(VEGF),basic fibroblast growth factor(bFGF) and microvessel density(MVD) were observed under light microscope.The average optical density and positive area of bFGF and MVD were analyzed by imaging analysis system.Results Shugan Tongluo Formula could significantly reduce the positive rate,average optical density and positive area of bFGF in mammary gland and statistically decreased the number and positive area of MVD in comparison with those in model group(P<0.05,or P<0.01).Shugan Tongluo Formula showed a tendency in decreasing the positive rate of VEGF and average optical density of MVD,but the difference was insignificant between Shugan Tongluo Formula treated groups and the model group(P>0.05).Conclusion Shugan Tongluo Formula can inhibit the angiogenesis in breast of rats with mammary gland hyperplasia by inhibiting the overexpression of bFGF,and decreasing MVD.
