Abstract:Hemoglobin (Hb) was immobilized successfully on nanometer-sized gold colloid particles associated with a cysteamine monolayer on a gold electrode surface, and was characterized by atomic force microscopy (AFM), UV–vis spectroscopy (UV–vis), surface enhanced Raman spectroscopy (SERS) and electrochemical impedance spectroscopy (EIS). The immobilized Hb was shown to keep its biological activity well. Direct electron transfer (eT) between Hb and the modified electrode was achieved without the aid of any electron mediator. In pH 7.2 phosphate buffer solution, the formal potential (E°′) of Hb was −0.051 V (vs. SCE) and the eT rate constant was 0.49 s−1. The average surface coverage of Hb immobilized on the gold colloid was about 6.71×10−11 mol cm−2. The immobilized Hb displayed the features of a peroxidase and gave an excellent electrocatalytic response to the reduction of H2O2. The Michaelis–Menten constant (Km) was 1.2×10−4 M. The currents were proportional to the H2O2 concentration from 3.6×10−7 to 8.6×10−4 M and the detection limit was as low as 1.2×10−7 M (S/N=3).

Abstract:In this paper, the electrochemical behavior of Hb assembled on glassy carbon (GC) electrode modified with nanostructured gold colloid-silk fibroin (NGC-SF) was investigated. Electrochemical results show that direct electron transfer of the immobilized Hb occurs as indicated by a couple of quasi-reversible redox peaks with a formal potential of −0.38V (vs. SCE) in 0.10M pH 7.0 phosphate buffer. This direct electron transfer process is accompanied by proton exchange and thus the formal potential of Hb changes linearly with solution pH in the range from pH 6.0 to 8.0 with a slope value of −45.2mV/pH. In addition, the immobilized Hb retains its native catalytic activity to H2O2 without the aid of any electron mediator. This indicates that the presence of NGC-SF can provide a suitable microenvironment for Hb and enhance the direct electron transfer between the hemoglobin (Hb) and electrodes.

Abstract:Several aspects of the application of silver electrode to the electrochemical studies of hemoglobin have been discussed in this paper . The silver electrode could not only be used directly as the electrode for the electrochemical studies of hemoglobin, but also react with phenothiazine and benzimidazole to give stable and useful mediator-coated electrodes. In addition, the silver electrode could help sodium dodecyl sulfate to give full play to its promoting effect on the protein.
Direct Electrochemistry of Hemoglobin in Gold Nanowire Array

Abstract:Gold nanowire array has been proven to be efficient support matrixes for the immobilization of hemoglobin (Hb). The vertically oriented nanowire array provides an ordered well-defined 3D structure with nanowire density approximately 5 x 10(8)cm(2). The adsorption of ferritin onto the nanowire surface was visualized by transmission electron microscopy. When Hb was adsorbed, UV-vis absorption and Fourier transform infrared (FT-IR) spectra show no obvious denaturation of Hb in the nanowire array. The Hb-modified nanowire array exerted direct electron transfer and gave a well-defined, nearly reversible redox couple with formal potential of -0.225 V. The quantity of electroactive Hb varied with the changing of the morphology of the electrode and found to increase with the increasing of the nanowire length. Comparisons of voltammetric and quartz crystal microbalance measurements show that 70% of the Hb molecules adsorbed are electroactive when the length of the nanowire was 2 microm. Both of the Hb-modified nanowire array and the unmodified nanowire array demonstrate good electrocatalytic reduction ability for hydrogen peroxide. With the adsorption of glucose oxidase onto the bare nanowire surface, sensitive and selective glucose biosensors can be fabricated.

Abstract:The direct electrochemistry of hemoglobin (Hb) was studied by cyclic voltammetry(CV) and flow injection analysis(FIA) on a silver electrode modified by a self-assembled monolayer of lipoic acid(LA). Lipoic acid molecules can strongly adsorb onto the Ag electrode surface through the cleavage of the S-H bond and the formation of the Ag-S bond, The observed adsorption coverage of LA demonstrates that the LA molecules spontaneously form a self-assembled monolayer. Experimental data show that LA can promote the redox process of Hb at the modified electrode surface. This chemically modified electrode (CME) exhibits good stability in the CV and FIA. Linear-sweep voltammetric measurement of Hb at the CME reveals a linear relationship between the oxidative peak current and the concentration of Hb in the range of 5.0 x 10(-7)-1.5 x 10(-5) mol/L. The relative standard derivation (RSD) for six replicate measurements of 5.0 x 10(-6) mol/L Hb in FIA is 2.8%. The detection limit is 2.0 x 10(-7) mol/L. The reaction mechanism involves the hydrogen bond/salt bridge formation between the carboxylate of LA and the protonated lysine residues of Hb that can enhance the electron transfer reaction. It can be used to detect Hb in teal examples.

Abstract:A novel hemoglobin (Hb) electrode has been presented through adsorbing lib onto graphene-Pt nanocomposite. The graphene-Pt with uniform sheet-like structure was obtained by microwave-assisted method. Positions of Soret absorption band showed lib retained its natural configuration after adsorption onto graphene-Pt. The electrochemical behavior of the immobilized Hb was studied by cyclic voltammetry. The immobilized Hb showed a pair of stable and well-defined peaks at the formal potential of -0.26 V. As a model, a kind of novel third generation biosensor for H(2)O(2) was developed. The resultant heme-protein electrode exhibited electrocatalytical responses to the reduction of H(2)O(2), excellent stability, interference and good reproducibility. The presence of graphene enhanced the protein loadings, accelerates interfacial electron transfer and improves thermal stability of the adsorbed protein. (C) 2011 Elsevier B.V. All rights reserved.

Abstract:The direct electrochemistry of hemoglobin (Hb) immobilized on a hexagonal mesoporous silica (HMS)-modified glassy carbon electrode was described. The interaction between Hb and the HMS was investigated using UV-Vis spectroscopy, FT-IR, and electrochemical methods. The direct electron transfer of the immobilized Hb exhibited two couples of redox peaks with the formal potentials of -0.037 and -0.232 V in 0.1 M (pH 7.0) PBS, respectively, which corresponded to its two immobilized states. The electrode reactions showed a surface-controlled process with a single proton transfer at the scan rate range from 20 to 200 mV/s. The immobilized Hb retained its biological activity well and displayed an excellent response to the reduction of both hydrogen peroxide (H2O2) and nitrate (NO2-). Its apparent Michaelis-Menten constants for H2O2 and NO2- were 12.3 and 49.3 microM, respectively, showing a good affinity. Based on the immobilization of Hb on the HMS and its direct electrochemistry, two novel biosensors for H2O2 and NO2- were presented. Under optimal conditions, the sensors could be used for the determination of H2O2 ranging from 0.4 to 6.0 microM and NO2- ranging from 0.2 to 3.8 microM. The detection limits were 1.86 x 10(-9) M and 6.11 x 10(-7) M at 3sigma, respectively. HMS provided a good matrix for protein immobilization and biosensor preparation.

Abstract:It is reported for the first time that hemoglobin (Hb) was immobilized on the surface of carbon black powders modified at the surface of a glassy carbon electrode. The cyclic voltammetric results showed that the immobilized Hb could undergo a direct quasi-reversible electrochemical reaction. Its formal potential, E-0, is -0.330 V in phosphate buffer solution (pH 6.9) at a scan rate of 100 mV/s and is almost independent of the scan rate in the range of 40-200 mV/s. The dependence of E-0, on the pH of the buffer solution indicated that the conversion of Hb-Fe(III)/Hb-Fe(II) is a one-electron-transfer reaction process coupled with one-proton-transfer. The experimental results also demonstrated that the immobilized Hb retained its bioelectrocatalytic activity for the reduction of H2O2. Furthermore, the immobilized Hb can be stored at 4 degrees C for several weeks without any loss of the enzyme activity. Thus, the immobilized Hb may be used as a biocathodic catalyst in biofuel cells. (c) 2007 Elsevier B.V. All rights reserved.

Abstract:It is one of main challenges to find the suitable materials to enhance the direct electron transfer between the electrode and redox protein for direct electrochemistry field. Nano-structured metal oxides have attracted considerable interest because of unique properties, well biocompatibility, and good stability. In this paper, the copper oxide nanowire bundles (CuO NWBs) were prepared via a template route, and the bioelectrochemical performances of hemoglobin (Hb) on the CuO NWBs modified glass carbon electrodes (denoted as Hb-CuO NWBs/GC) were studied. TEM and XRD were used to characterize the morphology and structure of the as synthesized CuO NWBs. Fourier transform-infrared spectroscopy (FT-IR) proved that Hb in the CuO NWBs matrix could retain its native secondary structure. A pair of well-defined and quasi-reversible redox peaks at approximately -0.325 V (vs. Ag/AgCl saturated KCl) were shown in the cyclic voltammogram curve for the Hb-CuO NWBs/GC electrode, which indicated the direct electrochemical behavior. The Hb-CuO NWBs/GC electrode also displayed a good electrocatalytic activity toward the reduction of hydrogen peroxide. These results indicate that the CuO NWBs are good substrates for immobilization of biomolecules and might be promising in the fields of (bio) electrochemical analysis.

Abstract:Multilayered hemoglobin (Hb) molecules were successfully immobilized on three-dimensional gold film electrode modified with self-assembled monolayers (SAMs) of 3-mercaptopropylphosphonic acid. Direct electrochemistry of the immobilized multilayered Hb occurs with high thermal stability and electrochemical stability. In the multilayered Hb film, the most inner Hb molecules can directly transfer electron with the electrode, while the Hb protein beyond this layer communicates electron with the electrode via protein–protein electron exchange. In addition, the proposed functional interface can greatly enhance electron transfer rate of the immobilized Hb protein (ks=15.8±2.0s−1) due to the increase of roughness of the gold substrate. Under optimized experimental conditions, the multilayered Hb film displays good bioelectrocatalytic activity toward the reduction of hydrogen peroxide. This electrochemical sensor shows fast response (less than 1s), wide linear range (7.8×10−8 to 9.1×10−5M) and low detection limit (2.5×10−8M), which can be attributed to good mass transport, large Hb proteins loading per unit area and fast electron transfer rate of Hb protein.

Positions of Soret absorption band showed Hb retained its natural configuration on the resulting films. Electrochemical impedance spectroscopy and cyclic voltammetry were employed to study the electrochemical behavior of adsorbed Hb. The immobilized Hb showed a pair of stable and well-defined peaks at about −284 and −174mV at 100mVs−1 in pH 6.0 buffers and displayed excellent electrocatalytical responses to the reduction of H2O2, NO2- and trichloroacetic acid. The presence of meso-WO3 film greatly enhances the protein loadings, accelerates interfacial electron transfer and improves thermal stability of the adsorbed protein.

Abstract:Direct electrochemistry was carried out to investigate the interaction between haemoglobin (Hb) and nitric oxide (NO), employing a Hb-DNA film modified electrode. DNA was used as an electrochemical "promoter" for the direct electron transfer of Hb at the electrode surface. Aside from a pair of peaks corresponding to the redox reaction of Hb, two additional peaks were observed in a solution containing NO. The pre-peak, located at -0.070 V vs. SCE, was proposed to come from the chemical redox reaction, i.e., NO-induced oxidation of Hb in the film, coupled electron transfer process of Hb. The other peak, at -0.684 V vs. SCE, was assigned to the reduction of heme-ligated NO. These phenomena were proposed to be related to the electron transfer and binding process between Hb and NO.

Abstract:Abstract Hemoglobin (Hb) was successfully immobilized on gold nanoparticles (GNP) associated with a 1,4‐benzenedimethanethiol (BDT) monolayer on a gold electrode surface and was characterized by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). Direct electron transfer (DET) between Hb and the modified electrode was achieved without the aid of any electron mediator. GNP can greatly promote the redox of Hb, on which a pair of obvious CV peaks for Hb Fe(III)/Fe(II) redox couple were obtained in pH 6.2 phosphate buffer solution (PBS). In pH 6.2 PBS, the formal potential (E°') of Hb was −0.180 V (versus SCE), and the electron transfer rate constant was 5.2 s−1. The formal potential of Hb on gold colloidal modified electrode shifts linearly with pH with a slope of −55.5 mV/pH at 20°C, showing that the electron transfer is accompanied by single proton transportation. The high efficiency of the Hb/GNP/BDT modified gold electrode toward the catalytic electroreduction of hydrogen peroxide has been observed. The immobilized Hb gave an excellent electrocatalytic response to H2O2. Potential application of Hb/GNP/BDT modified gold electrode as biosensors to monitor H2O2 is proposed.

Abstract:Nanoparticles (NPs) consisting of an Fe3O4 core and a thin gold shell (referred to as Au@Fe3O4 NPs) were self-assembled on the surface of a glassy carbon electrode modified with ethylenediamine. Following adsorption of hemoglobin, its interaction with the NPs was studied by UV–Vis spectroscopy, electrochemical impedance spectroscopy, and cyclic voltammetry. Stable and well-defined redox peaks were observed at about −350 mV and −130 mV in pH 6.0 buffer. The modified electrode was used as a mediator-free sensor for hydrogen peroxide (H2O2), with a linear range from 3.4 µM to 4.0 mM of H2O2, and with a 0.67 µM detection limit (at an S/N of 3). The apparent Michaelis-Menten constant is 2.3 mM.

Abstract:Hemoglobin (Hb) was successfully immobilized in dimethyldioctadecyl ammonium bromide (DOAB) film at pyrolytic graphite (PG) electrode. Electrochemical experiments revealed that Hb in DOAB film exhibited a pair of well-defined, quasi-reversible cyclic voltammetric peaks at about -0.160 V versus saturated calomel electrode (SCE) in pH 5.0 buffer, characteristic of the heme Fe(III)/Fe(II) redox couple of Hb. The electron transfer (eT) rate between Hb and the PG electrode was 0.10 s(-1). Positions of the Soret absorbance band indicated that the Hb retained its secondary structure and was similar to its native state. Furthermore, the Hb in DOAB film acted as a biological catalyst towards the reduction of nitric oxide (NO). Abstract:Titanium oxide nanotubes (TiO2-NTs) synthesized by the hydrothermal method had been prepared as the co-immobilization matrix to incorporate hemoglobin (Hb) successfully. The nanostructures of TiO2-NTs were investigated by X-ray diffraction and high-resolution electron microscopy. The Hb immobilized in TiO2-NTs had a similar structure to the native of Hb and retained its near-native conformations as characterized by the UV–vis and FT-IR spectroscopy. A couple of quasi-reversible redox peaks with a formal potential of −0.34V (vs. SCE) in 0.10M pH 7.0 phosphate buffered saline (PBS) were observed. The amperometric response of the immobilized Hb linearly to H2O2 concentration ranged from 4μM to 64μM with a detection limit of 4.637×10−6M and the high stability of the immobilized Hb in TiO2-NTs constituted a promising platform for the development of biosensors.

Abstract:Using chitosan as an effective linker between CMK-3 and glassy carbon electrode surface, {Hb/CMK-3}n multilayer film-modified electrodes were constructed through layer-by-layer assembly. The morphology of thus-formed {Hb/CMK-3}n film was characterized by scanning electron micrographs, and the interaction of hemoglobin (Hb) with CMK-3 was studied by UV–vis spectroscopy and electrochemical methods. Under optimal conditions, {Hb/CMK-3}6 film showed a couple of stable and well-defined redox peaks at about −377 and −296mV in pH 7.0 buffers. Furthermore, the {Hb/CMK-3}6 film displayed excellent electrocatalysis to the reduction of both H2O2 and O2. Based on thus-formed film and its direct electron transfer behavior, a novel biosensor was presented for the determination of H2O2 ranging from 1.2 to 57μM with the detection limit of 0.6μM at S/N=3. CMK-3 provided a desirable matrix for protein immobilization and biosensor preparation.
Original Article Electrochemistry of Hemoglobin I from Lucina pectinata immobilized on a modified gold electrode with 3-mercaptopropionic acid

Abstract:The extraordinary affinity of recombinant hemoglobin I from Lucina pectinata (rHbI) for hydrogen sulfide (H2S) allows us to postulate it as substrate for the preparation of future hydrogen sulfide sensor. The modification of a gold surface with 3- mercaptopropionic acid (3-MPA), nickel ion and recombinant hemoglobin I, and electrochemical kinetics parameters were reported here. Cyclic Voltammetry and X-ray photoelectron spectroscopy were employed to characterize the electrode modification process, while FTIR was used to verify the presence of hemoglobin on the surface. A pair of well-defined redox peaks for rHbIFe(III)-rHbIFe(II) was obtained. Electrochemical parameters of immobilized hemoglobin as formal potential (E o" ), charge transfer coefficient (α) and apparent heterogeneous electron transfer rate constant (ks) were estimated by cyclic voltammetry data. Also, the electroactivity of this hemoglobin-electrode was exemplified at the prepared electrode for association with hydrogen sulfide.

Abstract:Hemoglobin can exhibit a direct electron-transfer reaction after being entrapped in a SP Sephadex membrane. A pair of stable and well-defined redox waves are obtained at a hemoglobin-SP sephadex modified pyrolytic graphite electrode. The anodic and cathodic peak potentials are located at -0.244 and -0.336 V (vs SCE), respectively. On the other hand, the peroxidase activity of the protein in the membrane is also greatly enhanced. The apparent Michaelis-Menten constant is calculated to be 1.9 mM, which shows a large catalytic activity of hemoglobin in the SP Sephadex membrane toward hydrogen peroxide (H2O2) According to the direct electron-transfer property and enhanced peroxidase activity of Hb in the membrane, a Hb/SP Sephadex membrane-based H2O2 biosensor is prepared, with a linear range similar to5.0 x 10(-6) to 1.6 x 10(-4) mol/L.

Abstract:The direct electron transfer between hemoglobin (Hb) and the glassy carbon electrode (GC) can be readily achieved via a high biocompatible composite system based on biopolymer chitosan (CHT) and TiO2 nanorods (TiO2-NRs). TiO2-NRs greatly promote the electron transfer between Hb and GC, which contribute to the higher redox peaks. UV-vis spectra result indicated the Hb entrapped in the composite film well keep its native structure. The immobilized Hb remains its bioelectrocatalytical activity to the reduction of H2O2 with a lower detection limit. A novel, sensitive, reproducible and stable electrochemical biosensing platform of H2O2 based on Hb-TiO2-CHT electrode is explored.
