Relationship Between Spinal Glial Cell Activation and Excitatory Amino Acid in Rats with Prostatitis

Abstract:Objective:To observe the expression of phosphorylated p38MAPK(p-p38),glial cell activation and changes of excitatory amino acid in rat prostatitis models induced by intrathecal injection of SB203580.Methods:45 male SPF rats were divided into the normal group(5 rats),prostatitis group(10 rats)and the group of chronic prostate pain treated with SB203580(SB203580 group,30 rats).In SB203580 group,complete Freund's adjuvant was injected and intrathecal administration of 0.5,2.5 and 12.5 μg/10 μl SB203580 was applied respectively in every five rats.After 5 and 10 days,Western blot and ELISA were used to determine the changes of p-p38 and glial fibrillary acidic protein(GFAP)in the posterior horn of the L5~S2 spinal cord.Glutamic acid(Glu)and aspartate(Asp)changes were assayed by amino acid analyzer.Results:In prostatitis group,p-p38,GFAP,Glu and Asp increased significantly with time passing,while these indexes in SB203580 group significantly reduced compared with those in prostatitis group.GFAP level was positively correlated with the levels of Glu and Asp.The correlation coefficient was 0.457(P=0.0016)and 0.498(P=0.0005),respectively.Conclusion:Glial cell activation is an important reason for the excitatory amino acid changes in spinal dorsal horn of rats suffering from the chronic prostate pain.Blocking p38MAPK signaling pathway can effectively reduce the glial cell activation and the level of excitatory amino acids.
