A Potential Link Between Aromatics-Induced Oviposition Repellency Behaviors and Specific Odorant Receptor of Aedes Albopictus

Abstract:BACKGROUNDThe Asian tiger mosquito, Aedes albopictus, is a competent vector for the spread of several viral arboviruses including dengue, chikungunya, and Zika. Several vital mosquito behaviors linked to survival and reproduction are primarily dependent on a sophisticated olfactory system for semiochemical perception. However, a limited number of studies has hampered our understanding of the relationship between the A. albopictus acute olfactory system and the complex chemical world. RESULTSHere, we performed a qRT-PCR assay on antennae from A. albopictus of differing sex, age and physiological states, and found that AalbOr10 was enriched in blood-fed female mosquitoes. We then undertook single sensillum recording to de-orphan AalbOr10 using a panel of physiologically and behaviorally relevant odorants in a Drosophila 'empty neuron' system. The results indicated that AalbOr10 was activated by seven aromatic compounds, all of which hampered egg-laying in blood-fed mosquitoes. Furthermore, using a post-RNA interference oviposition assay, we found that reducing the transcript level of AalbOr10 affected repellent activity mediated by 2-ethylphenol at low concentrations (10(-4) vol/vol). Computational modeling and molecular docking studies suggested that hydrogen bonds to Y68 and Y150 mediated the interaction of 2-ethylphenol with AalbOr10. CONCLUSIONWe reveal a potential link between aromatics-induced oviposition repellency behaviors and a specific odorant receptor in A. albopictus. Our findings provide a foundation for identifying active semiochemicals for the monitoring or controlling of mosquito populations. (c) 2024 Society of Chemical Industry.

Abstract:Moths rely on plant volatiles to locate appropriate plants for feeding and laying eggs. While extensive research has been conducted on the global agricultural pests, Spodoptera frugiperda and Spodoptera litura, their molecular mechanisms for detecting plant volatiles remain mostly unknown. Here, we have demonstrated that nonanal, a common plant volatile, is attractive for both virgin and gravid females of the two species. Second, we have identified a conserved odorant receptor clade (SfruOR47 clade) that is primarily tuned to nonanal. Finally, by three-dimensional (3D) structure prediction, molecular docking, and site-directed mutagenesis, we have revealed that the His57 and Glu61 residues, also shared by other six orthologous ORs, are essential for nonanal binding in SfruOR47 and SlituOR9, indicating the conserved structure and function of ORs in the SfruOR47 clade. These findings offer novel insights into the molecular mechanisms and evolutionary aspects of moth behavior in response to plant volatiles.

Abstract:A variety of microorganisms inhabit the human skin, including bacteria, fungi, and viruses. Skin microbiota plays an important role in maintaining host skin homeostasis and the skin barrier function. Volatiles produced by skin microorganisms in different individuals have varying effects on the regulation of host-seeking behavior in the mosquito. In this review, we summarize the diversity of skin microorganisms, the volatiles produced by skin microorganisms and the regulatory effects of skin microorganisms on mosquito-borne infectious diseases, which could provide clues for developing new strategies for the prevention and intervention of mosquitoborne infectious diseases.

In this review, we summarized the sources and factors of human volatiles and their effects on mosquito behavior, which may provide a new approach to the development of mosquito attractants and repellents.

Abstract:Olfaction plays crucial role in mosquito host seeking,oviposition and searching for sugar sources. However, the role of semiochemicals in mating behavior remains speculative. In this paper we summarized the research on mosquito olfaction and chemical ecology in the past 20 years. The olfactory receptors in the sensilla of the antenna, maxillary palps and proboscis respond to the odors from the environment. The olfactory transduction and neural encoding and processing in the peripheral olfactory pathway start from the binding of odorant binding receptors (OBP) with odors. The changes in sensitivity and response profile of olfactory receptor neurons as a result of blood meal are involved in modulating behavior of mosquito females. The carbon dioxide, lactic acid and other odors released from animal and human skins attract mosquito upwind flight to the source in the host seeking. The volatiles from the oviposition site and the pheromone compounds originating from eggs, larvae and pupae have been found attractive to the gravid females. The plant volatiles especially the flower fragrance helps mosquito adults search for sugars. Current repellents act by interfering, directly or indirectly, with olfaction. Some repellents inhibit the mosquito orientation flight by impeding its olfaction. It could be promising to find the active substances in the odors from animals, human skins, plants or oviposition sites. The understanding of the olfactory mechanism can allow us to develop new and effective tools for mosquito surveillance and control.

Abstract:Vacuolar-type H+-ATPases (V-ATPases) play a crucial role in the life cycle of agricultural pests and represent a promising target for the development of novel insecticides. In this study, S18, a derivative of vanillin acquired from Specs database using a structure-based virtual screening methodology, was first identified as a V-ATPase inhibitor. It binds to subunit A of the enzyme with a Kd of 1 nM and exhibits insecticidal activity against M. separata. Subsequently, using S18 as the lead compound, a new series of vanillin derivatives were rationally designed and efficiently synthesized. and their biological activities were assessed. Among them, compound 3b-03 showed the strongest insecticidal activity against M. separata by effectively targeting the V-ATPase subunit A with Kd of 0.803 μM. Isothermal titration calorimetric measurements and docking results provided insights into its interaction with subunit A of V-ATPase, which could facilitate future research aimed at the development of novel chemical insecticides.

Abstract:Mosquito-borne diseases constitute a major health issue and a high burden for many countries. The biocontrol of mosquitoes is vital for combatting such diseases. Bacteria exhibit huge potential biotechnological uses due to their adaptability and ubiquity. Volatile organic compounds (VOCs) are typically molecules with low molecular weight and high vapor pressure. The volatiles produced by bacteria are complex and can impact insect behavior. This study aimed to compare the volatiles produced by the repellent Bacillus licheniformis strain Ba1 with a closely related, yet non-repulsive one (B957). The strains were isolated from extreme environments in Morocco (desert dunes and hot springs) and their resistance to both high temperature and salinity was investigated. After cultivation, the VOCs were extracted and identified by GC-MS. The VOCs detected showed little difference between the two strains, however, their relative abundance varied greatly, suggesting a dose-dependent effect. Ba1 was found to produce both repulsive and attractive compounds as well as molecules with larvicidal effects mainly found in plant extracts. To the best of our knowledge, this is the first study of VOCs emitted by Bacillus sp. With a repulsive effect against mosquitoes.

Abstract:This study explores the role of floral volatile organic compounds (FVOCs) in insect behavior, focusing on Aquilaria sinensis (AS), a valuable tropical plant threatened by Heortia vitessoides Moore. Despite H. vitessoides' attraction to AS and non-host plants like Elaeocarpus decipiens (ED) and Dalbergia odorifera (DO), little is known about their chemical interactions. FVOCs from these plants were analyzed at 9:00 and 18:00 using GC×GC-QTOF-MS and HS-SPME. The results showed that ED exhibiting the highest concentration (92.340 ng/mg), followed by DO (75.167 ng/mg) and AS (64.450 ng/mg). Through GC-EAD and EAG, a total of 11 FVOC compounds with electrophysiological activates were identified. These compounds, except linalool, showed dose-dependent responses. Y-Tube bioassays confirmed phenylethyl alcohol or the mixture of EAD-active compounds produced positive chemotactic responses in both males and females. FVOCs have the potential to be used as a natural and sustainable alternative to chemical insecticides in pest control.

Abstract:Mosquito-borne viral diseases are a major concern of global health and result in significant economic losses in many countries. As natural vectors, mosquitoes are very permissive to and allow systemic and persistent arbovirus infection. Intriguingly, persistent viral propagation in mosquito tissues neither results in dramatic pathological sequelae nor impairs the vectorial behavior or lifespan, indicating that mosquitoes have evolved mechanisms to tolerate persistent infection and developed efficient antiviral strategies to restrict viral replication to nonpathogenic levels. Here we provide an overview of recent progress in understanding mosquito antiviral immunity and advances in the strategies by which mosquitoes control viral infection in specific tissues.

Abstract:Fruits produce a range of volatile compounds that make up their characteristic aromas and contribute to their flavor. Fruit volatile compounds are mainly comprised of esters, alcohols, aldehydes, ketones, lactones, terpenoids and apocarotenoids. Many factors affect volatile composition, including the genetic makeup, degree of maturity, environmental conditions, postharvest handling and storage. There are several pathways involved in volatile biosynthesis starting from lipids, amino acids, terpenoids and carotenoids. Once the basic skeletons are produced via these pathways, the diversity of volatiles is achieved via additional modification reactions such as acylation, methylation, oxidation/reduction and cyclic ring closure. In this paper, we review the composition of fruit aroma, the characteristic aroma compounds of several representative fruits, the factors affecting aroma volatile, and the biosynthetic pathways of volatile aroma compounds. We anticipate that this review would provide some critical information for profound research on fruit aroma components and their manipulation during development and storage.

Abstract:Background: The development of economically viable and environmentally neutral tools to control insects that consume or damage over 20% of global agriculture or vector human and animal disease represents one of the most important challenges of the 21st century. The suite of chemical-based strategies currently employed to control insect populations rely primarily on insecticides, which are subject to rapid resistance and often have harmful off-target environmental and health-related impacts, and, to a lesser degree, repellents, which typically rely on masking attractive odors. The discovery and characterization of Vanderbilt University allosteric agonists (VUAAs), a family of small-molecule agonists that target the highly conserved, insect-specific odorant receptor coreceptor (Orco), raise the potential for the development of a novel repellent paradigm for vector/pest management. VUAAs have the potential to target nearly all insect olfactory sensory neurons, leading to highly aversive behavioral responses, but importantly have limited volatility, thereby reducing their utility as spatial repellents. Results: We have characterized VUAA thermolysis components and identified a suite of volatiles (VUAA-based active ingredients, VUAIs) that act specifically in novel binary combinations as robust and long-lasting spatial repellents against Anopheline mosquitoes. In mobility-based behavioral experiments, VUAIs act synergistically as effective spatial repellents and outperform parent VUAA compounds against host-seeking Anopheline mosquitoes. Conclusions: VUAIs are volatile alternatives to Vanderbilt University allosteric agonists (VUAAs) that have the potential for use as spatial repellents in disease vector and agricultural pest control. The repellency observed is odorant receptor coreceptor (Orco)-dependent, supporting the hypothesis that VUAIs and VUAAs similarly target an allosteric Orco recognition site. © 2024 The Author(s). Abstract:Mosquitoes (Diptera: Culicidae) are arthropod vectors involved in transmitting deadly infectious diseases to humans and animals, causing several million human deaths every year worldwide. Plant‐based products provide environment‐friendly mosquitocidal properties and are harmless to humans, non‐target organisms, and the environment. This review covers up‐to‐date information about plant products (plant compounds, extracts, and essential oils), which are utilized to control all the life cycle stages (egg, larva, pupa, and adult) of the mosquito genera Aedes, Anopheles, and Culex. Mosquitoes pose a severe threat to the environment as vectors of numerous harmful diseases affecting humans and animals. Mosquitoes transmit pathogens that cause dengue, chikungunya, malaria, zika, yellow fever, Japanese encephalitis, and filariasis. Today's mosquito control strategies heavily rely on the use of chemical insecticides such as N,N‐diethyl‐meta‐toluamide (DEET), N,N‐diethyl mandelic acid amide (DEM), and dimethyl phthalate (DMP). However, the widespread use of chemical insecticides has resulted in pollution, bio‐magnification, and other health and environmental issues. It has also become ineffective because of the mosquitoes' aptitude to develop resistance, emphasizing the urgent need for safe, effective, and long‐lasting strategies. An alternative and promising approach to circumventing these obstacles involves the implementation of insecticides made from natural compounds found in plants. Therefore, the scientific community has shifted its focus towards plant‐based phytochemicals, oils, and extracts, as these are eco‐friendly, safe, and cost‐effective alternatives to conventional chemical insecticides. This review aims to provide details on current advances in plant‐based products (plant compounds, extracts, and essential oils), which are used to control all the life cycle stages (egg, larva, pupa, and adult) of the mosquito genera Aedes, Anopheles, and Culex. Hopefully, this review will pave the way to devise control strategies against these challenging pests.

Abstract:We often consider mosquitoes through an 'anthropocentric lens' that disregards their interactions with nonhuman and nonpathogenic organisms, even though these interactions can be harnessed for mosquito control. Mosquitoes have been recognized as floral visitors, and pollinators, for more than a century. However, we know relatively little about mosquito-plant interactions, excepting some nutrition and chemical ecology-related topics, compared with mosquito-host interactions, and frequently use flawed methodology when investigating them. Recent work demonstrates mosquitoes use multimodal sensory cues to locate flowers, including ultraviolet visual cues, and we may underestimate mosquito pollination. This review focuses on current knowledge of how mosquitoes locate flowers, floral visitation assay methodology, mosquito pollination, and implications for technologies such as sterile male mosquito release through genetic control programs or Wolbachia infection.
Enhanced repellency of binary mixtures of Zanthoxylum armatum seed oil, vanillin, and their aerosols to mosquitoes under laboratory and field conditions

Abstract:The repellency of Zanthoxylum armatum seed oil (ZA-SO), alone or in combination with vanillin (VA), its six major constituents, and another four major previously known Zanthoxylum piperitum fruit oil constituents, as well as aerosol products containing 5 or 10% ZA-SO and 5% VA, was evaluated against female Aedes aegypti in laboratory and field studies. Results were then compared with those of N,N-diethyl-3-methylbenzamide (DEET) as a standard. Hand in cage laboratory tests showed that 0.2, 0.1, and 0.05 mg/cm2 ZA-SO resulted in > 92% protection through 30-min postexposure and was not significantly different than 0.05 mg/cm2 DEET. Skin treated with linalool and limonene (from Z. armatum) provided > 80% repellency to female Ae. aegypti at 10-min exposure, whereas cuminaldehyde, citronellal, geranyl acetate, and cuminyl alcohol (from Zanthoxylum piperitum) provided > 90% protection during this same time period. Only cuminaldehyde and citronellal provided complete protection comparable to DEET at 10-min postexposure. After that time, repellency of all plant constituents to mosquitoes was considerably decreased (< approximately 65%). An increase in repellency and duration of effectiveness was produced by a binary 1:4 mixture of ZA-SO and VA (0.05:0.2 mg/cm2) that was significantly more effective than 0.05 mg/cm2 DEET through 90 min. In field tests, an aerosol formulation containing 5 or 10% ZA-SO plus 5% VA gave 100% repellency at 60-min postexposure. Although these formulations were equal to the level of protection afforded by 10% DEET, repellency to the binary ZA-SO aerosol formulations at 90 min was significantly less effective than DEET. However, mixtures formulated from ZA-SO and VA merit further study as potential repellents for protection of humans and domestic animals from biting and nuisance caused by mosquitoes.
Insecticidal and Antifeedant Activities of Malagasy Medicinal Plant (Cinnamosma sp.) Extracts and Drimane-Type Sesquiterpenes against Aedes aegypti Mosquitoes

Abstract:The overuse of insecticides with limited modes of action has led to resistance in mosquito vectors. Thus, insecticides with novel modes of action are needed. Secondary metabolites in Madagascan plants of the genus Cinnamosma (Canellaceae) are commonly used in traditional remedies and known to elicit antifeedant and toxic effects in insect pests. Here we test the hypothesis that extracts of Cinnamosma sp. enriched in drimane sesquiterpenes are toxic and/or antifeedant to the yellow fever mosquito Aedes aegypti. We show that the bark and root extracts, which contain a higher abundance of drimane sesquiterpenes compared to leaves, were the most efficacious. Screening isolated compounds revealed cinnamodial to be the primary driver of adulticidal activity, whereas cinnamodial, polygodial, cinnafragrin A, and capsicodendrin contributed to the larvicidal activity. Moreover, an abundant lactone (cinnamosmolide) in the root extract synergized the larvicidal effects of cinnamodial. The antifeedant activity of the extracts was primarily contributed to cinnamodial, polygodial, and cinnamolide. Parallel experiments with warburganal isolated from Warburgia ugandensis (Canellaceae) revealed that aldehydes are critical for-and a hydroxyl modulates-insecticidal activity. Our results indicate that plant drimane sesquiterpenes provide valuable chemical platforms for developing insecticides and repellents to control mosquito vectors.
