Abstract:Flos Trollii, the flowers of Trollius chinensis Bunge, has been widely used in Chinese and Mongolian medicine for its efficacy of heat-clearing and detoxification. This drug has both medicinal and edible applications, and has led to various pharmacognosy, natural product chemistry, and pharmacology studies. As a result, its chemical constituents and bioactivities have been well-characterized in recent years. Nevertheless, a couple of critical issues, such as the major effective components, are still unresolved. The present review summarizes research progress on this drug regarding the constituents and bioactivities based on investigations in these laboratories and the results reported in recent publications. In addition, the pending issues are discussed and constructive suggestions for further investigation are proposed.

Abstract:Trollius chinensis Bunge (TCB) is a perennial plant of the Ranunculaceae family with medicinal and edible values. It is widely distributed and commonly used in various regions, including Asia, Europe, and North America. The main chemical components of TCB include alkaloids, flavonoids, phenolic acids, and volatile oil compounds. TCB is renowned for its anti-inflammatory, heat-clearing, detoxifying, and eyesight-improving properties. Its dried flowers are commonly used as a traditional Chinese medicine indicated for the treatment of upper respiratory tract infections, chronic tonsillitis, pharyngitis, influenza, and bronchitis. Modern pharmacology has demonstrated the anti-cancer, anti-inflammatory, antihypertensive, and antioxidant effects of TCB. This study presents a comprehensive overview of various aspects of TCB, including herbal textual research, botany, phytochemistry, pharmacology, traditional uses, clinical application, and quality control, aiming to provide new ideas on the scientific application of TCB as well as the integration of modern research with traditional medicinal uses.

Abstract:Trollius chinensis Bunge, a perennial herb belonging to the Ranunculaceae family, has been extensively used in traditional Chinese medicine. Documented in the Supplements to the Compendium of Materia Medica, its medicinal properties encompass a spectrum of applications, including heat clearance, detoxification, alleviation of oral/throat sores, earaches, eye pain, cold-induced fever, and vision improvement. Furthermore, T. chinensis is used in clinical settings to treat upper respiratory infections, pharyngitis, tonsillitis, esoenteritis, canker, bronchitis, etc. It is mainly used to treat inflammation, such as inflammation of the upper respiratory tract and nasal mucosa. This comprehensive review explores the evolving scientific understanding of T. chinensis, covering facets of botany, materia medica, ethnopharmacological use, phytochemistry, pharmacology, and quality control. In particular, the chemical constituents and pharmacological research are reviewed. Polyphenols, mainly flavonoids and phenolic acids, are highly abundant among T. chinensis and are responsible for antiviral, antimicrobial, and antioxidant activities. The flower additionally harbors trace amounts of volatile oil, polysaccharides, and other bioactive compounds. The active ingredients of the flower have fewer side effects, and it is used in children because of its minimal side effects, which has great research potential. These findings validate the traditional uses of T. chinensis and lay the groundwork for further scientific exploration. The sources utilized in this study encompass Web of Science, Pubmed, CNKI site, classic monographs, Chinese Pharmacopoeia, Chinese Medicine Dictionary, and doctoral and master's theses.

Abstract:A new compound, named trollioside, together with nine known compounds, proglobeflowery acid, isoswertisin, isoswertiajaponin, cirsimaritin, veratric acid, vitexin, orientin, beta-sitosterol and daucosterol, were isolated from the ethanol extract of the dried flowers of Trollius chinensis Bunge. The isolation and structural determination of these compounds are discussed.

Abstract:A new compound, named as trolline was isolated from the ethanol extract of the flowers of Trollius chinensis Bunge. The chemical structure of this compound was elucidated by NMR, MS, IR, UV spectra and single-crystal X-Ray analysis. The in vitro antibacterial and antiviral experiments revealed that trolline exhibited appreciable antibacterial activity against respiratory bacteria such as Staphylococcus aureus, Streptococcus pneumoniae and Klebsiella pneumoniae. Trolline was also demonstrated to have moderate antiviral activity against influenza virus A and B.
Evaluation of antioxidant and antibacterial properties of extracts from Trollius chinensis Bunge

Abstract:Trollius chinensis Bunge is a traditional edible and medicinal plant utilized popularly in north China. This study is intended to evaluate the antioxidant and antimicrobial properties of extracts obtained with different processing methods from Trollius chinensis Bunge. The results showed that all extract displayed moderate or great potential of antioxidant and antibacterial activity, especially ultrasound-assisted ethanol extracts (UAE). It exhibited the highest 2, 2-diphenyl-1-picrylhydrazyl (DPPH; EC50 0.215 ± 1.10 mg/mL), 2, 2-azino-bis (3-ethylbenzo-thiazoline-6-sulfonic acid) (ABTS; EC50 < 10 μg/mL) and reducing power (RP; EC50 0.25 ± 0.02 mg/mL) with the highest level of flavonoids content (71.56 ± 0.35 mg/g) and total phenolic (35.75 ± 0.35 mg/g). In addition, the UAE had high antibacterial activity against Staphylococcus aureus (19.7 ± 0.6 mm inhibition zone), Bacillus subtilis (16.4 ± 0.3 mm inhibition zone), Escherichia coli (15.3 ± 0.2 mm inhibition zone), respectively. The HPLC analysis show that the main flavonoids components in T. chinensis Bunge were revealed to be 2″-O-β-L-galactopyranosylorientin, orientin, vitexin, hyperin. These results suggested the extracts of T. chinensis Bunge could be potentially used as a promising source of antioxidant and antibacterial agents for using in the food or drug industries.

Abstract:Context: The flowers of Trollius chinensis Bunge (Ranunculaceae), used for respiratory tract infections, mainly contain flavonoids, phenolic acids, and alkaloids; however, the effective components are debatable because of their unclear in vivo activities.Objective: This study investigates the effective components from the perspectives of biotransformation and absorption.Materials and methods: Both single person derived- and multiple people-derived intestinal florae were used to investigate the biotransformation of aqueous extract of the flowers of T. chinensis (AEOF) at the concentrations of 15.0, 30.0, and 60.0mg/mL, respectively, for 72h. Both human colon adenocarcinoma cell line (Caco-2) monolayers and everted gut sacs were employed to evaluate the intestinal absorption of the intestinal bacterial transformed AEOF at the concentrations of 10, 20, and 30mg/mL, respectively, for 180min.Results: 2 '' O-beta-L-Galactopyranosylorientin, orientin, vitexin, quercetin, veratric acid, proglobeflowery acid, and trolline in AEOF were not transformed by intestinal bacteria, while isoquercetin and trollioside were completely transformed. The P-app values of 2 '' O-beta-L-galactopyranosylorientin, orientin, and vitexin calculated based on the experimental data of intestinal absorption were at the levels of 10(-5), whereas those of veratric acid, proglobeflowery acid, and trolline were at 10(-4). The mass ratio of flavonoids to phenolic acids to alkaloids changed from 16:10:7 to 9:12:8 before and after absorption.Discussion and conclusion: The dominant position of flavonoids was replaced by phenolic acids after absorption. In addition to flavonoids which are usually considered as the dominant effective ones, phenolic acids and alkaloids should be also very important for the efficacy of these flowers.

Abstract:Curcuma wenyujin is an indigenous medicinal material cultivated in Zhejiang province,China.The domestic and foreign scholars have conducted studies on the chemical components of Curcuma wenyujin.More than one hundred and thirty chemical components were isolated and identificated.Researches showed that the main chemical components of it was volatile file://C:\Users\user\AppData\Local\youdao\DictBeta\Application\7.1.0.0421 \resultui\dict\? keyword =oil,it amounted to one hundred and thirteen categories.Curcuma wenyujin is regarded as one of the traditional precious Chinese herbal medicines.Modern researches showed that its chemical constituents have pharmacological activities such as anti-tumor,anti-flammatory,anti-influenza virus,preventing and treating cardiovascular and cerebrovascular diseases,affecting nervous system and protecting kidney.Because of a long period of continuous cropping and vegetative propagation,its germchit degraded and the pathogenic diseases in Curcuma wenyujin were becoming more and more serious.This situations influenced both the output and quality of it.In this article,we described the symptomes and pathogens of main pathogenic diseases and summarized control measures in Curcuma wenyujin.
Extensive Profiling of Polyphenols from Two Trollius Species Using a Combination of Untargeted and Targeted Approaches

Abstract:Various species of globeflowers, belonging to the genus Trollius, have been extensively used in traditional Chinese medicine due to their anti-inflammatory, antimicrobial, and antiviral properties, which are mainly attributed to their high polyphenol content. Differences in polyphenol composition, and abundances, will lead to varying treatment efficacies of globeflowers. Herein, we employ a combination of targeted and untargeted mass spectrometry (MS) approaches to characterize and quantify a comprehensive array of polyphenols, mainly including flavonoids and phenolic acids in two globeflower species commonly used in Chinese medicine, Trollius chinensis Bunge and Trollius ledebouri Reichb. In addition, free radical scavenging activity was investigated to evaluate the association between polyphenol composition and antioxidation capacity. Liquid chromatography (LC)-based separation and multiple-reaction-monitoring (MRM) transitions were optimized using a library of 78 polyphenol reference compounds to achieve absolute quantification on triple quadrupoles MS (QqQ). The analytical method was further expanded via high-resolution MS to provide relative quantitation of an additional 104 endogenous polyphenols in globeflowers not included in our reference library. Our results revealed stark differences in polyphenol content between T. chinensis and T. ledebouri, emphasizing the need for systematic characterization of polyphenol composition to ensure treatment efficacy and consistency in standardizing the use of globeflowers in Chinese medicine.
Antibacterial and antioxidant activities of the endophytic fungi from medicinal herb Trillium tschonoskii

Abstract:Fourteen distinct endophytic fungal isolates were first separated from the healthy underground parts of Trillium tschonoskii, which is a traditional medicinal herb mainly distributed in China. They were taxonomically identified by morphological traits and internal transcribed spacer (ITS) rRNA gene sequence analysis. Seven genera, namely, Acremonium, Colletotrichum, Fusarium, Leptodontidium, Paraconiothyrium, Plectosphaerella, and Volutella, were identified on the basis of their morphological characterizations. Their ITS-rDNA sequences were compared with those available in the GenBank database to obtain the closest related species by BLAST analysis. A modified thin layer chromatography-bioautography assay was used to detect the antibacterial activity of the n-butanol extracts of the mycelia and culture filtrates of the isolates. Most of the fungal isolates were screened to have antibacterial activity, and antibacterial components were examined to mainly exist in the filtrate except isolates Trtsf09 and Trtsf14. Five isolates, that is, Trtsf05, Trtsf06, Trtsf11, Trts12 and Trts13, were also screened to have antioxidant activity. The endophytic fungi isolated from T. tschonoskii could be an alternative source to produce antibacterial and antioxidant agents.

Abstract:The flowers of Trollius species, named Jin Lianhua in Chinese, are widely used traditional Chinese herbs with vital biological activity that has been used for several decades in China to treat upper respiratory infections, pharyngitis, tonsillitis, and bronchitis. We developed a rapid and reliable method for simultaneous quantitative analysis of 19 flavonoids in trollflowers by using high-performance liquid chromatography (HPLC). Chromatography was performed on Inertsil ODS-3 C18 column, with gradient elution methanol–acetonitrile–water with 0.02% (v/v) formic acid. Content determination was used to evaluate the quality of commercial trollflowers from different regions in China, while three Trollius species (Trollius chinensis Bunge, Trollius ledebouri Reichb, Trollius buddae Schipcz) were explicitly distinguished by using hierarchical clustering analysis. The linearity, precision, accuracy, limit of detection, and limit of quantification were validated for the quantification method, which proved sensitive, accurate and reproducible indicating that the proposed approach was applicable for the routine analysis and quality control of trollflowers.
Optimization of the Extraction Process of Flavonoids from Trollius Ledebouri with Natural Deep Eutectic Solvents

Abstract:In recent years, natural deep eutectic solvents have been favored greatly due to their environment friendly, mild biological toxicity and simple biodegradability. Natural deep eutectic solvents gradually applied for the extracting bioactive compounds from natural products efficiently. In this study, 20 natural deep eutectic solvents were prepared and their physical and chemical properties were tested. The ultrasonic-assisted extraction method was used to extract flavonoids from Trollius ledebouri and high-performance liquid chromatography-ultraviolet was applied to examine two main bioactive flavonoids (orientin and vitexin). Compared with traditional solvents (water and 60% ethanol solution), natural deep eutectic solvents composed of L(-)-proline and levulinic acid (molar ratio 1:2) show a super extraction efficiency. On this basis, the response surface method was used to optimize the extraction temperature, extraction time, water contents, and solid-liquid ratio. As a consequence, the extraction temperature 60 degrees C, extraction time 18 min, water content 14% (v/v), and the solid-liquid ratio 48mL.g(-1) were chosen as the best extraction process. This study shows that natural deep eutectic solvents can effectively extract flavonoids from T. ledebouri, laying a foundation for the further application of natural deep eutectic solvents to extract bioactive compounds from natural products.

Abstract:The plants from genus Origanum are common folk Chinese herbs used to treat a variety of diseases. They are also used as a spice, a seasoning, and an ornament. Origanum plants are rich in essential oils and also have other compounds including terpenoids, flavonoids, organic acids, and sterols. They have a variety of biological activities such as antispasmodic, anti-inflammatory, growth-promoting, antibacterial, antioxidant, and anticancer properties. The chemical components and biological effects of genus Origanum were summarized by different scientific databases such as Web of Science, SciFinder, Baidu Scholar, PubMed, ScienceDirect, and SpringerLink. In conclusion, recent studies were mainly focused on the activities of their essential oils. The research studies for nonvolatile constituents and their pharmacological activities are few. Therefore, research on compounds in genus Origanum plants can be strengthened and their application prospect can be explored so as to make better use of the resources of these plants.

Abstract:Trolliusditerpenosides A-Q (1–17), seventeen new labdane-diterpenoid glycosides, were isolated from the dried flowers of Trollius chinensis Bunge, a plant that has been commonly used as both an anti-inflammatory folk medicine and a healthcare tea for its therapeutic and anti-viral and antibacterial properties. Their structures were corroborated via comprehensive spectroscopic analysis, ECD calculations, and single-crystal X-ray diffraction analysis. Furthermore, the inhibitory activities on lipopolysaccharide (LPS)-induced NO production in RAW 264.7 macrophages of all compounds (1–17) were evaluated in vitro. Compounds 3, 6, 7, and 11 displayed significant inhibitory activities against NO production, with IC50 values ranging from 1.6 ± 0.1 to 14.4 ± 0.2 μM. In addition, compounds 3, 6, 7, and 11 all down-regulated the mRNA expression of iNOS, COX-2, and IL-1β in RAW 264.7 cells mediated by LPS. These findings not only support the chemical context of genus Trollius but also the exploration of new chemical entities with pharmacological significance from this genus.

Network pharmacology-based study on the mechanism of action of Trollius chinensis capsule in the treatment of upper respiratory tract infection

Abstract:Background: Upper respiratory tract infection (URTI), one of the most common respiratory diseases, has a high annual incidence. Trollius chinensis capsule has been used to treat URTI in China. However, the underlying-mechanisms remain unclear. Methods: Network pharmacology was used to explore the potential mechanism of action of Trollius chinensis capsule in URTI treatment. The active compounds in Trollius chinensis were obtained from the TCMSP, SymMap, and ETCM databases. The TCMSP, PubChem, and SwissTargetPrediction databases were used to predict potential targets of Trollius chinensis. URTI-associated targets were gathered from GeneCards and DisGeNET databases. The key targets and signaling pathways associated with URTI were selected by network topology, GO, and KEGG pathway enrichment analysis. Molecular docking was used to verify the binding activity between active compounds and key targets. Results: Quercetin, pectolinarigenin, beta-sitosterol, acacetin and cirsimaritin are major active compounds in Trollius chinensis capsule. Eighty one candidate therapeutic targets were confirmed to be involved in protection of Trollius chinensis capsule against URTI. Among them, 7 key targets (TP53, IL6, AKT1, CASP3, CXCL8, MMP9, and EGFR) were verified to have good binding affinities to the main active compounds. Furthermore, enrichment analyses suggested that inflammatory response, virus infection and oxidative stress related biological processes and pathways were possibly the potential mechanism. Conclusion: Overall, the present study clarified that quercetin, pectolinarigenin, beta-sitosterol, acacetin and cirsimaritin are proved to be the main effective compounds of Trollius chinensis capsule treating URTI, possibly by acting on the targets of IL6, AKT1, CASP3, CXCL8, MMP9 and EGFR to play anti-infectious, anti-viral, and anti-oxidative effects. This study provides a new understanding of the active compounds and mechanisms of Trollius chinensis capsule in URTI treatment from the perspective of network pharmacology.
On-Line Study of Flavonoids of Trollius Chinensis Bunge Binding to Dna with Ethidium Bromide Using A Novel Combination of Chromatographic, Mass Spectrometric and Fluorescence Techniques

Abstract:The study of the interaction between drugs and DNA is an important way to understand the role of drug molecules. A novel online analytical method for this purpose combining high-performance liquid chromatography-diode array detector-electrospray ionization-ion-trap time-of-flight mass spectrometry (HPLC-DAD-ESI-IT-TOF-MS(n)) and DNA-ethidium bromide detection with a fluorescence detector (DNA-EB-FLD) was firstly developed, which could rapidly identify the chemical constituents and obtain the profile related to DNA binding activity. This method has been applied for a precise or probable identification of the chemical constituents by ultraviolet (UV) absorption and MS(n) data analysis, while the DNA binding profile has been characterized by directly measuring the fluorescence intensity of compound-DNA-EB. Using this method, Trollius chinensis Bunge was studied and 18 constituents were identified by MS(n) data; six of them (4'-methoxy-2″-O-(2‴-methylbutyryl)vitexin,2″-O-(3‴-methoxycaffeoyl)vitexin) and 4'-methoxy-2″-O-(2‴-methylbutyryl)orientin,acacetin-7-O-rutinoside,quercetin-3-O-xylosylglucoside,quercetin-3-O-arabinosylglucoside) were identified for the first time in T. chinensis Bunge, and 16 constituents accounted for its activity of binding to DNA. The established (HPLC-DAD-ESI-IT-TOF-MS(n) DNA-EB-FLD) system has proved to offer a useful strategy for correlating the chemical profile with the binding to DNA activities of the components without their isolation and purification, and may be used for multicomponent analysis of active substances in other herbs.

Abstract:Abstract Objectives Oxytropis DC is a perennial plant of Fabaceae family, which is widely distributed in the northern temperate zone. It is known as “locoweed” because of its toxic component swainsonine. However, it is widely used in Tibetan medicine and Mongolian medicine, mainly for the treatment of heat-clearing and detoxifying, pain-relieving, anti-inflammatory, hemostasis, and other diseases. To provide a basis for the further development and utilization of Oxytropis DC, the pieces of literature about the application, phytochemistry, pharmacological action, and toxicity of Oxytropis DC were reviewed and analyzed. Key findings A total of 373 chemical constituents were found from Oxytropis DC, including flavonoids, alkaloids, steroids, terpenoids, and others. Pharmacological actions mainly include antitumor, antioxidation, anti-inflammatory, analgesic, antibacterial, antifibrosis, and other pharmacological actions, among them, the antitumor effect is particularly prominent. Summary At present, studies on its pharmacological effects are mainly concentrated on the extracts, some flavonoids, and alkaloids. In the follow-up studies, research on the pharmacological activities of the other chemical constituents in Oxytropis should be strengthened. It has the potential to pave the way for research and development of novel Oxytropis medicines.

Abstract:Taxilli Herba (TH) is a well-known traditional Chinese medicine (TCM) with a wide range of clinical application. However, there is a lack of comprehensive research on its chemical composition in recent years. At the same time, Taxillus chinensis (DC) Danser is a semi parasitic plant with abundant hosts, and its chemical constituents varies due to hosts. In this study, the characterization of chemical constituents in TH was analyzed by ultra-fast liquid chromatography coupled with triple quadrupole-time of flight tandem mass spectrometry (UFLC-Triple TOF-MS/MS). Moreover, partial least squares discriminant analysis (PLS-DA) was applied to reveal the differential constituents in TH from different hosts based on the qualitative information of the chemical constituents. Results showed that 73 constituents in TH were identified or tentatively presumed, including flavonoids, phenolic acids and glycosides, and others; meanwhile, the fragmentation pathways of different types of compounds were preliminarily deduced by the fragmentation behavior of the major constituents. In addition, 23 differential characteristic constituents were screened based on variable importance in projection (VIP) and p-value. Among them, quercetin 3-O-β-D-glucuronide, quercitrin and hyperoside were common differential constituents. Our research will contribute to comprehensive evaluation and intrinsic quality control of TH, and provide a scientific basis for the variety identification of medicinal materials from different hosts.

Abstract:Taxus, as a globally prevalent evergreen tree, contains a wealth of bioactive components that play a crucial role in the pharmaceutical field. Taxus extracts, defined as a collection of one or more bioactive compounds extracted from the genus Taxus spp., have become a significant focus of modern cancer treatment research. This review article aims to delve into the scientific background of Taxus extracts and their considerable value in pharmaceutical research. It meticulously sifts through and compares various advanced extraction techniques such as supercritical extraction, ultrasound extraction, microwave-assisted extraction, solid-phase extraction, high-pressure pulsed electric field extraction, and enzymatic extraction, assessing each technology's advantages and limitations across dimensions such as extraction efficiency, extraction purity, economic cost, operational time, and environmental impact, with comprehensive analysis results presented in table form. In the area of drug formulation design, this paper systematically discusses the development strategies for solid, liquid, and semi-solid dosage forms based on the unique physicochemical properties of Taxus extracts, their intended medical uses, and specific release characteristics, delving deeply into the selection of excipients and the critical technical issues in the drug preparation process. Moreover, the article looks forward to the potential directions of Taxus extracts in future research and medical applications, emphasizing the urgency and importance of continuously optimizing extraction methods and formulation design to enhance treatment efficacy, reduce production costs, and decrease environmental burdens. It provides a comprehensive set of preparation techniques and formulation optimization schemes for researchers in cancer treatment and other medical fields, promoting the application and development of Taxus extracts in pharmaceutical sciences.

Abstract:Fritillaria is a traditional Chinese medicine(TCM) with a history of thousands of years. Fritillaria always contain saponins, alkaloids, amino acids, flavones, and polysaccharides. Among them, Fritillaria polysaccharide has a variety of biological activities. Its anti-inflammatory and antiaging activities are new study hotspots. The extraction, purification, quantitative determination, monosaccharide composition, and biological activity of Fritillaria polysaccharides have been examined for several years in an attempt to identify the active components and understand the pharmacological process. In this review, ample original publications related to the distribution, extraction, purification, quantitative determination, monosaccharide composition and biological activities of Fritillaria until 2023 were searched and collected by using various literature databases. Databases included the China National Knowledge Infrastructure, VIP database, Wan Fang database, PubMed, Elsevier, Springer, Science Direct, Google Scholar and Web of Science database, as well as the classic Chinese medical books and PhD and MSc theses. The properties and outcomes of various extractions, purifications, quantitative determination methods, monosaccharide compositions, and biological activities of Fritillaria polysaccharides are discussed here. Additionally, we summarize the research potential of Fritillaria polysaccharide and identify promising research direction candidates.
