PrLZ is more highly expressed in high-grade prostatic intraepithelial neoplasia (PIN) and prostate cancer (PCa) than in normal prostate or benign prostatic hyperplasia (BPH), which indicated its potential role in PCa progression. We previously reported that PrLZ promotes growth capability in vitro as well as tumorigenicity in vivo. In this work, we investigated its potential role in metastasis progression of PCa. METHODS PrLZ-overexpressing vector and PrLZ-specific microRNA-expressing vector were transfected into LNCaP and C4-2 cells respectively. Adhesion capability was determined following PrLZ alteration. Immunofluorescence was used to detect the expression of E-cadherin. Transwell assay was performed to determine the migration and invasion capability. Western blot was used to determine STAT3 phosphorylation and MMP-2 expression. RESULTS PrLZ-overexpressing LNCaP/PrLZ and PrLZ-knockdown C4-2/miPrLZ cells were obtained respectively. O.D. values stand for adherent LNCaP and LNCaP/PrLZ cells were 0.633±0.09 and 0.277±0.07 respectively, which showed a statistic significance between the two cells (P<0.05), vice versa PrLZ knockdown led to enhanced adhesion of C4-2/miPrLZ cells compared to parental C4-2 cells (P<0.01). Immunofluorescence showed that E-cadherin was dominant expressed in cell-cell interface of LNCaP cells, whereas LNCaP/PrLZ cells exhibited a diffused expression in cytoplasm. In addition, PrLZ overexpression enhanced the invasive capability rather than migration of LNCaP cells, however, PrLZ knockdown decreased both invasive and migration ability of C4-2 cells. Mechanically, PrLZ overexpression elevated STAT3 phosphorylation and upregulated MMP-2 expression in LNCaP/PrLZ cells, and inhibition of PrLZ results in inactivation of STAT3 and decreased expression of MMP-2 in C4-2/miPrLZ cells. Furthermore, when LNCaP/PrLZ and C4-2 cells were treated with siRNA targeting STAT3 or Stattic, an inhibitor of STAT3, decreased invasive ability and downregulation of MMP-2 were obtained regardless of the high expression level of PrLZ. CONCLUSIONS PrLZ decreased adhesion ability and enhanced invasive capability of PCa cells via activation of STAT3 and upregulation of MMP-2 expression, which indicated that PrLZ may play a critical role in initiating metastasis of PCa progression.
