Abstract:Leucine zipper sequences have been widely used to engineer protein-based hydrogels for biomedical applications. Previously, we have used this method to engineer tandem modular protein-based hydrogels as a step toward developing extracellular matrix-mimetic hydrogels. However, the spontaneous self-association of leucine zipper sequences in solution has made it challenging to express and purify tandem modular proteins carrying leucine zipper under native conditions. To obviate this problem, here we report a novel two-component approach to engineer tandem modular protein-based hydrogels. This methodology makes use of two complementary leucine zipper sequences (CCE and CCK), which do not self-associate but self-assemble into heterodimeric coiled-coils at neutral pH, as functional groups to drive the self-assembly of protein hydrogels. The two protein components are bifunctional and trifunctional tandem modular proteins carrying the leucine zipper functional groups. We found that the two proteins carrying CCE or CCK can be expressed and purified under native conditions with high yield. Upon mixing, the aqueous solution of the two proteins readily forms a transparent hydrogel. The resultant hydrogel can undergo reversible sol-gel transitions as a function of temperature, and shows much improved erosion properties. This method provides a new approach to tune the topology and physical properties of the protein hydrogels via genetic engineering, and opens the possibility to systematically explore the use of large native extracellular proteins to engineer extracellular matrix-mimetic hydrogels.

Abstract:The integration of functions in materials in order to gain macroscopic effects in response to environmental changes is an ongoing challenge in material science. Here, functions on different hierarchical levels are sequentially linked to translate a pH-triggered conformational transition from the molecular to the macroscopic level to induce directed movements in hydrogels. When the pH is increased, lysine-rich peptide molecules change their conformation into a beta-hairpin structure because of the reduced electrostatic repulsion among the deprotonated amino groups. Coupled to this conformation change is the capability of the beta-hairpin motifs to subsequently assemble into aggregates acting as reversible cross links, which are used as controlling units to fix a temporary macroscopic shape. A structural function implemented into the hydrogel by a microporous architecture-enabled nondisruptive deformation upon compression by buckling of pore walls and their elastic recovery. Coupled to this structural function is the capability of the porous material to enhance the diffusion of ions into the hydrogel and to keep the dimension of the macroscopic systems almost constant when the additional cross-links are formed or cleaved as it limits the dimensional change of the pore walls. Covalent cross-linking of the hydrogel into a polymer network acted as gear shift to ensure translation of the function on the molecular level to the macroscopic dimension. In this way, the information of a directed shape-shift can be programmed into the material by mechanical deformation and pH-dependent formation of temporary net points. The information could be read out by lowering the pH. The peptides reverted back into their original random coil conformation and the porous polymer network could recover from the previously applied elastic deformation. The level of multifunctionality of the hydrogels can be increased by implementation of additional orthogonal functions such as antimicrobicity by proper selection of multifunctional peptides, which could enable sophisticated biomedical devices.

Abstract:Relating the macroscopic properties of protein-based materials to their underlying component microstructure is an outstanding challenge. Here, we exploit computational design to specify the size, flexibility, and valency of de novo protein building blocks, as well as the interaction dynamics between them, to investigate how molecular parameters govern the macroscopic viscoelasticity of the resultant protein hydrogels. We construct gel systems from pairs of symmetric protein homo-oligomers, each comprising 2, 5, 24, or 120 individual protein components, that are crosslinked either physically or covalently into idealized step-growth biopolymer networks. Through rheological assessment, we find that the covalent linkage of multifunctional precursors yields hydrogels whose viscoelasticity depends on the crosslink length between the constituent building blocks. In contrast, reversibly crosslinking the homo-oligomeric components with a computationally designed heterodimer results in viscoelastic biomaterials exhibiting fluid-like properties under rest and low shear, but solid-like behavior at higher frequencies. Exploiting the unique genetic encodability of these materials, we demonstrate the assembly of protein networks within living mammalian cells and show via fluorescence recovery after photobleaching (FRAP) that mechanical properties can be tuned intracellularly in a manner similar to formulations formed extracellularly. We anticipate that the ability to modularly construct and systematically program the viscoelastic properties of designer protein-based materials could have broad utility in biomedicine, with applications in tissue engineering, therapeutic delivery, and synthetic biology.

Abstract:Reversible sol-gel transforming behaviors combined with tunable mechanical properties are vital demands for developing biomaterials. However, it remains challenging to correlate these properties with the hydrogels constructed by denatured protein as building blocks. Herein, taking advantage of naturally high-affinity coordination environments consisting of i, i + 4 His-Glu motifs offered by paramyosin, a ubiquitous nanofibrous protein, we found that Zn2+ rather than Ca2+ or Mg2+ has the ability to trigger the self-assembly of native abalone paramyosin (AbPM) into protein hydrogels under benign conditions, while the addition of EDTA induces the hydrogels back into protein monomers, indicative of a reversible process. By using such sol-gel reversible property, the AbPM gels can serve as a vehicle to encapsulate bioactive molecules such as curcumin, thereby protecting it from degradation from thermal and photo treatment. Notably, based on the high conserved structure of native AbPM, the mechanical property and biological activity of the fabricated AbPM hydrogels can be fined-tuned by its noncovalent interaction with small molecules. All these findings raise the possibility that native paramyosin can be explored as a new class of protein hydrogels which exhibit favorable properties that the traditional hydrogels constructed by denatured protein building blocks do not have.

Abstract:We demonstrate the synthesis of protein-polymer hybrid hydrogel that can be used as a platform for immobilizing functional proteins. Orthogonal chemistry was employed for cross-linking the hybrid network and conjugating proteins to the gel backbone, allowing for the convenient, one-pot formation of a functionalized hydrogel. The resulting hydrogel had tunable mechanical properties, was stable in solution, and biocompatible.

Abstract:Biomaterials that mimic the extracellular matrix in both modularity and crosslinking chemistry have the potential to recapitulate the instructive signals that ultimately control cell fate. Toward this goal, modular protein polymer-based hydrogels were created through genetic engineering and enzymatic crosslinking. Animal derived tissue transglutaminase (tTG) and recombinant human transglutaminase (hTG) enzymes were used for coupling two classes of protein polymers containing either lysine or glutamine, which have the recognition substrates for enzymatic crosslinking evenly spaced along the protein backbone. Utilizing tTG under physiological conditions, complete crosslinking occurred within 2 min, as determined by particle tracking microrheology. Hydrogel composition impacted the elastic storage modulus of the gel over 4-fold and also influenced microstructure and degree of swelling, but did not appreciably effect degradation by plasmin. Mouse 3T3 and primary human fibroblasts were cultured in both 2- and 3-dimensions without a decrease in cell viability and displayed spreading in 2D. The properties, which are controlled through the specific nature of the protein polymer precursors, render these gels valuable for in situ therapies. Furthermore, the modular hydrogel composition allows tailoring of mechanical and physical properties for specific tissue engineering applications. (C) 2010 Elsevier Ltd. All rights reserved.

Abstract:Biological load-bearing tissues are strong, tough, and recoverable under periodic mechanical loads. However, such features have rarely been achieved simultaneously in the same synthetic hydrogels. Here, we use a force-coupled enzymatic reaction to tune a strong covalent peptide linkage to a reversible bond. Based on this concept we engineered double network hydrogels that combine high mechanical strength and reversible mechanical recovery in the same hydrogels. Specifically, we found that a peptide ligase, sortase A, can promote the proteolysis of peptides under force. The peptide bond can be re-ligated by the same enzyme in the absence of force. This allows the sacrificial network in the double-network hydrogels to be ruptured and rebuilt reversibly. Our results demonstrate a general approach for precisely controlling the mechanical and dynamic properties of hydrogels at the molecular level.
An Injectable Self-Healing Protein Hydrogel with Multiple Dissipation Modes and Tunable Dynamic Response.

Abstract:Hydrogels with dynamic mechanical properties are of special interest in the field of tissue engineering and drug delivery. However, it remains challenging to tailor the dynamic mechanical response of hydrogels to simultaneously meet diverse application needs. Here, we report a hetero-coiled-coil complex cross-linked protein hydrogel exhibiting unusual multiple energy dissipation modes and tunable dynamic response. Such unique features confer on the hydrogel responsiveness to mechanical stimuli in a broad range of frequencies. Therefore, the hydrogels are injectable due to their shearing-thinning properties at low shear rates of 0.8 rad s(-1) and can fully recover their mechanical properties within a few seconds due to the intrinsic fast dynamics of the cross-linkers. Moreover, the dynamic response of these hydrogels can be fine-tuned by the temperature and the hydrogel network structures. We anticipate that these hydrogels are promising candidates for delivering therapeutic drugs, biological molecules, and cells in a broad spectrum of biomedical applications.

Abstract:A modular and straightforward strategy to build up an amyloid fibril‐based hydrogel platform is designed by combining a mussel foot protein and a soft amyloid core, displaying their original advanced properties. The resulting hydrogel exhibits excellent mechanical and adhesive performance and is potentially applied to biocatalysis, coating materials, and tissue engineering when endowed with tunable functionalities. Natural proteins display organized hierarchical structures and tailored functionalities that cannot be achieved by synthetic approaches, highlighting the increased interest in developing protein‐based materials. Protein self‐assembly allows fabricating sophisticated supramolecular structures from relatively simple building blocks, a strategy naturally employed by amyloid proteins and intrinsically disordered proteins. However, the design of self‐assembled bioinspired materials with multi functionalities is still challenging. Inspired by the natural self‐assembly proteins (such as mussel foot proteins and amyloid proteins), a temperature‐inducible engineering programable hydrogel‐like amyloid nanostructure is developed by using a genetically modular fusion approach. The resulting hydrogel‐like assemblies display outstanding adhesive capacity, high stability, and broad substrate universality. The employed SpyCatcher/SpyTag system allows modifying the hydrogel‐like assemblies with any functional proteins of interest. Owing to their strong adhesive capacity and functional flexibility, such amyloid fibril‐based hydrogel shows advantages in the immobilization of diverse enzymes for highly efficient biocatalysis, fabrication of multi‐layered functional coatings, and construction of functionalized 3D scaffold for cell culture. Overall, a modular and straightforward approach is established to obtain a genetically programable nanostructure platform. The novel hydrogel‐like assemblies described here may be potentially applied to but not limited to synthetic biology, surface/interface engineering, and tissue engineering.

Rationally Designed Synthetic Protein Hydrogels with Predictable and Controllable Mechanical Properties

Abstract:A key challenge in biomaterials research is to produce synthetic hydrogels that can replicate the diverse mechanical properties of the naturally occurring tissues for various biomedical applications, including tissue engineering, stem cell and cancer research, cell therapy, and immunomodulation. However, currently, the methods that can be used to control the mechanical properties of hydrogels are very limited and are mainly focused only on the elasticity of hydrogels. In this work, combining single molecule force spectroscopy, protein engineering and theoretical modeling, we show that synthetic protein hydrogels with predictable mechanical properties can be rationally designed using protein building blocks with known mechanical properties at the molecular level. We have successfully tuned the elasticity, extensibility, toughness and self-healing of hydrogels using this approach [1]. We have also demonstrated the engineering of physically crosslinked muscle-mimic hydrogels which are strong, tough, elastic and able to function under significant load and spontaneously self-heal when damaged [1]. Next, by using light-responsive proteins as building blocks, we have been able to engineer hydrogels whose mechanical properties are reversibly tunable by light illumination [2]. These hydrogels can capture the dynamic mechanical environments of naturally occurring extracellular matrices and are potentially suitable for cell culture and tissue engineering. Finally, we show that the hydrogel network structure is also important for the mechanical properties of hydrogels. The hydrogels made of the same protein building blocks but with different network structures can have distinct mechanical responses. These studies provide a biomechanical basis for the rational design of synthetic proteins hydrogels with predictable and controllable mechanical properties.

Abstract:Designing synthetic protein hydrogels with tailored mechanical properties similar to naturally occurring tissues is an eternal pursuit in tissue engineering and stem cell and cancer research. However, it remains challenging to correlate the mechanical properties of protein hydrogels with the nanomechanics of individual building blocks. Here we use single-molecule force spectroscopy, protein engineering and theoretical modeling to prove that the mechanical properties of protein hydrogels are predictable based on the mechanical hierarchy of the cross-linkers and the load-bearing modules at the molecular level. These findings provide a framework for rationally designing protein hydrogels with independently tunable elasticity, extensibility, toughness and self-healing. Using this principle, we demonstrate the engineering of self-healable muscle-mimicking hydrogels that can significantly dissipate energy through protein unfolding. We expect that this principle can be generalized for the construction of protein hydrogels with customized mechanical properties for biomedical applications.
Molecular Engineering of Metal Coordination Interactions for Strong, Tough, and Fast-Recovery Hydrogels.

Abstract:Many load-bearing tissues, such as muscles and cartilages, show high elasticity, toughness, and fast recovery. However, combining these mechanical properties in the same synthetic biomaterials is fundamentally challenging. Here, we show that strong, tough, and fast-recovery hydrogels can be engineered using cross-linkers involving cooperative dynamic interactions. We designed a histidine-rich decapeptide containing two tandem zinc binding motifs. Because of allosteric structural change-induced cooperative binding, this decapeptide had a higher thermodynamic stability, stronger binding strength, and faster binding rate than single binding motifs or isolated ligands. The engineered hybrid network hydrogels containing the peptide-zinc complex exhibit a break stress of ~3.0 MPa, toughness of ~4.0 MJ m-3, and fast recovery in seconds. We expect that they can function effectively as scaffolds for load-bearing tissue engineering and as building blocks for soft robotics. Our results provide a general route to tune the mechanical and dynamic properties of hydrogels at the molecular level.

Abstract:Stimuli-responsive shape-transforming hydrogels have shown great potential toward various engineering applications including soft robotics and microfluidics. Despite significant progress in designing hydrogels with ever more sophisticated shape-morphing behaviors, an ultimate goal yet to be fulfilled is programmable reversible shape transformation. It is reported here that transient structural anisotropy can be programmed into copolymer hydrogels of N-isopropylacrylamide and stearyl acrylate. Structural anisotropy arises from the deformed hydrophobic domains of the stearyl groups after thermomechanical programming, which serves as a template for the reversible globule-to-coil transition of the poly(N-isopropylacrylamide) chains. The structural anisotropy is transient and can be erased upon cooling. This allows repeated programming for reversible shape transformation, an unknown feature for the current hydrogels. The programmable reversible transformation is expected to greatly extend the technical scope for hydrogel-based devices.
One-Pot Fabrication of Supramolecular Synthetic Protein Hydrogel with Tissue-like Integrated Dynamic Features

Abstract:Protein-incorporated soft networks have received remarkable attention during the past several years. They possess desirable properties similar to native tissues and organs and exhibit unique advantages in applications. However, fabrication of protein-based hydrogels usually suffers from complex protein mutation and modification or chemical synthesis, which limited the scale and yield of production. Meanwhile, the lack of rationally designed noncovalent interactions in networks may result in a deficiency of the dynamic features of materials. Therefore, a highly efficient method is needed to include supramolecular interactions into protein hydrogel to generate a highly dynamic hydrogel possessing integrated tissue-like properties. Here, we report the design and construction of native protein-based supramolecular synthetic protein hydrogels through a simple and efficient one-pot polymerization of acrylamide and ligand monomers in the presence of a ligand-binding protein. The supramolecular interactions in the network yield integrated dynamic properties, including remarkable stretchability over 10,000% of their original length, ultrafast self-healing abilities within 3-4 s, tissue-like fast stress relaxation, satisfactory ability of adhesion to different living and nonliving substrates, injectability, and high biocompatibility. Furthermore, this material demonstrated potential as a biosensor to monitor small finger movements. This strategy provides a new avenue for fabricating synthetic protein hydrogels with integrated features.

Abstract:Designing proteins that fold and assemble over different length scales provides a way to tailor the mechanical properties and biological performance of hydrogels. In this study, we designed modular proteins that self-assemble into fibrillar networks and, as a result, form hydrogel materials with novel properties. We incorporated distinct functionalities by connecting separate self-assembling (A block) and cell-binding (B block) domains into single macromolecules. The number of self-assembling domains affects the rigidity of the fibers and the final storage modulus G' of the materials. The mechanical properties of the hydrogels could be tuned over a broad range (G' = 0.1 - 10 kPa), making them suitable for the cultivation and differentiation of multiple cell types, including cortical neurons and human mesenchymal stem cells. Moreover, we confirmed the bioavailability of cell attachment domains in the hydrogels that can be further tailored for specific cell types or other biological applications. Finally, we demonstrate the versatility of the designed proteins for application in biofabrication as 3D scaffolds that support cell growth and guide their function. STATEMENT OF SIGNIFICANCE: Designed proteins that enable the decoupling of biophysical and biochemical properties within the final material could enable modular biomaterial engineering. In this context, we present a designed modular protein platform that integrates self-assembling domains (A blocks) and cell-binding domains (B blocks) within a single biopolymer. The linking of assembly domains and cell-binding domains this way provided independent tuning of mechanical properties and inclusion of biofunctional domains. We demonstrate the use of this platform for biofabrication, including neural cell culture and 3D printing of scaffolds for mesenchymal stem cell culture and differentiation. Overall, this work highlights how informed design of biopolymer sequences can enable the modular design of protein-based hydrogels with independently tunable biophysical and biochemical properties.

Abstract:Injectable and self-repairing hydrogels with tunable gelation time have excellent application prospects in drug release and tissue engineering. In this study, we synthesized the above hydrogels through acylhydrazone bonds between the polyacrylate containing the aldehyde group and azobenzene group (P(Azo-co-EGMA)) and the P(N-isopropylacrylamide-co-acrylhydrazide) containing the hydrazide group (P(NIPAM-co-AH)). The gelation time (t sol–gel) could be controlled from a few seconds (<5 s) to several hours (20 h) by changing the total polymer concentration or the molecular weight of the polymers. After a period of hydrogel formation observed by the vial inversion method, the hydrogel reached a stable state, and the stable hydrogel state time (t stable) recorded by diffusing wave spectroscopy (DWS) was much longer than t sol–gel. The elasticity index (EI = 217391) of stabilized hydrogel Gel 7 formed by polymers with higher molecular weight was about five times that of hydrogel Gel 4 formed by polymers with lower molecular weight, and the elastic modulus (G′ = 1766 Pa) of Gel 7 at the same frequency was about four times that of Gel 4. Besides, the gelation behavior could also be adjusted by adding β-cyclodextrins (β-CDs) and UV irradiation. The hydrogels displayed fast self-healing ability without any external stimuli and achieved pH-responsive sol–gel transition. Moreover, the hydrogels exhibited injectable ability and biocompatibility, which made the hydrogels more promising.
