Abstract:Currently,study and development on tea green pigments(TGP)become the hot spots in green tea utilization.In the present paper,components of TGP were introduced.Mechanism of TGP degradation during the processing and storage of green tea were summarized.The processing techniques of TGP and the factors which affect their formation and stability,including metal ion,light,and heat,pH value,were discussed.The development and application prospects of TGP were also analyzed.

Abstract:Tea polyphenols is an ideal natural anti-oxidant,which has being widely applied in many fields.The methods of extraction of tea polyphenols from green tea have been become one of hot points of tea research.In the present paper,methods of extraction of tea polyphonols from green tea were reviewed,including solvent extraction,ionic precipitation,supercritical CO2 extraction,resin adsorption,ultrasonic assistant extraction,microwave assistant extraction and so on.We also discussed basic principle,producing routine and advantage or disadvantage of each method at the same time.

Abstract:Background: Enzymatic reactions during tea growth, processing, storage and post-processing conduce to the diversity in the organoleptic qualities and health benefits of tea. While chemical component transformations by enzymatic catalysis have been extensively explored, the dynamic alteration of endogenous and exogenous enzyme activities throughout manufacture chains remain in a mist.Scope and approach: This paper discusses main enzymatic reactions involved in tea growth, processing, storage and post-processing, including the dominant enzymes of specific processes, the chemical transformation driven by enzymes and the alteration of endogenous and exogenous enzymes activities. The direct and indirect regulatory factors influencing enzyme activities and their possible applications in tea production practice are also explored. Key findings and conclusion: The endogenous enzyme system of tea plants regulates secondary metabolism and stress defense, contributing to the primary accumulation of volatile and non-volatile compounds in tea plants. Endogenous enzymes activities altered unceasingly and are exposed to more substrates due to cell rupture during the withering, spread, rolling and tumbling stages, establishing the intricate substance basis for distinct organoleptic characteristics. Microbial or intentionally added exogenous enzymes, such as beta-glucosidase and tannase, dominate during fermentation, storage, and post-processing, transforming chemical profiles to a more radical extent. This review provides theoretical foundation for precise controlling and adequate utilization of enzymes throughout the tea manufacture chains.

Abstract:Background: Tea seed (TS), as the mature fruit and growth source of tea plant, has been characterized with appealing bioactive substances, remarkable health benefits and diverse application prospects. As one of the most significant products of TS, tea seed oil (TSO) has gained recognition as the "oriental olive oil". Tea seed meal (TSM) and tea seed shell (TSS) continue to be underutilized and are usually considered as low-value by-products. Nevertheless, current exploration of the underlying mechanisms is inadequate, and investigations concerning deep processing and potential prospects still remain at their infancy. Consequently, there exists a gap in the comprehensive aspects of TS utilization and the related advancements.Scope and approach: In this review, we have summarized the latest research progresses on TS, specifically focused on the bioactive substances, pharmacological activities, manufacturing process and potential applications. Furthermore, we have also elucidated the current challenges in existing studies, and proposed the research orientations and potential prospects of TS.Key findings and conclusions: TS contain abundant bioactive substances, and exhibit outstanding performance in antioxidant, anti-tumor, anti-obesity, anti-microbe, gastric protection, and so on. These properties contribute to its application potentials as crucial materials in industry, agriculture and ecology, as well as new perspective in drug development and clinical translation. Future researches should concentrate on the specific active in-gredients, intricate structure-activity relationships, bioactive mechanisms, undiscovered health benefits and manufacturing process, which would facilitate the transformation of waste resources into valuable treasures, thus fostering sustainable development and socio-economic growth.

Abstract:Microbiomes can greatly affect the quality of fermented food and beverages, including tea. In this study, microbial populations were characterized during black and green tea manufacturing, revealing that tea processing steps can drive both the bacterial and fungal community structure. Tea leaves were found to mostly harbor Proteobacteria, Bacteriodetes, Firmicutes, and Actinobacteria among bacteria and Ascomycetes among fungi. During processing, tea microbial populations changed especially between sterilized and unsterilized samples. The surface sterilization of fresh leaves before processing can remove many microbes, especially the bacteria of the genera Sphingomonas and Methylobacteria, indicating that these are mostly phylloplane microbes on tea leaves. The surface sterilization removed most fungi, except the Debaryomyces. We also observed a fluctuation in the content of several tea quality-related metabolites during processing. Caffeine and theanine were found in the same quantities in green tea with or without leaf surface sterilization. Abstract:Black tea is highly favored by consumers worldwide, with enzymatic reactions being recognized as a pivotal factor influencing tea quality. The role of microorganisms in shaping the composition of black tea has emerged as a focus of research due to their involvement in enzyme catalysis and metabolic processes. In this study, full-length amplicon sequencing combined with qPCR more accurately reflected microbial profile, and Pantoea, Pseudomonas, Paucibacter, and Cladosporium were identified as the main microbial genera. Moreover, by comprehensively analyzing color, aroma, and taste components over time in black tea samples, correlations were established between the dominant genus and various quality factors. Notably, peroxidase activity levels, total soluble sugar content, and tea pigments concentration exhibited significant associations with the dominant genus. Consequently, this microbiological perspective facilitated the exploration of driving factors for improving black tea quality while establishing a theoretical foundation for quality control in industrial production.

Abstract:Tea (Camellia sinensis L.) is a crop with both commercial and medicinal value with remarkably high polyphenol content in the form of catechins. To understand the molecular regulation of catechin biosynthesis in tea, we treated the tea plants with darkness. We used qRT-PCR to validate the expression of genes involved in catechin biosynthesis. It indicated that dark treatment displayed different effects on the genes participating in tea flavonoid (FL) pathway. The early genes of FL biosynthesis pathway, CHSI, F3H and DFR, remained at steady expression levels when treated by darkness. It is noteworthy that the expression level of LAR increased and the level of ANS decreased under dark conditions. The vanillin assay showed that the dark-treated plants contained lower levels of total catechins than those grown under normal conditions. The HPLC analysis further demonstrated the changes in biosynthesis of catechins under these conditions. In accordance with the gene expression pattern, the content of epicatechins (ECs) declined and that of catechins (Cs) was elevated in response to the darkness. Our study uncovered the molecular mechanisms and biochemical changes of shading in tea cultivation.

Abstract:The fermentation is the main process to form the unique flavor and health benefits of dark tea. Numerous studies have indicated that the microorganisms play a significant part in the fermentation process of dark tea. Dark tea has the quality of "The unique flavor grows over time," but unscientific storage of dark tea might cause infestation of harmful microorganisms, thereby resulting in the remaining of fungi toxins. Mycotoxins are regarded as the main contributor to the quality of dark tea, and its potential mycotoxin risk has attracted people's attention. This study reviews common and potential mycotoxins in dark tea and discusses the possible types of masked mycotoxins in dark tea. A summary of the potential risks of mycotoxins and masked mycotoxins in dark tea is presented, intending to provide a reference for the prevention and risk assessment of harmful fungi in dark tea.

Microbial-Transferred Metabolites and Improvement of Biological Activities of Green Tea Catechins by Human Gut Microbiota

Abstract:Green tea catechins (GTCs) are dietary polyphenols with broad bioactivities that undergo extensive microbial metabolism in the human gut. However, microbial-transferred metabolites and their health benefits are not fully understood. Herein, the microbial metabolism of GTCs by human fecal microbiota and dynamic alteration of the microbiota were integrally investigated via in vitro anaerobic fermentation. The results showed that the human gut microbiota exhibited a strong metabolic effect on GTCs via UHPLC-MS/MS analysis. A total of 35 microbial-transferred metabolites were identified, far more than were identified in previous studies. Among them, five metabolites, namely EGCG quinone, EGC quinone, ECG quinone, EC quinone, and mono-oxygenated EGCG, were identified for the first time in fermented GTCs with the human gut microbiota. Consequently, corresponding metabolic pathways were proposed. Notably, the antioxidant, α-amylase, and α-glucosidase inhibitory activities of the GTCs sample increased after fermentation compared to those of the initial unfermented sample. The results of the 16S rRNA gene sequence analysis showed that the GTCs significantly altered gut microbial diversity and enriched the abundancy of Eubacterium, Flavonifractor, etc., which may be further involved in the metabolisms of GTCs. Thus, these findings contribute to a better understanding of the interactions between GTCs and gut microbiota, as well as the health benefits of green tea consumption.

Abstract:Endophytic fungi and bacteria are the most ubiquitous and representative commensal members that have been studied so far in various higher plants. Within colonization and interaction with their host plants, endophytic microbiota are reportedly to modulate not only the host’s growth but also holobiont resilience to abiotic and biotic stresses, providing a natural reservoir and a promising solution for sustainable agricultural development challenged by global climate change. Moreover, possessing the talent to produce a wide array of high-value natural products, plant endophytic microbiota also serve as an alternative way for novel drug discovery. In this review, tea, one of the world’s three largest nonalcoholic beverages and a worldwide economic woody crop, was highlighted in the context of endophytic microbiota. We explore the recent studies regarding isolation approaches, distribution characteristics and diversity, and also biological functions of endophytic microbiota in Camellia sinensis (L.) O. Kuntze. Profoundly, the future insight into interaction mechanism between endophytic microbiota and tea plants will shed light on in-depth exploration of tea microbial resources.
Metabolic improvements of novel microbial fermentation on black tea by Eurotium cristatum

Abstract:Due to its traditional fermentation, there are obvious limits on the quality improvements in black tea. However, microbial fermentation can provide an abundance of metabolites and improve the flavor of tea. The "golden flower" fungi are widely used in the microbial fermentation of tea and has unique uses in healthcare. To further explore the improvements in black tea quality achieved via microbial fermentation, we used widely targeted metabolomics and metagenomics analyses to investigate the changes in and effects of metabolites and other microorganisms during the interaction between the "golden flower" fungi and black tea. Five key flavor metabolites were detected, the levels of catechin, epigallocatechin gallate, (−)-epicatechin gallate were decreased by different degrees after the inoculation of the "golden flower" fungus, whereas the levels of caffeine and (+)-gallocatechin increased. Botryosphaeriaceae, Botryosphaeriales, Dothideomycetes, Aspergillaceae, Trichocomaceae, and Lecanoromycetes play a positive role in the black tea fermentation process after inoculation with the "golden flower" fungi. D-Ribose can prevent hypoxia-induced apoptosis in cardiac cells, and it shows a strong correlation with Botryosphaeriaceae and Botryosphaeriales. The interaction between microorganisms and metabolites is manifested in tryptophan metabolism, starch and sucrose metabolism, and amino sugar and nucleotide sugar metabolism. In conclusion, the changes in metabolites observed during the fermentation of black tea by "golden flower" fungi are beneficial to human health. This conclusion extends the knowledge of the interaction between the "golden flower" fungi and black tea, and it provides important information for improving the quality of black tea.

Abstract:Qingzhuan tea (QZT) is a unique type of dark tea exclusively produced in Hubei Province of China. In the current study, liquid chromatography-mass spectrometry (LC-MS) coupled with multivariate analysis was applied to characterize the chemical composition of QZT and investigate the effect of QZT processing on its metabolic profile and sensory quality. The contents of polyphenols and flavonoids decreased significantly while the polysaccharides content remained stable, while the theabrownin content inversely increased during QZT processing. LC-MS-based metabolomics analyses revealed that the tea sample after microbial fermentation (MFT) was dramatically different from the sample before microbial fermentation (UFT), while MFT was very similar to QZT. A total of 102 compounds were identified as critical metabolites responsible for metabolic changes caused by QZT processing, with the contents of catechins and flavonoids significantly decreased, and some novel phenolic acids and catechin derivatives were formed. The sensory quality of QZT was mainly formed during microbial fermentation, which greatly reduced the astringency and bitterness of raw tea leaves and produced its characteristic woody and stale aroma as well as mellow taste. These results suggested that microbial fermentation is the critical process in changing the metabolic profile of raw tea leaves and forming the sensory quality of QZT.

Abstract:Tea is one of the most popular beverages worldwide, with several health benefits attributed for its rich chemical composition and further associated with fermentation process to improve its quality attributes. Most tea types originate from the leaves of Camellia sinensis with differences in fermentation levels yielding black tea, green tea, pouchong tea, oolong tea. Teas like pu-erh or kombucha to encompass both green and red types are further post-fermented. Tea fermentation is a traditional process involving physical, biochemical, and microbial changes which are associated with improved organoleptic characters, nutritive value, and health outcomes. The production of fermented tea relies on naturally occurring enzymes and microbial metabolic activities. This review focuses on presenting a holistic overview on the effect of different microorganisms including bacteria, yeast, and fungi on the biochemical changes and sensory attributes of fermented tea products reported in research articles along the last 15 years. Moreover, production conditions and major biochemical changes are dissected to present the best factors influencing fermented tea quality. This review presents an evidence-based reference for specialists in tea industry to optimize tea fermentation process for targeted attributes.

Abstract:Processing of dark tea varieties, such as Fu brick tea, Liupao tea, Qianliang tea, and Qing brick tea, includes solid-state fermentation involving microorganisms. In this study, we analyzed the major chemical constituents of dark tea extracts and evaluated their modulatory effect on the gastrointestinal function in normal mice, including the improvement of gastrointestinal transit and intestinal microbial, as well as the attenuation of intestinal microbial dysbiosis and intestinal pathological damage, and the adjustment of immune function in antibiotic-treated mice. Substantial differences in major chemical constituents, including total polyphenols, total organic acids, water extract content, 18 free amino acids, gallic acid, and six tea catechins, were observed among Fu brick tea, Qianliang tea, Qing brick tea, and Liupao tea extracts. Extracts from the four dark tea varieties significantly promoted gastrointestinal transit and colonization of beneficial Bifidobacterium and Lactobacillus, and inhibited the growth of harmful Escherichia coli and Enterococcus in normal mice. In addition, Qianliang tea, Qing brick tea, and Liupao tea extracts significantly accelerated the reversal of the ampicillin sodium-induced pathological damage in the ileum, intestinal bacterial dysbiosis (Bifidobacterium, Lactobacillus, E. coli, and Enterococcus), and low immunity.

Abstract:Tea is one of the most popular drinks in the world. Dark tea is a kind of post-fermented tea with unique sensory characteristics that is produced by the special fermentation of microorganisms. It contains many bioactive substances, such as tea polyphenols, theabrownin, tea polysaccharides, etc., which have been reported to be beneficial to human health. This paper reviewed the latest research on dark tea's potential in preventing and managing cancer, and the mechanisms mainly involved anti-oxidation, anti-inflammation, inhibiting cancer cell proliferation, inducing cancer cell apoptosis, inhibiting tumor metastasis, and regulating intestinal flora. The purpose of this review is to accumulate evidence on the anti-cancer effects of dark tea, the corresponding mechanisms and limitations of dark tea for cancer prevention and management, the future prospects, and demanding questions about dark tea's possible contributions as an anti-cancer adjuvant.

Abstract:As far as health benefit is concerned, dark tea is one of the best beverages in the world. Theabrownins are the major ingredient contributing to the health benefits of dark tea and known as "the soft gold in dark tea." A growing body of evidence indicated that theabrownins are macromolecular pigments with reddish-brown color and mellow taste, and mainly derived from the oxidative polymerization of tea polyphenols. Theabrownins are the main active ingredients in dark tea which brings multiple health-promoting effects in modulating lipid metabolism, reducing body weight gain, attenuating diabetes, mitigating NAFLD, scavenging ROS, and preventing tumors. More importantly, it's their substantial generation in microbial fermentation that endows dark tea with much stronger hypolipidemic effect compared with other types of tea. This review firstly summarizes the most recent findings on the preparation, structural characteristics, and health-promoting effects of theabrownins, emphasizing the underlying molecular mechanism, especially the different mechanisms behind the effect of theabrownins-mediated gut microbiota on the host's multiple health-promoting benefits. Furthermore, this review points out the main limitations of current research and potential future research directions, hoping to provide updated scientific evidence for their better theoretical research and industrial utilization.

Abstract:This study investigated the changes of main quality components and the effect of bioactivities of black tea during a manufacturing process that adds to enzymatic treatment. It used black tea harvests in summer and autumn for manufacturing and analyzed the number of main quality components and bioactivities of the black tea produced by treating cellulase during rolling and polyphenol oxidase (PPO) and peroxidase (POD) during fermentation. The results found that the contents of total catechins (TC), total polyphenols (TP), reducing sugars (ReS), and individual catechins in the tea infusions of summer and autumn black tea that were obtained during enzymatic treatments decreased significantly ( p < 0.05). However, the content of free amino acid (FAA), the tea quality parameter such as the total liquor color (TLC), the briskness index (BI), and the tea pigments including total theaflavin (TF), thearubigens (TR), TRSI, and TRSII all increased significantly ( p < 0.05). Moreover, the biological activity enhanced black tea's antioxidant and metabolic capacity with enzymatic treatments were proved by in vitro experiments. The results of this study contribute to gaining further knowledge on how black tea's quality and potential health benefits are maximized by using exogenous enzyme addition methods in the manufacturing process preparation.

Abstract:Background: Tea (Camellia sinensis) has a rich history and is widely consumed across many countries, and is categorized into green tea, white tea, oolong tea, yellow tea, black tea, and dark tea based on the level of fermentation. Based on a review of previous literature, the commonly recognized bioactive substances in tea include tea polyphenols, amino acids, polysaccharides, alkaloids, terpenoids, macro minerals, trace elements, and vitamins, which have been known to have various potential health benefits, such as anticancer, antioxidant, anti-inflammatory, anti-diabetes, and anti-obesity properties, cardiovascular protection, immune regulation, and control of the intestinal microbiota. Most studies have only pointed out the characteristics of tea's bioactivities, so a comprehensive summary of the pharmacological characteristics and mechanisms of tea's bioactivities and their use risks are vital. Aim of review: This paper aims to summarize tea's bioactive substances of tea and their pharmacological characteristics and mechanisms, providing a scientific basis for the application of bioactive substances in tea and outlining future research directions for the study of bioactive substances in tea. Key scientific concepts of review: This review summarizes the main biologically active substances, pharmacological effects, and mechanisms and discusses the potential risks. It may help researchers grasp more comprehensive progress in the study of tea bioactive substances to further promote the application of tea as a natural bioactive substance in the medical field.
Effect of Microbial Fermentation on Internal Quality of Oolong Tea

Abstract:Microbial fermentation of oolong tea was studied in this paper.Obviously changes of the internal quality of oolong tea were found in microbial fermentation by Penicillium, Aspergillius niger and Rhizopus.The contents of water extracted substances, tea polyphenols, catechins and soluble sugars decreased and the tea polyphenol content of the fermented oolong tea decreased by 45.3% with a combined use of the above-mentioned 3 kinds of microbes.Meanwhile both the contents of tea pigments and amino acid increased in the Aspergillius niger-mediated fermentation, and especially, the theabrownins (TB)increased by 34.8%.
