Iron-overloading Promotes Protein Nitration in Mouse Liver

Abstract:Protein nitration is a post-translational modification which will induce the changes of protein catalytic activities,cell signaling and cell skeletal structure.Under iron-overloaded condition,there are advantageous features to promote protein nitration.However,up to date,direct evidence on the role of iron-overloaded elevating hepatic protein nitration is little.In this article,iron-overloaded mice were obtained by peritoneal injection with iron-dextran.Western blotting indicated that iron-overloading could increase the expression of inducible nitric oxide as well as the extent of protein tyrosine nitration in liver,and some new protein nitration bands were found(about 57 kD and 35 kD),indicating that iron overloading caused the selectivity of protein nitration in vivo.
