Callus Induction and Establishment of the Plant Regeneration System for Cortaderia Selloana

Abstract:Mature seeds of Cortaderia selloana were used as explants and cultured on MS medium supplemented with different combinations of phytohormones.The optimal tissue culture system for callus induction and plantlet regeneration for this species was identified.The seeds were soaked in sterilized water for 24 h before sterilization with 75% alcoho1 for 20 s and with 0.1%(w/v) hydrargyrum chloride for 10 min.The best medium for callus induction was MS+2,4-D 3.0 mg/L+6-BA 0.1 mg/L.The best medium for callus cytodifferentiation was MS+6-BA 2.0 mg/L+NAA 0.1 mg/L,and the best medium for plant upgrowth was MS+6-BA 1.5 mg/L+NAA 0.2 mg/L.The best rooting medium was 1/2MS+NAA 0.8 mg/L+6-BA 0.2 mg/L+(0.02%)AC.
