HLA gene polymorphisms and long-term humoral response in children immunized with hepatitis B vaccine at birth

Abstract:Objective To study the association between human leukocyte antigen(HLA)class II gene polymorphisms and the long-term humoral response in children vaccinated with hepatitis B vaccine at birth.Methods A total of 297 children between 5 to 7 years old who were immunized with hepatitis B vaccine at birth with the 0,1-,and 6-month schedule were enrolled.Of them,86 children with anti-HBs<10 mIU/ml were considered as long-term non-and hypo-responders;the remaining 211 subjects with anti-HBs≥10 mIU/ml were defined as long-term responders.A total of 10 alleles in HLA-DR and-DQ subregions were detected with polymerase chain reaction(PCR)using sequence-specific primers.Results The frequencies of HLA-DRB1*04(27.9%)and DRB1*07(36.0%)in long-term non-and hypo-responders were higher than that in responders(18.5% and 27.0%,respectively)but with no statistical significance(P=0.072 and 0.122,respectively).The frequency of HLA-DQB1*0401 in long-term non-and hypo-responders was significantly higher than that in responders(15.1% vs 7.6%,P=0.047).The allele distributions of five other alleles in the DR subregion and two other alleles in the DQ subregion between long-term non-and hypo-responders and responders were similar.Conclusions HLA-DQB1*0401 is associated with the long-term humoral response in children vaccinated against hepatitis B during infancy.
