Abstract:The aim of the present study was to investigate the change in heme oxygenase (HO)-carbon monoxide (CO)-cyclic guanosine monophosphate (cGMP) pathway in vascular calcification. Vascular calcification model was established in rats by using vitamin D(3) and nicotine. Vascular calcium content, alkaline phosphatase (ALP) activity, HO activity, HbCO formation and content of cGMP in vessels were measured. Immunochemistry (IH) for HO 1 expression and in situ hybridization (ISH) for HO 1 mRNA were observed. Compared to those of control rats, the aortic calcium content and vascular ALP activity in rats of the calcified group (VDN group) were obviously increased, but HO 1 activity, CO concentration and cGMP content in vessels of rats in VDN group were markedly decreased. Expressions of HO-1 protein and mRNA were significantly decreased compared to control rats. Vascular calcification might induce a down regulation in vascular HO-CO-cGMP pathway.

Abstract:Aims: To explore the effects of heme oxygenase-1 (HO-1) on vascular dysfunction in high fat diet streptozotocin-induced type 2 diabetic (T2D) rats. Methods: Rats received a high-fat diet followed by a low dose of streptozotocin (30 mg/kg) to induce T2D. T2D rats were treated with hemin (1, 5, or 25 mg/kg) or carbon monoxide-releasing molecule-2 (CORM-2, 5 mg/kg) for 4 weeks. Isometric contractions of aortic rings were measured. The expression of cyclooxygenase-2 (COX-2) and activities of HO, SOD, and MDA were evaluated. Results: The fasting blood glucose, blood insulin levels, and IR index in T2D rats were higher than those in the control group, which were ameliorated by HO-1 inducer hemin. The antidiabetic effect was accompanied by enhanced HO activity. The vascular relaxation response to ACh was decreased in T2D rats, while treatment with hemin could prevent such decrease in vasorelaxation. An increase in COX-2 expression was found in the aortas of T2D rats. Treatment of T2D rats with COX-2 inhibitor NS398 restored ACh-induced vasodilation. COX-2 overexpression in T2D rats was inhibited by hemin. Hemin treatment also inhibited the decline of SOD activity and the increase of MDA content in the aorta of T2D rats. CORM-2, an agent which releases the HO-1 product CO, could mimic the beneficial effect of hemin. Conclusion: Induction of HO-1 with hemin ameliorates the abnormality of endothelium-dependent vascular relaxation in T2D rats. Abstract:AIM:To examine the effect of HO-1 inducer hemin on hydrogen peroxide (H2O2) caused decrease in contraction of isolated rat aortic rings, and to elucidate the underlying mechanism.METHODS:The thoracic aortic rings with endothelium of male Sprague-Dawley rats were mounted on a bath system. Isometric contractions of aortic rings were measured.RESULTS:(1) After intraperitoneal injection of HO-1 inducer hemin, HO-1 activity of thoracic aorta and COHb concentration in rat blood enhanced. And it also prevented the decrease in contraction responses to PE which pretreatment of arteries with 300 micromol/L H2O2. (2) Pretreatment of ATP-sensitive potassium channel inhibitor glibenclamide, but not GC inhibitor methylene blue, could partly abolish the protection of hemin in arteries with H2O2 exposure. (3) Hemin could not influence the shift of concentration-response curve to [Ca2+]o in arteries with H2O2 exposure. (4) In Ca(2+) -free K-H solution, exposure of H2O2 reduced caffeine and PE-induced constriction in the rat aortic rings. After pretreatment of hemin, could prevent the decrease in contraction responses to caffeine and PE.CONCLUSION:Increase in HO-1 activity could prevent the H2O2 induced decrease in contraction responses to PE in intact aortic rings. The mechanism might be involved in activation of ATP-sensitive potassium channel and mobilization of intracellular calcium stores, but had no relationship with the GC pathway.

Abstract:Aim To explore the effects of homocysteine(Hcy) on the expression of matrix metalloproteinase-2(MMP-2) in rat vascular smooth muscle cells(VSMC). Methods Cultured rat VSMC was incubated with different concentration of Hcy in vitro for 24,48 and 72 h.The expression of MMP-2 was determined by using the methods of gelatin zymography and western blotting. Results 0.05～1.0 mmol/L Hcy increased the expression of MMP-2 significantly.Incubated with the same concentration of Hcy the level of MMP-2 of 72 h was higher than that of 24 h and 48 h. Hcy >5.0 mmol/L reduced the expression of MMP-2.Incubated with the same concentration of Hcy(>5.0 mmol/L) the level of MMP-2 of 72 h was lower than that of 24 h and 48 h. Conclusions These datas suggested that Hcy can affect the expression of MMP-2 in VSMC.It may be one of factors in the pathogenesis of atherosclerosis induced by Hcy.
Alteration of Enzyme Expressions in Mevalonate Pathway

Abstract:The mevalonate pathway is an important metabolic pathway that plays a key role in multiple cellular processes. The aim of this study was to define whether the enzyme expression in mevalonate pathway changes during cardiovascular remodelling in spontaneously hypertensive rats (SHR).Hearts and thoracic aortas were removed for the study of cardiovascular remodeling in SHR and Wistar-Kyoto rats (WKY). The protein expression of the enzymes in hearts, aortas and livers was analyzed by western blot. The histological measurements showed that the mass and the size of cardiomyocytes, the media thickness and the media cross-sectional area (MCSA) of the thoracic aorta were all increased in SHR since 3 weeks of age. In the heart, there was overexpression of some enzymes, including 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMGR), farnesyl diphosphate synthase (FDPS), and geranylgeranyltransferase type I (GGTase-I), and downregulation of squalene synthetase (SQS) in SHR since 3 weeks of age. In the aorta, besides similar expressions of HMGR, SQS, FDPS and GGTase-I as in the heart, there was upregulation of farnesyltransferase α at 16 and 25 weeks of age and of farnesyltransferase β in 25-weeks-old SHR. Western blot demonstrated overexpression of HMGR and downregulation of SQS in SHR livers at all ages tested.The cardiovascular remodeling of SHR preceded the development of hypertension, and altered expression of several key enzymes in the mevalonate pathway may play a potential pathophysiological role in cardiovascular remodeling.
The Direct Effect of Carbon Monoxide on K-Ca Channels in Vascular Smooth Muscle Cells

Abstract:The vasorelaxation induced by carbon monoxide (CO) has been demonstrated previously, Both a guanosine cyclic monophosphate (cGMP) signalling pathway and cGMP-independent mechanisms have been proposed to be responsible for the vascular action of CO. A direct effect of CO on the activity of calcium-activated K (K-Ca) channels in vascular smooth muscle cells (SMCs) and the underlying mechanisms were investigated in the present study. It was found that CO hyperpolarized single SMCs isolated from rat tail arteries. The whole-cell outward K+ channel currents in vascular SMCs, but not in neuroblastoma cells, were enhanced by CO. Extracellularly or intracellularly applied CO increased the open probability of single high-conductance K-Ca channels concentration-dependently without affecting the single channel conductance. Although it did not increase the resting level of intracellular free calcium concentration, CO significantly enhanced the calcium sensitivity of single K-Ca channels in SMCs. Furthermore, the effect of CO on K-Ca channels was not mediated by cGMP or guanine nucleotide-binding proteins (G proteins, G(i)/G(o) or G(s)) in excised membrane patches. Our results suggest that the direct modulation of high-conductance K-Ca channels in vascular SMCs by CO may constitute a novel mechanism for the vascular effect of CO.

Abstract:Hypoxic pulmonary vascular structural remodeling (HPVSR) is the important pathologic basis of hypoxic pulmonary hypertension (HPH). The discoveries of endogenous gaseous messenger molecules, nitric oxide (NO) and carbon monoxide (CO), have been moving the research of HPVSR to a very new phase. But the effect and significance of heme oxygenase (HO)/CO on the development of HPVSR have not been fully understood. In this study, we observed the alteration of endogenous HO/CO system in five time points during 14 days and found that the content of CO in lung homogenates in rats with HPVSR increased in a time-dependent double-peak manner. Exogenous supply of ZnPP-IX, an inhibitor of HO-1, decreased the content of CO in lung homogenate, decreased the expression of Fas and apoptotic cells in pulmonary artery smooth muscle cells (PASMCs), up-regulated the expression of PCNA in PASMCs, and worsened HPH and HPVSR of hypoxic rats. Meanwhile, exogenous supply of CO played an adverse action. The results showed that the up-regulation of HO/CO exerted a protective role in the development of HPVSR.

Abstract:objective To observe the effect of exogenous H_2S on vascular calcification in rat.Methods vascular calcification in rat was induced by administration of Vitamin D_3 plus nicotine.To Estimate the extent of calcification by Von Kossa staining,calcium content,45Ca2+ deposition and alkaline phosphatase activity.Osteopontin (OPN) mRNA was determined by competitive quantitative RT-PCR.Results A strong positive staining of black/brown areas among the elastic fibers of the medial layer in calcified aorta by Von Kossa staining,Calcium content,45Ca2+ accumulation and ALP activity in calcified arteries increased by 6.77-,1.42-,and 1.87-fold as compared with the controls;The expression of OPN mRNA is up-regulated(P<0.01).However,calcium content,45Ca2+uptake and ALP activity in VDN plus NaHS group were lower than those in VDN group.Low and high dosage NaHS obviously relieve degree of calcification in the cardiovascular tissues in a dosage-dependent manner(P<0.01).Conclusion H_2S plays a role in the pathogenesis of vascular calcification by reducing vascular calcification.
Influence Of Carbon Monoxide On Growth And Apoptosis Of Human Umbilical Artery Smooth Muscle Cells And Vein Endothelial Cells

Abstract:Carbon monoxide (CO) is a vasoactive molecule that is generated by vascular cells as a by-product of heme catabolism and it plays an important physiological role in circulation system. In order to investigate whether exogenous CO can mediate the growth and proliferation of vascular cells, in this study, we used 250 parts per million (ppm) of CO to treat human umbilical artery smooth muscle cell (hUASMC) and human umbilical vein endothelial cell (HuVEC) and further evaluated the growth and apoptosis status of SMC and HuVEC. After SMC and HuVEC were exposed to CO for 7-day, the growth of SMC and HuVEC was significantly inhibited by CO in vitro on day 5 of CO exposure. And CO blocked cell cycle progress of SMC and HuVEC, more SMC and HuVEC stagnated at G0/G1 phase by flow cytometric analysis. Moreover, CO treatment inhibited SMC and HuVEC apoptosis caused by hydrogen peroxide through decreasing caspase 3 and 9 activities. To confirm the molecular mechanism of CO effect on SMC and HuVEC growth, we compared the gene expression profile in SMC and CO-treated SMC, HuVEC and CO-treated HuVEC. By microarray analysis, we found the expression level of some genes which are related to cell cycle regulation, cell growth and proliferation, and apoptosis were changed during CO exposure. We further identified that the down-regulated CDK2 contributed to arresting cell growth and the down-regulated Caspase 3 (CASP3) and Caspase 9 (CASP9) were associated with the inhibition of cell apoptosis. Therefore, CO exerts a certain growth arrest on SMC and HuVEC by inhibiting cell cycle transition from G0/G1 phase to S phase and has regulatory effect on cell apoptosis by regulating the expression of apoptosis-associated genes.

Abstract:AIM: To study alterations of nitric oxide synthase (NOS) in cardiac sarcop lasmic reticulum from rats with myocardial calcification, and to explore the mec hanism of inhibition of SR function in the rats with myocardial calcification. METHODS: The myocardial calcification rat models were prepared by vit amin D3 plus nicotine for 2 weeks and 6 weeks. Cardiac SR was separated by centrifugating. T he nitric oxide (NO) production, NOS activity and NOS protein expression in the SR were perfor med. RESULTS: Compared with control, myocardial calcium content in t he 6 weeks i ncreased by 408%(P0 01), the NO production, NOS activity and NOS pro tein expression in the SR with myocardial calcification increased versus control( P 0 01).Myocardial calcium content was not alterations significantly, but the NO S /NO pathway in the SR was up-regulated slightly in the 2 weeks. CONCLUSION : The up-regulated NOS/NO system in the SR with myocardial calcification is the impor tant mechanism of function inhibition of the SR.
Down- Regulation of Glutamyl Cysteine Cystathionine- Lyase/hydrogen Sulfide System in Rat Cardiovascular Calcification

Abstract:AIM: To observe the changes of cystathionine-lyase/hydrogen sulfide(CSE/H_2S) in vivo in vascular calcification and to explore the role of CSE/H_2S in vascular calcification.METHODS: Vascular calcification model in rats was induced by administration of vitamin D_3 plus nicotine.The extent of calcification was estimated by assaying calcium content.[~(45)Ca~(2+)] deposition and alkaline phosphatase(ALP) activity were detected.CSE mRNA amount was determined by using competitive quantitative RT-PCR.The content of H_2S and activity of CSE in the plasma and cardiovascular tissues were also determined with biochemical methods.RESULTS: Calcium content in myocardium increased by 3.8 folds in a calcification model.Compared to control,calcium content,[~(45)Ca~(2+)] accumulation and ALP activity in calcified arteries increased by 6.8,1.4,and 1.9 folds,respectively(P0.01).H_2S contents in plasma,myocardium and aorta were 39%,39% and 31% lower than those in control group(P0.01).The gene expression of CSE was down-regulated in myocardium and aorta.Compared to control group,the amount of CSE mRNA in myocardium and calcified aorta were decreased by 28% and 36%,respectively(P0.01).The activity of CSE was 56% and 53% lower than that in control(P0.01).CONCLUSION: The production of H_2S,the gene expression and activity of CSE are down-regulated in the cardiovascular calcification,suggesting that the decrease in H_2S production plays a role in the pathogenesis of vascular calcification.
Hydrogen Sulfide Ameliorates Vascular Calcification Induced by Vitamin D3 Plus Nicotine in Rats

Abstract:Aim: To investigate the role of the endogenous cystathionine γ-synthase (CSE)/ hydrogen sulfide (H 2 S) pathway in vascular calcification in vivo . Methods: A rat vascular calcification model was established by administration of vitamin D3 plus nicotine (VDN). The amount of CSE and osteopontin (OPN) mRNA was determined by using semi-quantitative reverse-transcription polymerase chain reaction. The calcium content, 45 Ca 2+ accumulation and alkaline phosphatase (ALP) activity were measured. H 2 S production and CSE activity were measured. Results: von Kossa staining produced strong positive black/brown staining in areas among the elastic fibers of the medial layer in the calcified aorta. The calcium content, 45 Ca 2+ accumulation and ALP activity in calcified arteries increased by 6.77-, 1.42-, and 1.87-fold, respectively, compared with controls. The expression of the OPN gene was upregulated ( P < 0.01). Expression of the CSE gene was downregulated. However, calcium content, 45 Ca 2+ uptake and ALP activity in the VDN plus NaHS group was lower than that in the VDN group. The content of calcium and 45 Ca 2+ accumulation and activity of ALP in the aorta were 34.8%, 40.75% and 63.5%lower in the low-dosage NaHS group than in the VDN group, respectively ( P < 0.01), and the calcium content and deposition of 45 Ca 2+ and activity of ALP was 83.9%, 37.8 % and 46.2% lower in the aorta in the high-dosage NaHS group than in the VDN group, respectively ( P < 0.01). The expression of the OPN gene was downregulated. Conclusion: The production of H 2 S, and CSE activity were decreased and CSE gene expression was downregulated in rats with vascular calcification. H 2 S can ameliorate vascular calcification, suggesting that the H 2 S/ CSE pathway plays a regulatory role in the pathogenesis of vascular calcification.

Abstract:This work was aimed to explore the changes and significance of adrenomedullin (ADM) mRNA and receptor activity modifying protein 2 (RAMP2) mRNA in calcified vascular smooth muscle cells (VSMCs). Calcification of cultured rat VSMCs was produced by incubation with β-glycerophosphate. Content of ADM released by VSMCs was measured by radioimmunoassay (RIA). The amount of ADM mRNA and RAMP2 mRNA was determined by competitive quantitative RT-PCR. The intracellular calcium content, alkaline phosphatases activity and cellular 45Ca2+-uptake were determined. The results showed that the content of calcium, 45Ca2+-uptake and alkaline phosphatases activity in calcified VSMCs were increased by 118%, 174% and seven-fold (all P<0.01), respectively, compared with control VSMCs. Content of ADM in medium was increased by 99% (P<0.01). Furthermore, it was found that the amount of ADM mRNA and RAMP2 mRNA in calcified cells was elevated by 78 and 56% (all P<0.05), respectively, compared with control. The elevated levels of RAMP2 mRNA were in positive correlation with ADM mRNA (r=0.76, P<0.05) in calcified VSMCs. In conclusion, calcified VSMCs generated an increased amount of ADM, and up-regulated gene expressions of ADM and RAMP2.

Abstract:Objective To explore the impact of carbon monoxide on proliferation and apoptosis of smooth muscle cells in the pulmonary arteries of hypoxic rats.Methods Twenty four rats were randomly divided into four groups: control group(n=7), hypoxic group (n=6), hypoxic + ZnPP group (n=6)and hypoxic +CO group(n=5). An immunohistochemical technique was used to assess the abundance and localization of proliferous cells nucleus antigen(PCNA) and Fas in pulmonary artery smooth muscle cells . TUNEL (TdT-mediated dUTP-biotin nick end labeling) was also used to detect apoptosis cells.Results 1.In rats of hypoxic + CO group, proliferous index (PI) was markedly lower than that of hypoxic group and hypoxic + ZnPP group (P0.01) ;2.Apoptosis index (AI) in hypoxic + CO group rats was increased significantly compared with rats in control group, hypoxic group and hypoxic + ZnPP group (P0.01);3.The expression of Fas in hypoxic + CO group rats was markedly higher than control group, hypoxic group and hypoxic + ZnPP group (P0.01);4.The ratio of PI to AI in rats of hypoxic + CO group was similar to that of control group (≈1).Conclusion Carbon monoxide may suppress proliferation and stimulate apoptosis of smooth muscle cells in pulmonary arteries of rats under hypoxic.
AN ULTRASTRUCTURAL STUDY ON EFFECT OF HYPOXIC PULMONARY VASCULAR IN YOUNG RATS BY HEME- OXYGENASE/CARBON-MONOXIDE SYSTEM

Abstract:Objective To explore the effect of hemeoxygenease/carbon monoxide system on hypoxic pulumonary vascular structural remodeling. Methods Twenty six Wister rats were randomly divided into four groups:control groups( n =6),hypoxia group( n =7),hypxia+ZnPP group( n =6) and hypoxia+CO group( n =7).Pulmonary artery mean pressure(mPAP) of each rat was elaluated by using right cardiac catheterization,the ultrastructaral changes in intra acinar pulmonary muscularized arteries were observed. Results mPAP was significantly increased in hypoxic rats(22±2 31?mmHg)as compared with that of normal controls(16 57±2 51mmHg)( P 0 01),however,mPAP was significantly increased in hypoxic rats treated with ZnPP(26 33±2 34?mmHg)as compared with that of hypoxic rats(22±2 31?mmHg)( P 0 01),mPAP was siginificantly decreased in hypoxic rats treated with CO(19 17±0 98?mmHg) as compared with that of hypoxic rats(22±2 31?mmHg)( P 0 01).Ultrastructure of pulmonary arteries changed obviously in hypoxic rats with the development of hypoxic pulmonary vasuclar structural remodeling,ZnPP increased pulmonary vascular strutral remodeling of hypoxic rats,CO ameliorated pulmonary vascular structural remodeling of hypoxic rats.Conclusion HO/CO plays an important role in the regulation of development of hypoxic pulmonary hypertension and hypxoic pulmonary vascular remodeling.\;[
[The Role of Endogenous CO in the Regulation of Endothelin-Induced VSMC Proliferation and MAPK Activity].

Abstract:Heme oxygenase (HO) is a rate-limiting enzyme of heme degradation, which converts the cellular heme to bilirubin and carbon monoxide (CO). Recently it is suggested that endogenous CO plays an important role in regulating vascular tone under both physiological and pathological conditions, but it is not clear whether endogenous HO/CO system regulates vascular smooth muscle cell (VSMC) proliferation. In the present study, VMSC 3H-TdR incorporation, mitogen-activated protein kinase (MAPK) activity, HO activity and CO release were determined to study the role of endogenous HO/CO system in regulating the VSMC proliferation induced by endothelin-1 (ET-1) in a cultured system. The results showed that ET-1 increased VSMC 3H-TdR incorporation, MAPK activity, HO activity, and CO release were up-regulated. Pretreatment of HO inhibitor, zinc protoporphyrin-9 (ZnPP-9), increased the ET-1-induced VSMC 3H-TdR incorporation and MAPK activity by 31.8% and 36.6% (P < 0.01, respectively), whereas pretreatment of heme-L-lysinate (HLL), a HO substrate, inhibited these activities. This study demonstrated that up-regulation of VSMC endogenous HO represents a cellular protective response to stress or injury. Inhibition of HO may enhance VSMC proliferation induced by ET-1 in vitro, suggesting that endogenous HO/CO system may be directly involved in the regulation of VSMC proliferation through MAPK signaling pathway.
Endogenous Sulfur Dioxide Inhibits Vascular Calcification in Association with the TGF-β/Smad Signaling Pathway

Abstract:The study was designed to investigate whether endogenous sulfur dioxide (SO2) plays a role in vascular calcification (VC) in rats and its possible mechanisms. In vivo medial vascular calcification was induced in rats by vitamin D3 and nicotine for four weeks. In vitro calcification of cultured A7r5 vascular smooth muscle cells (VSMCs) was induced by calcifying media containing 5 mmol/L CaCl2. Aortic smooth muscle (SM) α-actin, runt-related transcription factor 2 (Runx2), transforming growth factor-β (TGF-β) and Smad expression was measured. VC rats showed dispersed calcified nodules among the elastic fibers in calcified aorta with increased aortic calcium content and alkaline phosphatase (ALP) activity. SM α-actin was markedly decreased, but the osteochondrogenic marker Runx2 concomitantly increased and TGF-β/Smad signaling was activated, in association with the downregulated SO2/aspartate aminotransferase (AAT) pathway. However, SO2 supplementation successfully ameliorated vascular calcification, and increased SM α-actin expression, but inhibited Runx2 and TGF-β/Smad expression. In calcified A7r5 VSMCs, the endogenous SO2/AAT pathway was significantly downregulated. SO2 treatment reduced the calcium deposits, calcium content, ALP activity and Runx2 expression and downregulated the TGF-β/Smad pathway in A7r5 cells but increased SM α-actin expression. In brief, SO2 significantly ameliorated vascular calcification in association with downregulation of the TGF-β/Smad pathway.

Abstract:Background Vascular contractility and blood pressure (BP) are regulated by soluble guanylyl cyclase (sGC) and cyclic guanosine monophosphate (cGMP) pathway, which can be influenced by heme oxygenase (HO)-derived carbon monoxide (CO). The age-related changes in sGC/cGMP pathway in tail artery smooth muscle cells (SMCs) in hypertension have not been systematically investigated. Methods In the present study, spontaneously hypertensive rats (SHR) of 4, 8, and 20 weeks old were used. The basal and hemin-modulated levels of sGC and cGMP in tail artery tissues were examined. Results Although BP of 20-week SHR was significantly elevated, sGC and cGMP levels were unaltered compared with age-matched Wistar-Kyoto rats (WKY). The levels of sGC and cGMP were significantly lower in 4- and 8-week SHR compared with age-matched WKY although BP of 4-week SHR was normotensive. Hemin administration resulted in a significant decrease in BP in 8-week (158.7 +/- 2.4 versus 123.5 +/- 1.3 mmHg, P < 0.01), but not in pre-hypertensive (4 weeks) or 20-week SHR or WKY at all ages. Coincidently, sGC and cGMP levels in 8-week SHRs were significantly elevated and so did the expression levels of HO-1. Hemin treatment did not increase the cyclic adenosine monophosphate (cAMP) content of tail artery from 8-week SHR. The constitutive HO-2 levels remained unchanged in 8- and 20-week SHR and age-matched WKY. Conclusion The HO-activity inhibitor, chromium, mesoporphyrin, abolished the BP-lowering and HO-stimulating effects of hemin in young SHR. Our results suggest that alteration in sGC/cGMP pathway in vascular SMCs precedes the occurrence of hypertension but returns to normal once hypertension is fully manifested.
