A Study on the NF-κB activation, serum level and PBMC mRNA expression of cytokine in first-episode schizophrenia

Abstract:Objective To measure the activation and the serum levels of the nuclear factor kappa B(NF-κB),PBMC mRNA expression of IL-1β,TNF-α and IL-6,and to analyze the interaction among them for exploring the role of NF-κB in the synthesis of cytokine in schizophrenia.Methods 83 patients with first-episode schizophrenia and 65 healthy control subjects were included in the study.Transcription Factor Assay Kits was employed to measure NF-κB activation and reverse transcription-polymerase chain reaction(RT-PCR)technique to semi-quantitatively analyze IL-1β,IL-6 and TNF-α mRNA expression in the PBMC.The serum levels of IL-1β,IL-6 and TNF-α were estimated using commercial ELISA kits according to the manufacturer's instructions.Results NF-κB activation in the schizophrenic patients was significantly higher than that in the controls(P<0.05).Serum IL-1β,IL-6 and TNF-α levels in the schizophrenics were significantly higher than those in the controls(P<0.05).IL-1β,IL-6 and TNF-α mRNA expression in the PBMC of the schizophrenics were significantly higher than those of the controls(P<0.05).There were significantly positive correlation among the activation and serum level of NF-κB,mRNA expression of IL-1β or TNF-α in both the schizophrenic group and control group(P<0.05).But there were no significant correlation between the activation and serum level of NF-κB,mRNA expression of IL-6 in both the schizophrenia group and control group(P>0.05).Conclusions Schizophrenic patients tended to be in a state of immune activation.The activating PBMC was one of major sources of cytokine for schizophrenic patient.Activated NF-κB plays an important role in mediating the expression of IL-1β and TNF-α gene in schizophrenia.
