A STUDY ON THE CORRELATION BETWEEN THE REGULATED EXPRESSION OF MATRIX METALLOPROTEINASE 9(MMP-9) AND THE METASTATIC PHENOTYPE IN HUMAN MELANOMA CELLS

Abstract:Objective To investigate the correlation between MMP-9 and tumor metastasis.Methods One self-contained tetracycline-regulated retroviral vector containing antisense cDNA of MMP-9 was constructed and transfected into a metastatic human melanoma cell line WM451,which has a high MMP-9 expression level.In vitro and in vivo test such as Zymography as well as xenograftion in nude mice experiment were used to analyse expression of MMP-9 and in vivo behavior of tumor cells before and after gene transfection of mmp-9 and in vivo behavior of tumor cells before and after gene transfection.Results After transfection of antisense MMP-9 the expression and activity of MMP-9 decreased,the capacity of spontaneous metastasis in nude mice was suppressed significantly.Conclusion The decrease of MMP-9 expression in human melanoma cell line by MMP-9 antisense RNA could inhibit the metastatic phenotype of it and demonstrated that MMP-9 play a central role in metastasis.
