Abstract:Some agronomic characteristics such as height, stem diameter, relative leaf area and leaf length breadth ratio and components such as water content, total polysaccharide, water-soluble polysaccharide, and cellulose contents of 8 kinds Dendrobium officinale from different distribution areas were determined and analyzed. It was found that the differences among these agronomic characteristics have to some extent related to where they are from. Dendrobium officinale from Yandang and Yunnan areas which have moderate plant morphology and robust stem are better then that in other areas for planting in Zhejiang, but for Dendrobium officinale from Yunnan, the greenhouse is nessesary.

Abstract:New jute variety, “Zhonghuangma No.1( former name C90-2)”, was bred from progeny of 71-8×79-51 backcrossed with 79-51, by systematical selection and recurrent selection method for six generations selection in 1990. In the variety yield comparison test in 1993-1994, it's average yield was 2814.00kg/hm2, which was 14.32%more than that of the CK Yueyuan No.5. The average yield of C90-2 was 2808.00kg/hm2, 13.59%higher than that of CK Yueyuan No.5 (2472.15kg/hm2) in the national jute regional trial at 5 locations for 2 years (2002-2003), and the difference was to significant level. Its average strength was 401N, fineness 439.0m/g, better than that of the CK. The main Characteri stics of the variety were larger plant, thicker stem, higher resistance to anthracnose disease, high yield stability and wide adaptability, which is one of the most promising varieties at present and can completely substitute imported “Bangladeshi fiber and provide material for civil luxurious jute product.
Comparative Study of the Impact of Drought Stress on P.centrasiaticum at the Seedling Stage in Tibet

Abstract:Pennisetum centrasiaticum is widely distributed in arid and semi-arid areas of Tibet.Its rhizome system is developed and has strong resistance to adversity.In this study,the physiological characteristics and drought re-sistance of P.centrasiaticum seedlings from 12 drought-stressed sites in Tibet were examined at the Lhasa Plateau Ecosystem Research Station of the Chinese Academy of Sciences.PEG-6000 solution with five levels of water potential(0,-0.7,-1.4,-2.1,and-2.8 MPa)was used to simulate drought stress,and malondialdehyde(MDA),proline(Pro)and chlorophyll contents were determined.The balance between production and elimination of reac-tive oxygen species in P.centrasiaticum was destroyed,leading to membrane lipid peroxidation and the production of MDA,and accelerating the decomposition of chlorophyll.P.centrasiaticum absorbed water from the outside to resist drought by secreting proline and other osmotic regulating substances.The Pro and chlorophyll contents in P.centrasiaticum showed a temporary rising trend,and then decreased with the decrease in water potential.MDA content increased with the decrease in water potential.By using the membership function method,the drought re-sistance of P.centrasiaticum seedlings from the 12 areas was evaluated,and the results showed that the drought resistance at the sites went from strong to weak in this order: Xietongmen > Linzhou > Sog > Damxung > Tingri >Namling>Gyirong>Linzhi>Purang>Dingjie>Longzi>Sa'gya.The drought resistance of P.centrasiaticum was strong in Xietongmen,Linzhou and Sog.Whether P.centrasiaticum from these three areas is suitable for cultivation in arid and semi-arid areas of Tibet needs further study.

Abstract:We analyzed vitamin C, beta-carotene and nitrate content in thirty different carrot (Dacus carota var. sativa DC) genotypes sown in the spring and autumn. The genotypes were divided into three classes, high-quality, medium-quality and low-quality, by the cluster analysis method of the NOSA (National Occupational Safety Association) software. In the high-quality class, two genotypes, 'Shenhesusheng' and Japanese 'Sanhongqicun,' were selected to be recommended for fall planting. The Japanese 'Heitianwucun,' was recommended for spring planting.
Evaluation of drought-tolerant varieties based on root system architecture in cotton (Gossypium hirsutum L.)

Abstract:Background: Root system architecture (RSA) exhibits significant genetic variability and is closely associated with drought tolerance. However, the evaluation of drought-tolerant cotton cultivars based on RSA in the field conditions is still underexplored. Results: So, this study conducted a comprehensive analysis of drought tolerance based on physiological and morphological traits (i.e., aboveground and RSA, and yield) within a rain-out shelter, with two water treatments: well-watered (75 ± 5% soil relative water content) and drought stress (50 ± 5% soil relative water content). The results showed that principal component analysis identified six principal components, including highlighting the importance of root traits and canopy parameters in influencing drought tolerance. Moreover, the systematic cluster analysis was used to classify 80 cultivars into 5 categories, including drought-tolerant cultivars, relatively drought-tolerant cultivars, intermediate cultivars, relatively drought-sensitive cultivars, and drought-sensitive cultivars. Further validation of the drought tolerance index showed that the yield drought tolerance index and biomass drought tolerance index of the drought-tolerant cultivars were 8.97 and 5.05 times higher than those of the drought-sensitive cultivars, respectively. Conclusions: The RSA of drought-tolerant cultivars was characterised by a significant increase in average length-all lateral roots, a significant decrease in average lateral root emergence angle and a moderate root/shoot ratio. In contrast, the drought-sensitive cultivars showed a significant decrease in average length-all lateral roots and a significant increase in both average lateral root emergence angle and root/shoot ratio. It is therefore more comprehensive and accurate to assess field crop drought tolerance by considering root performance.
What Drives the Morphological Traits of Stress-Tolerant Plant Cynodon Dactylon in a Riparian Zone of the Three Gorges Reservoir, China

Abstract:The cyclical process of water storage and recession in the regular operation of the Three Gorges Reservoir creates a unique habitat stress that alters the structural and functional attributes of vegetation ecology within the riparian zone. The stress-tolerant plant Cynodon dactylon (L.) Pers is the dominant plant species in the riparian zone of the Three Gorges Reservoir. In this study, the riparian zone of the Daning River, a tributary located in the center of the Three Gorges Reservoir, was selected as our study area. To identify the drivers of the morphological traits of C. dactylon in the riparian zone of Daning River, we examined plant biomass and plant characteristics across different elevation gradients, with reference to abiotic factors to determine the distribution patterns of plant morphological traits. Results indicated that in the two main soil types of the riparian zone, plant biomass showed a consistent trend along the elevation gradient, with a “middle-height expansion” pattern; biomass increased and then decreased with rising water levels. Plant biomass positively correlated with soil total nitrogen and negatively correlated with soil pH, electrical conductivity, and total phosphorus. C. dactylon adapted to prolonged flooding in the riparian zone by having a significant negative correlation between plant height and erect stem length with soil moisture content to facilitate root respiration.
Evaluation of Diversity Based on Morphological Variabilities and ISSR Molecular Markers in Iranian Cynodon dactylon (L.) Pers. Accessions to Select and Introduce Cold-Tolerant Genotypes

Abstract:The main goals of the present study were to screen Iranian common bermudagrasses to find cold-tolerant accessions and evaluate their genetic and morphological variabilities. In this study, 49 accessions were collected from 18 provinces of Iran. One foreign cultivar of common bermudagrass was used as control. Morphological variation was evaluated based on 14 morphological traits to give information about taxonomic position of Iranian common bermudagrass. Data from morphological traits were evaluated to categorize all accessions as either cold sensitive or tolerant using hierarchical clustering with Ward's method in SPSS software. Inter-Simple Sequence Repeat (ISSR) primers were employed to evaluate genetic variability of accessions. The results of our taxonomic investigation support the existence of two varieties of Cynodon dactylon in Iran: var. dactylon (hairless plant) and var. villosous (plant with hairs at leaf underside and/or upper side surfaces or exterior surfaces of sheath). All 15 primers amplified and gave clear and highly reproducible DNA fragments. In total, 152 fragments were produced, of which 144 (94.73%) being polymorphic. The polymorphic information content (PIC) values ranged from 0.700 to 0.928. The average PIC value obtained with 15 ISSR primers was 0.800, which shows that all primers were informative. Probability identity (PI) and discriminating power between all primers ranged from 0.029 to 0.185 and 0.815 to 0.971, respectively. Genetic data were converted into a binary data matrix. NTSYS software was used for data analysis. Clustering was done by the unweighted pair-group method with arithmetic averages and principle coordinate analysis, separated the accessions into six main clusters. According to both morphological and genetic diversity investigations of accessions, they can be clustered into three groups: cold sensitive, cold semi-tolerant, and cold tolerant. The most cold-tolerant accessions were: Taft, Malayear, Gorgan, Safashahr, Naein, Aligoudarz, and the foreign cultivar. This study may provide useful information for further breeding programs on common bermudagrass. Selected genotypes can be evaluated for other abiotic stresses such as drought and salinity.

Abstract:Drought stress considered a key restrictive factor for a warm-season bermudagrass growth during summers in China. Genotypic variation against drought stress exists among bermudagrass ( Cynodon sp .), but the selection of highly drought-tolerant germplasm is important for its growth in limited water regions and for future breeding. Our study aimed to investigate the most tolerant bermudagrass germplasm among thirteen, along latitude and longitudinal gradient under a well-watered and drought stress condition. Current study included high drought-resistant germplasm, "Tianshui" and "Linxiang", and drought-sensitive cultivars; "Zhengzhou" and "Cixian" under drought treatments along longitude and latitudinal gradients, respectively. Under water deficit conditions, the tolerant genotypes showed over-expression of a dehydrin gene cdDHN4 , antioxidant genes Cu/ZnSOD and APX which leads to higher antioxidant activities to scavenge the excessive reactive oxygen species and minimizing the membrane damage. It helps in maintenance of cell membrane permeability and osmotic adjustment by producing organic osmolytes. Proline an osmolyte has the ability to keep osmotic water potential and water use efficiency high via stomatal conductance and maintain transpiration rate. It leads to optimum CO 2 assimilation rate, high chlorophyll contents for photosynthesis and elongation of leaf mesophyll, palisade and thick spongy cells. Consequently, it results in elongation of leaf length, stolon and internode length; plant height and deep rooting system. The CdDHN4 gene highly expressed in "Tianshui" and "Youxian", Cu/ZnSOD gene in "Tianshui" and "Linxiang" and APX gene in "Shanxian" and "Linxiang". The genotypes "Zhongshan" and "Xiaochang" showed no gene expression under water deficit conditions. Our results indicate that turfgrass show morphological modifications firstly when subjected to drought stress; however the gene expression is directly associated and crucial for drought tolerance in bermudagrass. Hence, current research has provided excellent germplasm of drought tolerant bermudagrass for physiological and molecular study and future breeding.

EVALUATION AND UTILIZATION OF MAJOR CHARACTERS OF CHINESE UNIQUE WHEAT GERMPLASM RESOURCES

Abstract:57 accessions of Chinese unique wheat germplasm from Tibet, Xinjiang. Yunnan and semi-wild wheat from Tibet have been evaluated. Cytological observation on root tip cells showed that all Tibet wheat, Yunnan wheat, Xinjiang wheat and Tibet semi-wild wheat had chromosome number of 2n= 42 and two pairs of satellite chromosome. The meiosis processes of pollen mother cells (PMC) from Yunnan wheat and Tibet wheat were found to be relatively unstable. Its esterase isozyme is tissue specificity. Cluster analysis illustrated that Yunnan wheat and Tibet semi-wild wheat were genetical close related and belong to the same taxonomical group, while Xinjiang wheat was an unique taxonomical group. Tibet semi-wild wheat has high crossability with rye. The seed set rates of Tibet semi-wild wheat varieties T494, T511-1 and T473 crossed with rye were higher than that of Chinese spring wheat crossed with rye, suggesting the genotype of above varieties was kr1kr1kr2kr2. The chromosome cross number of PMC at meta-phase I of F1 hybrid between Tibet semi-wild wheat and rye was higer than those of F1 hybrids between Chinese spring wheat and rye, suggesting varieties T445-3, T498-2, T488, T511-1 might have similar ph2 gene.

Evaluation on the Germplasm of Maize (zea Mays L.) Landraces from Southwest China.

Abstract:Because of their local adaptation and economic factors that limit the adoption of commercial hybrids, farmer-saved maize landraces are still grown over a considerable area concentrated in southwest China. To evaluate the potential of using maize landraces, the germplasm characteristics of 96 landraces from southwest China were evaluated at phenotypic, cellular, and molecular levels. The existence of high phenotypic variation and elite germplasm tolerant to low-N, low-P, as well as drought stress was observed. Of the total landraces, 81.25, 7.29, 5.21, and 2.08% were found with 0, 1, 2, 3, and 4 B chromosomes. Using 42 microsatellite (simple sequence repeat) loci, 246 alleles were detected among the landraces. The number of alleles per SSR locus varied from 2 to 10, averaging 5.67 allele per locus, which revealed a high level of genetic diversity of maize landraces in southwest China. Cluster analysis showed that 96 landraces could distinctly be clustered into four groups, which tended to associate with their geographic origins. We propose that the genetic diversity center of maize landraces in southwest China might be in Sichuan. A sharp genetic deviation from Hardy-Weinberg equilibrium was observed from heterozygosity deficiency and a considerable genetic variation was revealed within, rather than among, the landraces. Based on their germplasm characteristics, the innovation and utilization of maize landraces in southwestern China for theoretical and applied research could be achieved by constructing heterosis groups, developing inbred lines with high combining ability, and maintaining the landraces with elite germplasm and B chromosomes using bulked pollen.

Abstract:The genetic mechanism of favorable allele variation in germplasm resources was revealed to fully exploit their excellent agronomic and drought resistance traits. For many years, phenotypic evaluation and quality analysis were used to identify 66 strong drought-resistant varieties and one strong gluten variety from 323 wheat local germplasm resources. The 55 K SNP marker was used to investigate the association between major agronomic traits. Under irrigation conditions, 17 SNP loci were significantly correlated with major agronomic characteristics, 8 related candidate genes were annotated. Under drought stress and irrigation conditions, 32 SNP loci were significantly correlated with major agronomic characteristics, 24 related candidate genes were annotated. Three SNP loci were significantly associated with multiple traits, one SNP loci was stable and correlated considerably with spike length in multiple environments.154 significant association sites with drought resistance-related traits in wheat maturity were identified using whole gene association analysis of drought tolerance coefficient of yield-related characteristics, 50 functional candidate genes were annotated and six candidate genes were screened out that may participate in the drought stress response of yield-related traits at the adult stage. These findings also served as a foundation for subsequent gene cloning and molecular marker-assisted drought-resistant wheat breeding.

Abstract:Sixteen cotton cultivars widely planted in China were sowed under five different drought concentrations (0, 2.5, 5, 7.5, and 10%) using PEG6000 to screen the indices of drought resistance identification and explore the drought resistance of different cotton cultivars. Eighteen physiological indices including root, stem, and leaf water contents (RWC, SWC, and LWC), net photosynthetic rate (Pn), the maximum photochemical quantum yield (Fv/Fm), the actual photochemical quantum yield (φPSII), non-photochemical quenching coefficient (NPQ), leaf water potential (LWP), osmotic potential (ψs), leaf relative conductivity (REC), leaf proline content (Pro), leaf and root soluble protein contents (LSPC and RSPC), leaf and root malondialdehyde (MDA) contents (LMDA and RMDA), root superoxide dismutase, peroxidase, and catalase activities (RSOD, RPOD, and RCAT) were measured. Results indicated the 18 physiological indices can be converted into five or six independent comprehensive indices by principal component analysis, and nine typical indices (Fv/Fm, SWC, LWP, Pro, LMDA, RSPC, RMDA, RSOD, and RCAT) screened out by a stepwise regression method could be utilized to evaluate the drought resistance. Moreover, the 16 cotton cultivars were divided into four types: drought sensitive, drought weak sensitive, moderate drought resistant, and drought resistant types. The resistance ability of two selected cotton cultivars (drought resistant cultivar, Dexiamian 1; drought sensitive cultivar, Yuzaomian 9110) with contrasting drought sensitivities were further verified by pot experiment. Results showed that the responses of final cotton biomass, yield, and yield composition to drought were significantly different between the two cultivars. In conclusion, drought resistant cultivar Dexiamian 1 and drought sensitive cultivar Yuzaomian 9110 were screened through hydroponics experiment, which can be used as ideal experimental materials to study the mechanism of different cotton cultivars with contrasting drought sensitivities in response to drought stress.

Abstract:Abstract To assess the adaptability of Camellia oleifera for introduction in new growth locations, this study evaluated 10 representative C. oleifera cultivars from the main areas in China where this oil-producing evergreen crop is grown. Cluster analysis, correlation analysis, and membership function analysis were used to evaluate various indices of the selected C. oleifera cultivars, including flowering phenology, cold tolerance, leaf structure, pollen characteristics, and pollen viability. The correlation analysis identified the full blossoming time, leaf palisade and spongy tissue thickness, pollen deformity rate, and pollen activity as key indices for determining the adaptability of the cultivars to new areas. The membership function analysis of the 10 C. oleifera cultivars revealed the following order of adaptability: ‘XLC25’ > ‘Changlin4hao’ > ‘Ganzhouyou8hao’ > ‘Ganzhouyou6hao’ > ‘Tiechengyihao’ > ‘Eyou465’ > ‘XLC10’ > ‘Changlin3hao’ > ‘Changlin18hao’ > ‘QY235.’ When introducing C. oleifera cultivars to new regions, the higher-ranked cultivars are more likely to be successful. The results of this study may provide a new direction for the comprehensive assessment of plant introduction and domestication potential, i.e., the assessment of the vegetative and reproductive growth, adversity resistance, and blossoming time of plants.

Ecotype Division and Chemical Diversity of Cynomorium songaricum from Different Geographical Regions

Abstract:Cynomorium songaricum is an important endangered plant with significant medicinal and edible values. However, the lack of resources and quality variation have limited the comprehensive developments and sustainable utilization of C. songaricum. Here, we evaluated the chemical and genetic traits of C. songaricum from the highly suitable habitat regions simulated with species distribution models. The PCA and NJ tree analyses displayed intraspecific variation in C. songaricum, which could be divided into two ecotypes: ecotype I and ecotype II. Furthermore, the LC-MS/MS-based metabolomic was used to identify and analyze the metabolites of two ecotypes. The results indicated that a total of 589 compounds were detected, 236 of which were significantly different between the two ecotypes. Specifically, the relative content and the kind of flavonoids were more abundant in ecotype I, which were closely associated with the medicinal activities. In contrast, amino acids and organic acids were more enriched in ecotype II, which may provide better nutritional quality and unique flavor. In summary, our findings demonstrate the ecotype division and chemical diversity of C. songaricum in China from different geographical regions and provide a reference for the development of germplasm and directed plant breeding of endangered medicinal plants.

Abstract:Background: Drought-resistant varieties are an important way to address the conflict between wheat's high water demand and the scarcity of water resources in the North China Plain (NCP). Drought stress impacts many morphological and physiological indicators in winter wheat. To increase the effectiveness of breeding drought-tolerant varieties, choosing indices that can accurately indicate a variety's drought resistance is advantageous. Results: From 2019 to 2021, 16 representative winter wheat cultivars were cultivated in the field, and 24 traits, including morphological, photosynthetic, physiological, canopy, and yield component traits, were measured to evaluate the drought tolerance of the cultivars. Principal component analysis (PCA) was used to transform 24 conventional traits into 7 independent, comprehensive indices, and 10 drought tolerance indicators were screened out by regression analysis. The 10 drought tolerance indicators were plant height (PH), spike number (SN), spikelet per spike(SP), canopy temperature (CT), leaf water content (LWC), photosynthetic rate (A), intercellular CO2 concentration (Ci), peroxidase activity (POD), malondialdehyde content (MDA), and abscisic acid (ABA). In addition, through membership function and cluster analysis, 16 wheat varieties were divided into 3 categories: drought-resistant, drought weak sensitive, and drought-sensitive. Conclusion: JM418, HM19,SM22, H4399, HG35, and GY2018 exhibited excellent drought tolerance and,therefore, can be used as ideal references to study the drought tolerance mechanism in wheat and breeding drought-tolerant wheat cultivars.

Abstract:Dendrobium nobile Lindl. is one of the most important Orchid plants worldwide. The genotype-by-sequencing (GBS) method has now been widely used to access genetic diversity because of its high-throughput and cost-effective in molecular markers. The goal of this study was to employ the GBS technique for diversity evaluation of D. nobile and determine genetic differences between populations. A total of 129 accessions of D. nobile collected from 10 imitation-wild cultivated populations growing in Sichuan, Guizhou and Yunnan of southwestern China were sequenced, a total of 135 G clean reads and a total of 836,786 high quality SNPs were yielded and used for analysis of genetic diversity and population structure. The accessions of D. nobile were classified into three groups, excepts for 14 accessions belonging to admixed group. Genetic variation was greater within populations (87.80%) than among populations (12.20%). There was gene exchange between different populations. The 10 populations were classified into three groups, and group III could be further divided into two subgroups. The results will not only provide valuable information for the level of genetic diversity of D.nobile growing in southwestern of China, but also help for formulation of strategies for genetic resource protection and utilization. The accessions used in this study could be applied to the scientific research, breeding and production of D. nobile .

Methods of Comprehensive Evaluation for Drought Resistance in Sweet Potato Cultivars

Abstract:The methods of drought coefficient,cluster analysis and subordinate function were used in comprehensive evaluation for drought resistance adaptability in sweet potato cultivars.The tested components included morphological characters,plant growth,economic traits,physiological and biochemical values in the cultivars grown under water stress.On the basis of correlation analysis between the drought resistance indices obtained from the above-mentioned components and the drought resistance in cultivars,it was shown that 15 tested cultivars could be divided into four classes of tolerance.Most tolerant cultivars were Y3 and Chaoshu 1,followed by 92-111-107,Yushu 34,Nanshu 88 and 92-103-30.Less tolerant were Suyu 1,9318-58,Yusu 303,89-1,95-411-153 and Ximeng.The rest 3 cultivars were not tolerate at all.Above several comprehensive evaluation methods for drought resistance adaptability in sweet potato cultivars are well coincident.

Assessment of Pear Germplasm from the Middle Area of Gansu Province Using Amplified Fragment Length Polymorphism Markers

Abstract:Genetic diversity and genetic relationship of 40 accessions of pear (Pyrus L.) germplasm native to the middle area of Gansu province in China were analyzed using amplified fragment length polymorphism (AFLP)marker. 6 primer combinations generated 472 scored bands, among which 404 (86%) were polymorphic, which showed the abundant genetic diversity of these pear gerrmplasm. Based on the UPGMA cluster analysis, 40 pear accessions could be obviously divided into 7 major groups at the level of the coefficient of 0.72: P. communis L, P. sinkiangensis Yu, P. pyrifolia Nakai var. sinensis Y. Teng et K. Tanabe (Chinese white pears), P. xerophila Yu, P. phaeocarpa Rehd. and two P. ussuriensis Maxim groups. All the Chinese white pears were clustered into one group with Huanghuali, which belonged to P. pyrifolia. Unidentified pear cultivars or types were clustered with P. ussuriensis, P. communis, P. xerophila and Chinese white pears, respectively. The research indicated that the classification system based on AFLP could reflect the genetic diversity and relationship of Pyrus species and cultivars from Gansu province.
