Early Life Forepaw Sensorimotor Deprivation Impairs the Synaptic Plasticity at the Schaffer-CA1 Synapse in Hippocampus of Young Rats

Abstract:【Objective】To examine the influence of forepaw sensorimotor deprivation on synaptic plasticity at the Schaffer-CA1 synapse in hippocampus of young rats. 【Methods】Newborn SD rats were randomly assigned into two groups,the experiment group(deprived of forepaw sensorimotor function) and the control group(sham operated).Rats of the experiment group were deprived of forepaw sensorimotor function at PN13.Electrophysiological recordings in vitro were performed at the time point of PN25,PN35,PN45 and PN60 to measure long-term potentiation in the Schaffer-CA1 pathway of hippocampal slices from young rats. 【Results】Both groups exhibited LTP,while experiment group showed significantly less LTP in comparison to sham control group at PN25 and PN35,as the changes of field excitatory postsynaptic potentials(fEPSPs) slope were much lower [(13.39±2.90)% vs(30.39±4.76)%,F=9.31,P0.01;(18.92±4.00)% vs(39.38±4.84)%,F=10.95,P0.01]. 【Conclusions】Conclusion Forepaw sensorimotor deprivation in early life inhibits the induction and maintenance of LTP in the Schaffer-CA1 pathway and leads to the impairments on synaptic plasticity of hippocampus in rats,this is the possible electrophysiological mechanism for impairments of learning and memory.
