The Effects of Salvia Miltiorrhiza on the Expression of Heme Oxygenase-1 in Rat Isolated Liver

Abstract:Objective To study the effects of salvia miltiorrhiza on the expression of HO-1 mRNA and protein in low temperature preserved rat isolated liver.Methods Isolated liver was harvested and preserved in the 4℃ preserved solution simulating clinical procedures.Rats were divided into 3 groups according to different perfusion and preserved solutions: Balance solution group(Group A),Salvia miltiorrhiz group(Group B),and Salvia miltiorrhiza and inhibitor preconditioning group(Group C).Each group was subdivided into 4 subgroup according preserved times(0,1,3,6h;n=6).Pretreatment intraperitoneal injection of saline 24h before operation was performed in A and Group B,while ZnPP replaced of saline in Group C.Expression of HO-1 mRNA and protein was examined with RT-PCR and immunohistochemisty,respectively.Hepatocellular morphologic change was assessed with pathological section and scoring system.Correlations between the expression level of HO-1 mRNA and protein and liver injury score were analyzed.Results(1) The expression levels of HO-1 mRNA and protein were significantly higher in Group B than Group A and Group C(P0.05);(2) At all time points except for 0h,Group B had the mildest liver injury and the longest preservation time while Group C had the most severe liver injury;(3) There were significant correlations between the expression levels of HO-1 mRNA and protein and the liver injury score.Conclusion Induction of HO-1 expression by Salvia miltiorrhiza may account for its protective effects in low temperature-preservated rat isolated liver.
