The relationship between model predictive ability and validated method in QSAR/QSPR study

Abstract:Validation of the model predictive ability is a crucial aspect for any quantitative structure-activity relationship(QSAR) model. Since long,leave-one-out or leave-multiple-out cross-validation were used to determine the model predictive ability.However,at an international workshop held in Setubal(Portugal),several principles for assessing the validity of QSAR were proposed in 2004,as the"Organization for Economic Co-operation and Development(OECD) validation Principles",which emphasize that the model predictive ability must be validated by external validation set.In this study,the QSAR study is developed,employing 2D-autocorrelation descriptors with a set of 37 naphthoquinone ester derivatives and employing topological index and connective index with a set of 45 testosterone and dihydrotestosterone analogues,where all descriptors are calculated by E-Dragon software.The increase n subtraction 1 algorithm is used to select descriptors.Then support vector machine(SVM) is utilized to construct the quantitative structure-activity relationship models.At last we have used different methods to confirm the model predictive ability.The results show that the model prediction ability and the results of internal validation are not necessarily connected,but the evaluation function which combines the results of external validation set and training set can be a very good method to judge the model prediction ability.
