Abstract:Response surface methodology(RSM) was employed to optimize the extraction conditions of total flavonoids from Anoectochilus roxburghii.Based on single-factor tests,Box-Benhnken center composite experiment was carried out with four factors,including alcohol concentration,extraction temperature,extraction time and ratio of liquor to material.Meanwhile,the experimental data were analyzed by Design Expert 7.1.6 software.The results showed that the factors affecting the flaronoids content from Anoectochilus roxburghii were extraction temperature,alcohol concentration,extraction time and ratio of liquor to material.The optimum extraction conditions by RSM were extraction time 89.8 min,extraction temperature 62.25 C,alcohol concentration 80.68％ and ratio of liquor to material 79.38∶ 1.Under these conditions,the extraction yield of total flavonoids was 2.243 7 mg/g,which was close to the predicted value 2.338 6 mg/g.Using the method of response surface methodology to analyze the optimization process of flavonoids extraction from Anoectochilus roxburghii is accurate,reliable and feasible,which is useful for the extraction of flavonoids from Anoectochilus roxburghii and provides the certain experimental basis for deep research of Anoectochilus roxburghii resources.
