The Ultrasonic Mapping of Frog Muscular Electro-Mechanical Activity

Abstract:Objective To investigate frog muscular electron-mechanic activity under physiological or pathological conditions by using a M model of Doppler ultrasound.Methods One hundred thirty-two samples of bullfrog legs skeletal muscle were prepared.The samples were divided into 3 groups: single point monopole(N = 54),double point monopole(N = 56),and electromechinical conductionblock(N = 53),respectively.The muscles were stimulated by pulse of different voltages.The electron-mechanic conduction delay-time(t 1),mechanic conduction time(t 2) and duration time of mechanic contraction(Tm) were recorded by using the M model of Doppler.At the same time,time interval difference of two poles was measured by using an Electro-Physiography.Differences of all indexes within or among groups were compared.Results ①Within groups there were no significant differences in t 1,t 2 and Tm under conditions of different voltage and identical distance.However,changing Doppler scale but keeping identical voltage and distance,there was significantly difference for t 2(P 0.05).②No significant difference in t 1 or t 2 was observed between single monopole and double monopole groups under conditions of identical stimulant distance and stimulant voltage while t 2 of one or two voltages in single point monopole groups was longer than that in double point monopole group under conditions of identical distance(P 0.05).③After injection of alcohol between two poles Tm was significantly longer but t 2 shorter than injection before(P 0.05).④ There were no significant correlations among T,t 1,t 2 and Tm.Conclusions Single point monopole stimulation did not speed up t 2 of frog skeletal muscle even with high voltages,but double point monopole stimulation shortened t 2 with increased voltages.Therefore,the results may provide some important reference of electrophysiological and mechanical information for mechanical conduction effect of different pacing treatments used in clinic,and thereby may improve pacing modes in the future.
