Abstract:Aim To investigate whether ketamine suppresses lipopolysaccharide (LPS)-induced increase in Toll-like receptor 4 (TLR4) expression and nuclear factor-kappa B (NF-kappa B) activity in the intestines of rats.Methods Six groups of rats received one of the following: normal saline control, LPS (5 mg/kg) plus saline, LPS (5 mg/kg) plus ketamine (0.5 mg/kg), LPS (5 mg/kg) plus ketaminc (2-5 mg/kg), LPS (5 mg/kg) plus ketamine (10 mg/kg), or ketamine (10 mg/kg) alone. Intestinal TLR4 mRNA expression was analyzed by reverse transcription polymerase chain reaction (RT-PCR), and NF-kappa B activity was tested by electrophoretic mobility shift assay (EMSA) 1, 3, or 5 hours after the LPS injection.Results Lipopolysaccharide increased TLR4 expression and NF-kappa B activity in the intestines of rats. Ketamine at the dosage of 0.5, 2.5, and 10 mg/ kg suppressed the LPS-induced increase in TLR4 expression and NF-kappa B activity. Ketamine alone had no effect. Conclusion The study demonstrated that ketaminc inhibits NF-kappa B activation in the intestines of LPS-treated rats, possibly by suppressing TLR4 expression.Conclusion The study demonstrated that ketamine inhibits NF-kappa B activation in the intestines of LPS-treated rats, possibly by suppressing TLR4 expression.

Abstract:The aim of the study was to investigate the effects of ketamine on pulmonary Toll-like receptor 4 (TLR4) expression and nuclear factor kappa-B (NF-kappa-B) activity in rats with endotoxemia. Rats were treated with lipopolysaccharide (LPS, 5 mg/kg) to induce endotoxemia, followed immediately by ketamine (0.5, 5 and 10 mg/kg) or 0.9% NaCl (5 ml/kg). At 1, 3 and 5 h posttreatment, TLR4 mRNA expression and NF-kappa-B activity of the rat lungs were analyzed by reverse transcription-polymerase chain reaction (RT-PCR) and electrophoretic mobility shift assay (EMSA), respectively. LPS increased TLR4 expression and NF-kappa-B activity in the rat lungs. Ketamine at the dose levels of 5 and 10 mg/kg suppressed the LPS-induced elevation of TLR4 expression and NF-kappa-B activity. Ketamine alone had no effect on either one of them. The present study revealed that ketamine could inhibit pulmonary NF-kappa-B activity during endotoxemia in rats, and this antiinflammatory effect possibly acts through suppressing TLR4 expression.
Ketamine Inhibits NF-kappa B Activation and TNF-α Production Induced by LPS in Vivo

Abstract:Objective: To investigate the effect of ketamine on the endotoxin-induced NF-kappa B (NF-κB) activation and TNF-α production in vivo. Methods: The sepsis model was induced by a bolus injection of endotoxin (5 mg/kg) into the tail vein of adult male Wistar rats. Then all animals were immediately treated with different doses of ketamine (0.5, 5, 50 mg/kg) or normal saline (10 ml/kg) intraperitoneally(ip). After two hours, the intestines, livers, lungs and kidneys of each rat were dissected and kept for NF-κB and TNF-α detection. Results: Endotoxin enhanced NF-κB activity and TNF-α production in the intestine, liver, lung and kidney. Ketamine at a dose of more than 0.5 mg/kg suppressed endotoxin-induced NF-κB activation and TNF-α production in the intestine and kidney. However, the least dose of ketamine to inhibit NF-κB activation and TNF-α production in the lung was 5 mg/kg. Three different doses of ,etamine (0.5, 5, 50 mg/kg) could not inhibit NF-κB activation and TNF-α production in the liver, while ketamine at the dose of 50 mg/kg enhanced NF-κB activity and TNF-α production in the liver. Conclusion: These findings suggest that ketamine can suppress NF-κB and TNF-α production in vivo. Subanesthetic dose of ketamine has anti-inflammatory effect. However, ketamine at a large dose of more than 50 mg/kg may be harmful to the endotoxin-induced sepsis.

Abstract:. Objective and design: To observe the effect of ketamine on polymorphonuclear leucocytes (PMN) adhesion and respiratory burst activity in endotoxemia rats. Materials: 30 rats were randomly allocated to five groups: rats challenged with intraperitoneal injection of saline (saline group); challenged with intraperitoneal injection of LPS 10 mg/kg (LPS group); challenged with intraperitoneal injection of LPS 10 mg/kg and treated by intraperitoneal injection of ketamine 5, 25, 50 mg/kg at 0, 1, 2, 3, 4, 5 h after the injection of LPS, respectively (three ketamine treatment groups). Methods: PMN respiratory burst and CD11b expression were measured with flow cytometry at the end of 1 h, 4 h, and 6 h. Results: LPS challenge significantly increased PMN respiratory burst activity and CD11b expression when compared with the saline group (p < 0.01). There was a significant decrease in LPS-induced PMN respiratory burst activity and CD11b expression in three ketamine treatment groups when compared with LPS group (p < 0.01). Conclusions: Ketamine significantly inhibits PMN CD11b expression and respiratory burst activity in endotoxemic rats.

Abstract:The effects of ketamine on endotoxin-induced NF-KB activation and TNF-alpha expression were studied in vivo. A sepsis model was created by bolus injection of endotoxin (5 mg/kg) into the tail vein of adult Wistar rats. The rats were immediately treated with various doses of ketamine (0.5, 5, or 50 mg/kg) or 0.9% NaCl (10 ml/kg) ip. At 1, 4, or 6 hr post-treatment, NF-KB and TNF-alpha were assayed in the intestine, liver, and lung by electrophoretic mobility shift assay (EMSA) and reverse-transcription polymerase chain reaction (RT-PCR), respectively. Serum TNF-alpha was analyzed by ELISA. Endotoxin enhanced NF-KB activity and TNF-alpha. expression in the intestine, liver, and lung and it increased TNF-alpha, concentration in serum. Ketamine dosages 0.5 mg/kg suppressed the endotoxin-induced NF-KB activation and TNF-alpha expression in the intestine. The lowest dose to inhibit NF-KB activity and TNF-alpha expression in the lung was 5 mg/kg. Ketamine did not inhibit endotoxin-induced NF-KB activity or TNF-a expression in the liver; ketamine itself at a dose of 50 mg/kg enhanced NF-KB activity and TNF-alpha expression in the liver. Ketamine dosage 0.5 mg/kg inhibited endoto xin-induced TNF-alpha elevation in the serum. In conclusion, ketamine can suppress the induction of NF-KB and TNF-alpha by endotoxin in vivo. Subanesthetic dosages of ketamine have an anti-inflammatory effect, but large dosages may be harmful.

Methods Forty adult male SD rats weighing 200-250 g were randomly divided into 4 groups: (Ⅰ)control group(n=10); (Ⅱ) endotoxin group received intravenous endotoxin(Escherichia coli O111: B4, Sigma) 5 mg·kg~(-1)(n=10); (Ⅲ, Ⅳ)endotexin+ketamine group received ketamine or 5 or 10mg·kg~(-1)·h~(-1) after endotoxin(n=10). The animals were anesthetized with urethane i.p. (1g·kg~(-1)). Carotid artery was cannulated for BP and HR monitoring and jugular vein was cannulated for fluid or drug administration. Two hours after endotoxin administration the animals were sacrificed by exsanguination. Blood was collected and peripheral blood monocytes(PBMC) were isolated. NF-κB activity in PBMC was measured by EMSA and plasma TNF-α and IL-6 levels were determined by ELISA. Results Progressive hypotension and tachycardia developed after endotoxin administration. Endotexin also increased NF-κB activity in PBMCs and plasma TNF-α and IL-6. Ketamine 10 mg·kg~(-1) attenuated the endotexin-induced hemedynamie levels. Ketamine(5, 50 mg·kg~(-1)·h~(-1)) suppressed NF-κB activity in PBMC and inhihited plasma TNF-α level but plasma IL-6 level was not affected. Conclusion Ketamine can suppress endotoxin-induced NF-κappa B activation. Subanesthetic dose of ketamine has anti-inflammatory action.

Abstract:Ketamine has been advocated for anesthesia in endotoxemic and other severely ill patients because it is a cardiovascular stimulant. However, ketamine also suppresses serum levels of endotoxin-induced tumor necrosis factor-alpha, and reduces mortality in mice in endotoxin shock. Our study was designed to investigate the protective effect of ketamine on the endotoxin-induced proinflammatory cytokines and nuclear factor kappa B (NF-kappaB) activation in vivo.Adult male Wistar rats were randomly divided into six groups: saline controls; rats challenged with endotoxin (5 mg kg(-1)) and treated with saline; challenged with endotoxin (5 mg kg(-1)) and treated with ketamine (0.5 mg kg(-1)); challenged with endotoxin (5 mg kg(-1)) and treated with ketamine (5 mg kg(-1)); challenged with endotoxin (5 mg kg(-1)) and treated with ketamine (50 mg kg(-1)); and saline injected and treated with ketamine (50 mg kg(-1)). TNF-alpha, IL-6 and NF-kappaB were investigated in the tissues of the intestine (jejunum) after 1, 4 and 6 h.Endotoxin caused transient production of TNF-alpha and IL-6 and activation of NF-kappaB in the intestine at peak times of 1, 4 and 1 h, respectively. Ketamine 0.5 mg kg(-1) suppressed endotoxin-induced TNF-alpha elevation and inhibited NF-kappaB activation in the intestine; a dose of 5 mg kg(-1) was required to inhibit IL-6.Ketamine suppresses the production of proinflammatory cytokines such as TNF-alpha and IL-6 in the intestine, possibly via inhibition of NF-kappaB.
Effects of Low Doses of Ketamine on Pyramidal Neurons in Rat Prefrontal Cortex.

Abstract:Indirect evidence suggests that low doses of ketamine disinhibit (excite) pyramidal neurons in the prefrontal cortex (PFC). In this study, we directly examined the effect of ketamine on PFC pyramidal neurons using simultaneous single-cell and local-field-potential (LFP) recording in chloral hydrate-anesthetized rats. In all animals studied, PFC LFPs showed oscillations (0.3-1.5 Hz) between the active UP state and the relatively quiescent DOWN state, and pyramidal neurons fired preferentially during the UP state. Ketamine (1.25-20 mg/kg, i.v.) inhibited 80% of cells tested and consistently shifted PFC LFPs toward the DOWN state. The inhibitory effect of ketamine was mimicked by MK801, but not by the NR2B-selective NMDA receptor antagonist Ro25-6981. It was not blocked by the dopamine receptor antagonist fluphenazine, the GABAA receptor antagonist picrotoxinin, or the GABAB receptor antagonist CGP46381. These results are consistent with the high density of NMDA receptors expressed on PFC pyramidal neurons and our previous studies showing that blockade of NMDA receptors by ketamine inhibits dendritic NMDA receptor-mediated bursting in PFC pyramidal neurons. Thus, in addition to the previously proposed disinhibitory effect mediated through PFC interneurons, our data suggest that ketamine has an inhibitory effect on PFC pyramidal neurons. Our evidence further suggests that the effect is mediated through non-NR2B-containing NMDA receptors, independent of ketamine's effect on dopamine and GABA transmission. Further understanding of the two opposing effects of ketamine on PFC pyramidal neurons may provide important new insights into its mechanism of action.
Effects of Ketamine on Lipopolysaccharide-Induced Depressive-Like Behavior and the Expression of Inflammatory Cytokines in the Rat Prefrontal Cortex.

Abstract:The present study was designed to investigate the effects of ketamine on lipopolysaccharide (LPS)-induced depressive-like behavior and the expression of inflammatory cytokines in the rat prefrontal cortex. Thirty male Wistar rats were randomly divided into 3 groups (n=10): saline group (S group), LPS only group (L group) and LPS plus ketamine group (LK group). A forced swimming test (FST) was performed. On the first day, rats were placed into water for 15 min. Twenty-four hours later, rats were treated again as in the first test for a 5 min session, and the immobility time was recorded. The prefrontal cortex was harvested for the determination of the interleukin (IL)‑1β, IL-6 and IL-10 levels. Compared with the S group, rats in the L group had significantly increased immobility time in the FST and expression of IL-1β and IL-6, and significantly decreased expression of IL-10 in the prefrontal cortex (P<0.05). However, rats in the LK group had significantly decreased immobility times in the FST and expression of IL-1β and IL-6, and significantly increased expression of IL-10 in the prefrontal cortex compared with the L group (P<0.05). Ketamine can alleviate LPS-induced depressive-like behavior, and its effect is likely associated with changes in the expression of inflammatory cytokines in the rat prefrontal cortex.
Extrasynaptic CaMKIIα is Involved in the Antidepressant Effects of Ketamine by Downregulating GluN2B Receptors in an LPS-induced Depression Model

Abstract:Background A subanesthetic dose of ketamine provides rapid and effective antidepressant effects, but the molecular mechanism remains elusive. It has been reported that overactivation of extrasynaptic GluN2B receptors is associated with the antidepressant effects of ketamine and the interaction between GluN2B and calcium/calmodulin-dependent protein kinase IIα (CaMKIIα) is important for GluN2B localization and activity. Here, we tested whether changes of CaMKIIα and GluN2B are involved in the antidepressant effects of ketamine. Methods Lipopolysaccharide (LPS) was injected intraperitoneally (i.p.) into male C57BL/6 mice. For the interventional study, mice were administrated with ketamine (10 mg/kg, i.p.) or a CaMKIIα inhibitor KN93. Behavioral alterations were evaluated by open-field, novelty-suppressed feeding, and forced-swimming tests. Physiological functions were evaluated by the body weight and fur coat state of mice. The levels of p-CaMKIIα, CaMKIIα, p-GluN2B, GluN2B, p-CREB, CREB, BDNF, GluR1, and GluR2 in the hippocampus were detected by western blotting. The interaction between GluN2B and CaMKIIα was studied using immunoprecipitation assay and small interfering RNA (siRNA) assays. The colocalizations of GluN2B/PSD95 and p-GluN2B/PSD95 were detected by immunofluorescence. The long-term potentiation (LTP) in SC-CA1 of the hippocampus was detected by electrophysiology. Results LPS injection induced depression-like behaviors, which were accompanied by significant increases in extrasynaptic p-CaMKIIα expression, extrasynaptic GluN2B localization, and phosphorylation and decreases in p-CREB, BDNF, and GluR1 expressions and LTP impairment. These changes were prevented by ketamine administration. Immunoprecipitation assay revealed that LPS induced an increase in the p-CaMKIIα–GluN2B interaction, which was attenuated by ketamine administration. SiRNA assay revealed that CaMKIIα knockdown reduced the level and number of clusters of GluN2B in the cultured hippocampal neurons. KN93 administration also reduced extrasynaptic p-CaMKIIα expression, extrasynaptic GluN2B localization, and phosphorylation and exerted antidepressant effects. Conclusion These results indicate that extrasynaptic CaMKIIα plays a key role in the cellular mechanism of ketamine’s antidepressant effect and it is related to the downregulation of extrasynaptic GluN2B localization and phosphorylation.

Abstract:About 16% of the world's population has major depressive disorder. Traditional antidepressants have slow effect rates and low response rates. Many studies have shown that low doses of ketamine can produce rapid and effective antidepressant effects. However, its mechanism of action needs further exploration. Lipopolysaccharide (LPS) was used to establish a depression model in rats and PC12 nerve cells were used for in vitro experiments. (2,4)-Dimethoxybenzylidene anabaseine dihydrochloride (GTS-21), a specific agonist of alpha 7 nicotinic acetylcholine receptors (alpha 7 nAChRs), was used to compare the rapid antidepressant effect of ketamine. Different doses of alpha 7 nAChR antagonist methyllycaconatine (MLA) and alpha 7 nAChR-siRNA were used to interfere with the protective effects of ketamine on neuroinflammation in rats and PC12 cells, respectively. MLA intervention downregulated the anti-inflammatory effects of ketamine and decreased the effects of ketamine on behavior, synaptic plasticity, and Nissl bodies in the neuronal cells. Moreover, the dose of MLA was positively correlated with the inhibitory effect in rat hippocampi and the protective effects of GTS-21 were consistent with ketamine. These results demonstrated that low-dose ketamine could produce neuroprotective effects by activating the alpha 7 nAChR-mediated cholinergic anti-inflammatory pathway (CAP) in depression, resulting in a rapid antidepressant effect.
Role of NF-κB in ketamine-induced apoptosis in rat cortical neurons in vitro

Abstract:Objective To investigate the role of NF-κB in ketamine-induced apoptosis in rat cortical neurons in vitro. Methods Cortical neurons were isolated from newborn pathogen-free SD rats according to the method described by Kimney. After being cultured for 6 days the cortical neurons were randomly divided into 3 groups:group Ⅰ control (C); group Ⅱ ketamine (K) and group Ⅲ ketamine + SN50 (NF-κB translocation inhibitor) (KS). The cortical neurons were exposed to ketamine l mmol/L and ketamine 1 mmol/L + SN502.5 μmol/L for 24 h in group K and KS respectively. Twenty-four hours after the drugs were washed out, NF-κB activity, the expression of Bcl-XL and Bax and apoptosis in the neurons were measured. The ultrastructure of the neurons were examined by electron microscopy. Results NF-κB activity and the percentage of apoptotic neurons were significantly higher and Bcl-XL/Bax ratio was significantly lower in group K than in group C. There was no significant difference in NF-κB activity, Bcl-XL/Bax ratio and percentage of apoptotic neurons between group C and KS. Conclusion NF-κB is involved in ketamine-induced apoptosis in contical neurons in vitro.
Antidepressant Actions of Ketamine: Potential Role of L-Type Calcium Channels

Abstract:Recently, the United States Food and Drug Administration approved esketamine, the S-enantiomer of ketamine, as a fast-acting therapeutic drug for treatment-resistant depression. Although ketamine is known as an N-methyl-d-aspartate (NMDA) receptor antagonist, the underlying mechanisms of how it elicits an antidepressant effect, specifically at subanesthetic doses, are not clear and remain an advancing field of research interest. On the other hand, high-dose (more than the anesthetic dose) ketamine-induced neurotoxicity in animal models has been reported. There has been progress in understanding the potential pathways involved in ketamine-induced antidepressant effects, some of which include NMDA-receptor antagonism, modulation of voltage-gated calcium channels, and brain-derived neurotrophic factor (BDNF) signaling. Often these pathways have been shown to be linked. Voltage-gated L-type calcium channels have been shown to mediate the rapid-acting antidepressant effects of ketamine, especially involving induction of BDNF synthesis downstream, while BDNF deficiency decreases the expression of L-type calcium channels. This review focuses on the reported studies linking ketamine's rapid-acting antidepressant actions to L-type calcium channels with an objective to present a perspective on the importance of the modulation of intracellular calcium in mediating the effects of subanesthetic (antidepressant) versus high-dose ketamine (anesthetic and potential neurotoxicant), the latter having the ability to reduce intracellular calcium by blocking the calcium-permeable NMDA receptors, which is implicated in potential neurotoxicity.
Effects of Ketamine on Proinflammatory Cytokines and Nuclear Factor Kappab in Polymicrobial Sepsis Rats

Abstract:AIM: To explore the effects of ketamine on hemodynamics, plasma proinflammatory cytokine (TNF-alpha and IL-6) levels and nuclear factor kappa B (NF-kappa B) activation during polymicrobial sepsis.METHODS: Male Sprague-Dawlay rats were subjected to cecal ligation and puncture (CLP) or sham operation. The rats were randomly assigned into four equal groups: sham CLP group, CLP group, ketamine (KT) I group and KT II group. Thirty minutes before CLP, ketamine (5 mg/kg per hour and 10 mg/kg per hour, respectively) was infused continuously through the left femoral vein cannula in KT I group or KT II group. Sham CLP group and CLP group received 0.9% saline only (5 mL/kg per hour). The right femoral artery was cannulated to monitor mean arterial pressure (MAP) and heart rates (HR),and draw blood samples. The proinflammatory cytokine (TNF-alpha and IL-6) levels of plasma were measured using enzyme-linked immunosorbent assays (ELISA). The hepatic NF-kappa B activation was determined by Western blot and HPIAS 2000 image analysis system. Twenty hours after CLP, the rats were killed by right femoral artery phlebotomization.RESULTS: CLP produced progressive hypotension, and a first increase followed by a decrease in HR. The hypotension was prevented, and the HR was slightly steady in ketamine treated rats. TNF-alpha levels of plasma reached a peak value at 2 h after CLP. Ketamine (KT I group or KT II group) caused a significant decrease compared with CLP group at 2, 5 and 9 h time points after CLP (14.3 +/- 1.9 vs 4.3 +/- 0.9, 9.7 +/- 1.4 vs 4.3 0.9; 9.3 +/- 1.5 vs 4.3 +/- 0.9, 8.7 +/- 1.4 vs 4.3 +/- 0.9; 6.0 +/- 1.5 vs 5.0 +/- 1.7, 5.3 +/- 0.8 vs 5.0 +/- 1.7; P < 0.01, respectively). The IL-6 levels of plasma firstly ascended and then descended in CLP group, and reached a peak value at 9 h after CLP. Ketamine (KT I group or KT II group) caused a significant decrease compared with CLP group at 5, 9 or 20 h after CLP (135.0 +/- 52.6 vs 60.0 +/- 16.3, 112.5 +/- 52.6 vs 60.0 +/- 16.3; 410.0 +/- 68.7 vs 62.5 +/- 12.5, 250.0 +/- 28.0 vs 62.5 +/- 12.5; 320.0 +/- 25.9 vs 52.5 +/- 10.1, 215.0 +/- 44.6 vs 52.5 +/- 10.1; P < 0.05, respectively). The IL-6 levels of plasma in KT II group were lower than those of KT I group at 9 h after CLP (250.0 28.0 vs 410.0 68.7; P < 0.05). In addition, CLP increased hepatic NF-kappa B expression compared with sham CLP. Ketamine suppressed NF-kappa B activation in a dose-dependent manner at 4 h after CLP (237.7 +/- 3.5 vs 246.9 +/- 3.1; P < 0.05).CONCLUSION: Ketamine stabilizes the hemodynamics, attenuates the proinflammatory cytokine responses, and inhibits hepatic NF-kappa B activation. Ketamine May Exert Antidepressant Effects Via Suppressing NLRP3 Inflammasome to Upregulate AMPA Receptors.

Abstract:Recent studies have indicated that sub-anesthetic dose of ketamine exerts rapid antidepressant effects. Upregulation of α-amino-3-hydroxy-5-methylisoxazole-4-propionic acid (AMPA) receptors contributes to rapid antidepressant effects of ketamine. A recent study indicated that the increasing expression of AMPARs is related to the inhibition of NLRP3 inflammasome. Thus, we postulated that NLRP3 inflammasome might play an important role in ketamine's antidepressant effects. We found that sub-anesthesia dose of ketamine (10 ml/kg) ameliorated LPS-induced depressive-like behaviors, including decreased sucrose preference and increased immobility times in the tail suspension test. Ketamine also abrogated LPS-induced over expression of IL-1β and NLRP3 and reversed LPS-induced down-regulation of AMPA GluA1 subunits in hippocampi. The selective NLRP3 inflammasome inhibitor Ac-YVAD-CMK exhibited similar anti-inflammatory and antidepressant effects like ketamine. Combination of ketamine and Ac-YVAD-CMK showed no enhanced anti-inflammatory and antidepressant effects than ketamine or Ac-YVAD-CMK administration alone. These results indicated that the NLRP3 inflammasome might be an important mediator, through which ketamine could regulate AMPA receptors to exert rapid antidepressant effects.
Effect of Ketamine on High-voltage-activated Calcium Currents in Rat Hippocampal Neurons

Abstract:Objective: To investigate the effect of ketamine on the high-voltage-activated calcium currents (ICa(HVA)) in rat hippocampal neurons. Methods: Neurons were cultured from Wistar rat hippocampus. I Ca(HVA) was recorded using whole-cell patch clamp technique. After application with ketamine at different concentrations (10, 30, 100, 300, and 1 000 μmol/L), the effect of ketamine on ICa(HVA) was evaluated. Results: ICa(HVA) was inhibited by ketamine in a concentration- dependent manner. Ketamine at 10 μmol/L showed no effect on I Ca(HVA). Four concentrations of ketamine (30, 100, 300, and 1 000 μmol/L) reduced the peak ICa(HVA) currents by (17.5 ± 4.5)%, (25.5 ± 6.9)%, (38.5 ± 4.1)%, and (42.3 ± 4.6)% respectively, with a mean half maximal inhibitory concentration of 68.2 μmol/L and Hill coefficient of 0.47. The maximal activation membrane potential was shifted to (5.3 ± 0.8) from (5.4 ± 0.9). The half maximal activation membrane potential of inactivation curve was shifted from (-26.7 ± 3.9) mV to (-32.8 ± 4.2) mV. Conclusion: Ketamine can remarkably inhibit calcium currents in the central neurons, which may explain at least partly the action of ketamine on central nervous system.
Acute Ketamine Administration Attenuates Lipopolysaccharide-Induced Depressive-Like Behavior by Reversing Abnormal Regional Homogeneity in the Nucleus Accumbens

Abstract:Ketamine has rapid antidepressant effects, but no study to date has investigated changes in resting-state brain activity following ketamine administration in inflammation-induced depression. The purpose of this study was to use blood oxygen level-dependent functional MRI to explore changes in the resting-state brain activity in a rat model of depression induced by lipopolysaccharide (LPS, 1mg/ kg) challenge and to examine whether acute ketamine administration can reverse LPS-induced depressive-like behavior. Here, we showed LPS-induced depressive-like behavior as evidenced by significantly reduced motility in the forced swim test. In addition, LPS-induced increases in plasma levels of proinflammatory cytokines were not completely reversed by acute ketamine administration, suggesting that ketamine exerts its antidepressant effects independently of a possible interference with LPS-induced inflammatory signaling. However, increased regional homogeneity (ReHo) was observed in some brain regions of LPS-exposed animals, including bilateral caudate putamen and nucleus accumbens, which were parts of the mood-regulating circuit. Moreover, ReHo values of bilateral caudate putamen and nucleus accumbens showed significant positive correlations with immobility time. Notably, the LPS- induced depressive- like behavior and increase of ReHo value in the right nucleus accumbens was reversed by acute ketamine administration. In summary, this study suggests that acute ketamine administration is capable of attenuating LPS- induced depressive- like behavior, at least partially by reversing abnormal ReHo in the right nucleus Copyright (c) 2019 Wolters Kluwer Health, Inc.
