Comparison of Learning and Memory of APPswe/PS1dE9 Double Transgenic Mice Model of Alzheimer’s Disease and Wild-type Mice

Abstract:To compare learning and memory of APPswe/PS1dE9 double transgenic mice of Alzheimer’s disease(AD) and wild-type mice at the stage of different age. Methods: Female C57BL/6J mice were mated with male C3H/HeJ mice and the first filial generation(F1) were gotton. Then F1 mice were mated with 3 month-old APPswe/PS1dE9 double transgenic mice to get F2. DNA of each pup were extracted from the tails of F2 mice and amplified by polymerase chain reaction (PCR). According to the results of PCR, F2 mice were classified into transgenic group and wild-type group. Learning and memory were assessed by Morris water maze at the age of 6 or 8 months respectively and by Y-maze at 11 months. Results: Eighteen APPswe/ PS1dE9 double transgenic mice were identified from thirty three pups. At 6months, no significant statistical difference was found about the mean latency in place navigation test of APPswe/PS1dE9 double transgenic mice and wild-type mice, nor were the percentage of time and distance spent in the target quadrant in probe trials(P>0.05). At 8 months, compared with the wild-type, the mean latency in place navigation test of APPswe/PS1dE9 double transgenic mice was prolonged in the first day (P<0.05). At 11 months, the time of training to learn standard of APPswe/PS1dE9 double transgenic mice increased(P<0.05). Conclusion: APPswe/PS1dE9 double transgenic mice had dysfunction of learning and memory about at the age of 11 months.
