Glycan Profiling Changes of Peripheral Regulatory T Cells in Patients with Hepatocellular Carcinoma

Abstract:Objective:To assess the changes of glycosylation profiling of characteristic glycan structures of peripheral regulatory T cells(T reg) in hepatocellular carcinoma(HCC) patients.Methods:The 4-color flow cytometry was used to sort CD4+CD25+CD127low/-CD49d-T cells from peripheral blood mononuclear cells(PBMCs) in HCC patients(n=32) and healthy controls(n=10).Lectin microarray was used to analyze the extracted and labeled membrane protein of T reg.Results: The α-1,6-mannose,α-1,6-fucose,GalNAc,β-galactose and complex N-glycan were elevated on the surface of T reg cell membrane from HCC patients.Conclusions: The changes of T reg surface glycan profiling in HCC patients may be related with suppressive activity and may provide a novel target for HCC immunotherapy.
