Abstract:Seven new quinoxaline derivatives 3a-3g were synthesized through two steps convenient approach. Their structures were confirmed by Fourier transform infrared, proton nuclear magnetic resonance, and high-resolution mass spectrometry. The synthesized compounds 3a-3g showed good fluorescent intensity in dimethyl sulfoxide. They were also shown to possess good antifungal activities. The structure-activity relationship was studied by density functional theory calculations.

Abstract:We report herein the design and synthesis of novel 7-(3-alkoxyimino-5-amino/methylaminopiperidin-1-yl)fluoroquinolone derivatives based on the structures of new fluoroquinolones IMB and DZH. The antibacterial activity of these newly synthesized compounds was also evaluated and compared with gemifloxacin, ciprofloxacin, and levofloxacin. Results revealed that all of the target compounds 10-27 have good potency in inhibiting the growth of Staphylococcus aureus including MSSA (MIC: 0.125-8 μg/mL), Staphylococcus epidermidis including MRSE (MIC: 0.25-16 μg/mL), Streptococcus pneumoniae (MIC: 0.125-4 μg/mL), and Escherichia coli (MIC: 0.25-0.5 μg/mL). In particular, some compounds showed useful activity against several fluoroquinolone-resistant strains, and the most active compound 15 was found to be 16-128, 2-32, and 4-8-fold more potent than the three reference drugs against fluoroquinolone-resistant MSSA, MRSA, and MRSE.

Abstract:A series of novel 7-(3-alkoxyimino-4-amino-4-methylpiperidin-1-yl)fluoroquinolone derivatives were designed, synthesized and evaluated for their in vitro antibacterial activity and cytotoxicity. All of the target compounds have potent antibacterial activity against the tested Gram-positive and Gram-negative strains, and exhibit good potency in inhibiting the growth of Staphylococcus aureus including MRSA, Staphylococcus epidermidis including MRSE and Streptococcus pneumoniae (MICs: 0.125-4 μg/mL). Compound 22, with the best activity against Gram-positive strains, is 4-16 fold more potent than gemifloxacin, gatifloxacin and levofloxacin against Enterococcus faecalis, and 16- and 4-fold more potent than levofloxacin against S. epidermidis 09-6 and S. pneumoniae 08-4, respectively.

Abstract:We report herein the design and synthesis of novel 7-(4-alkoxyimino-3-aminomethylpiperidin-1-yl) fluoroquinolone derivatives. The antibacterial activity of the newly synthesized compounds was evaluated and compared with gemifloxacin, levofloxacin and ciprofloxacin. Results reveal that compounds 10, 16, and 17 have good activity against all of the tested Gram-positive organisms including drug-resistance strains (MICs: 0.125-4 mu g/mL). In addition, compounds 16 and 17 (MICs: 4 mu g/mL) were 2- to 8-fold more potent than the reference drugs against Pseudomonas aeruginosa. (C) 2011 Elsevier Ltd. All rights reserved.

Abstract:A series of novel 7-(3-alkoxyimino-4-methyl-4-methylaminopiperidin-1-yl)fluoroquinolone derivatives were designed, synthesized, and characterized by H-1-NMR, MS, and HRMS. These fluoroquinolones were evaluated for their in-vitro antibacterial activity against representative Gram-positive and Gram-negative strains. Generally, all of the target compounds have considerable antibacterial activity against the tested forty strains, and exhibit exceptional potency in inhibiting the growth of methicillin-sensitive Staphylococcus aureus (MSSA) and methicillin-resistant S. aureus (MRSA) ATCC33591 (MICs: 0.06 to 2 mu g/mL). In particular, compounds 14, 19, 28, and 29 are fourfold more potent than ciprofloxacin against MSSA 08-49. Compounds 23, 26, and 27 are twofold more potent than ciprofloxacin against MRSA ATCC33591 and MSSA ATCC29213. In addition, compound 14 exhibits excellent activity (MIC: 0.06 mu g/mL) against Acinetobacter calcoaceticus, which is two- to 16-fold more potent than the reference drugs gemifloxacin, levofloxacin, and ciprofloxacin.

Abstract:1-cyclopropyl-7-(S,S)-2,8-diazabicyclo4.3.0non-8-yl-6-fluoro-8-difloro-methoxy-1,4-dihydro-4-oxo-3-quinoline carboxylic acid was prepared and evaluated for antibacterial activity in vitro and in vivo. The compound had better activity than ciprofloxacin in vitro and has similar activity as or better activity than moxifloxacin in vitro and in vivo. Especially against S.pneumoniae 29074, the compound and moxifloxacin have i.v. ED50 of 1.84 and 3.67mg/kg respectively. Consequently, the compound was selected as candidate for further evaluation.

Abstract:AIM:To design and synthesize 1-(5-fluoro-2-pyridyl) quinolone derivatives, and to study their in vitro antibacterial activities.METHODS:Eight new compounds of 5-amino-6,8-difluoro-1-(5-fluoro-2-pyridyl)-7-(3-methyl-1-piperazinyl)-1, 4-dihydro-4-oxo-3-quinolinecarboxylic acid and its analogues were synthesized from ethyl 6-nitro-2, 3, 4, 5-terafluorobenzoylacetate and ethyl 3-methoxy-2, 4, 5-trifluorobenzoylacetate through 5 or 6 steps.RESULTS:Fifteen new compounds (1-15) were obtained including 8 desired compounds (8-15). The structures of the compounds were determined by 1HNMR, MS.CONCLUSION:In vitro antibacterial activities of the compounds (8-15) against Staphylococcus aureus-16, Escherichia coli-26 and Pseudomonas aeruginosa-17 were lower than control of ciprofloxacin.

Abstract:A series of 1-substituted-6-fluoro-7-(1-(4-((Z)-2-(2-aminothiazol-4-yl)-2-methoxyi-minoacetyl)piperazinyl))-1,4-dihydro-4-oxo-3-quinoline carboxylic acid has been prepared and evaluated for antibacterial activity. These compounds were prepared by the combination of 1-substituted- 6-fluoro-7- piperazinyl- 1,4-dihydro-4-oxo-3-quinolinecarboxylic acid and S-2-benzo- thiazoyl-(Z)-2-(2-aminothiazol-4-yl)-2-methoxyimino acetate(MAEM) under the condition of Schotten-Baumann. The title compounds are confirmed with NMR,UV, IR, FAB-MS, et al.

Abstract:To explore new agents of quinolone derivatives with high antibacterial activity, 7-(4-alkoxyimino-3-methyl-3-methylaminopiperidin-1-yl)quinolones were designed and synthesized, and their activity against gram-positive and gram-negative strains was tested in vitro. Sixteen target compounds were obtained. Their structures were established by 1H NMR, HRMS and X-ray crystallographic analysis. Compounds 14k and 14m-14o show good antibacterial activity against the tested five gram-positive strains and five gram-negative strains (MIC: 0.25-16 micromg x mL(-1)), of which the most active compound 14o is 8-fold more potent than levofloxacin against S. pneumoniae (MIC: 4 microg x mL(-1)), and comparable to levofloxacin against S. aureus, S. epidermidis, E. faecalis and E. coli (MIC: 0.25-1 microg x mL(-1)), but generally less potent than gemifloxacin.

Abstract:AIM:To find new antibacterial agents of quinolone with high activity and low toxicity.METHODS:To design and synthesize 7-(7-aminomethyl-5-azaspiro [2,4] hept-5-yl)-1-cyclopropyl-6-fluoro-8-methoxy-1,4-dihydro-4-oxo-3-quinolinecarboxylic acid and its analogues, and to study their antibacterial activity in vitro and in vivo.RESULTS:Twenty new compounds (2 - 11, 17 - 26) were obtained including five targeted compounds (22 - 26). The structures of the compounds were confirmed by 1HNMR, MS and HRMS. Compounds 22 - 26 showed broad spectrum of antibacterial activity against Gram-positive and Gram-negative organisms. Especially for compound 24, the relevant MIC values for 13 strains of Gram-positive organisms were < 0.001 - 0.03 mg(-1), including 4 strains of S. pneumoniae, 2 strains of S. pyogenes, 3 strains of S. aureus and 2 strains of Enterococci which exhibited more potent activity than contrast agents (clinafloxacin and gatifloxacin). The MIC values of 24 for 6 strains Gram-positive organisms were 0.01 - 1 mg x L(-1), which exhibited equal or lower activity than contrast agents. They were more effective than ciprofloxacin and gatifloxacin against intraperitoneal infections caused by S. pneumoniae and S. aureus in mice.CONCLUSION:Compounds (23, 24 and 26) showed excellent antibacterial activity in vitro and in vivo and should be worth further investigation.

Abstract:In order to find new agents of quinolones with high and broad - spectrum antibacterial activities and low toxicity, we have designed and synthesized nine compounds of 7-(7-methyl aminomethyl-5-azaspiro[2, 4] hept-5-yl)-1-cyclopropyl-6-fluoro-1, 4-dihydro-4-oxo-1, 8-naphthyridine-3-carboxylic acid and its analogues. The structures of the compounds were determined by 1H - NMR, MS. Their antibacterial activity in vitro have been tested. The results indicated that the new compounds showed a broad spectrum of antibacterial activity against Gram - positive and Gram - negative organisms. The MIC values of compound 23,26,27 for 10 strains of Gram - positive organisms were 0.01-0.12μg/ml, which exhibited more potent activity than reference agents gatifloxacin (0.12-1μg/ml) and ciprofloxacin (0.25-4μg/ml). The MIC values of them for 10 Gram - negative strains were 0.01-2μg/ml, which exhibited equal activity of reference agents.

Abstract:Objective To find new antibacterial agents of quinolones with high activity. Methods Seven fluoroquinolone derivatives with more water solubility substituent at C-7 position were designed, synthesized and evaluated for their in vitro activity against seventeen Gram-positive and thirteen Gram-negative strains. Results The activity of compound 10(MIC: 0.06~4μg/mL) was comparable to levofloxacin and gemifloxacin against S. aureus including MRSA and S. epidermidis including MRSE. Compound 1(MIC: 0.25μg/mL) is 32~128 fold more potent than both the reference drugs against S. pneumoniae 08-2, and compound 12(MIC: 1μg/mL) is 16 and 4 fold more potent than levofloxacin and gemifloxacin against both K. pneumoniae 09-22 and 09-23, respectively. Conclusion The water solubility of the substituent at C-7 position is not main parameter affecting antibacterial activity.

Abstract:To explore new agents of quinolone derivatives with high activity against Gram-positive and Gram-negative microorganisms, 7-(3-amino-4-alkoxyimino-1-piperidyl) quinolones were designed and synthesized, and their activity against Gram-positive and Gram- negative microorganisms were tested in vivo and in vitro. Twenty one target compounds were obtained. Their structures were established by 1H NMR, HRMS and X-ray crystallographic analysis. The target compounds possess different antimicrobial activities against both Gram-negative and Gram-positive microorganisms. Compounds 14a and 14m have broad spectral antibacterial activities. They show better antibacterial activities against 12 strains Gram-positive bacteria than three references. In particular, their activities against S. aureus and S. epidermidis (including MRSA and MRSE) were 4 - 16 times than that of gemifloxacin and balofloxacin, and 8 - 64 times than that of levofloxacin. The MIC values to S. aureus strains of compounds 14a and 14m were 0.25 - 1 mg x L(-1) and 0.125 - 1 mg x L(-1), to S. epidermidis strains were 0.5 - 4 mg x L(-1) and 1 - 8 mg x L(-1) respectively. The in vivo results showed that they have as good internal protection as gemifloxacin and moxifloxacin against systemic infection model in mice (P > 0.05).

Abstract:A series of gatifloxacin, ciprofloxacin, and 8-OCH3 ciprofloxacin coumarin derivatives with remarkable improvement in lipophilicity as compared to the parent fluoroquinolones was designed, synthesized, and characterized by 1H-NMR, MS, and HRMS. These derivatives were evaluated for their in-vitro activity against Mycobacterium smegmatis CMCC 93202 and MTB H37Rv ATCC 27294. All of the synthesized compounds were less active than the parent compounds against M. smegmatis CMCC 93202, but the activity of compound 6 was found to be 28-fold more potent than ciprofloxacin, 8-OCH3 ciprofloxacin, moxifloxacin, and rifampin, and comparable to gatifloxacin against MTB H37Rv ATCC 27294. These results indicated that the lipophilicity of the tested compounds is not the sole parameter affecting antimycobacterial activity.

Abstract:AIM:To explore new agents of quinolone derivatives with high activity against Gram-positive organisms.METHODS:dl-7-(4,4-Dimethyl-3- aminomethylpyrrolidinyl)-quinolones were designed and synthesized, and their activity against Gram-positive organisms was tested in vitro.RESULTS:Ten target compounds were obtained. The structures of these compounds were confirmed by 1H NMR, MS. The target compounds with dl-4,4-dimethyl-3-( methyl) aminomethylpyrrolidine side chains had high activity against Gram-positive organisms. Especially the MIC values of compound 22 for 4 strains of Gram-positive resistant bacteria (two strains of MRSA and two of MRSE) were 0.015 -0.5 mg x L(-), which exhibited more potent activities than gatifloxacin (4 - 128 times). Its MIC value for Pseudomonas aeruginosa 03-5 (0.008 mg x L(-1)) was 4 times as that of gatifloxacin (0.03 mg x L(-1)).CONCLUSION:The compound 22 showed high activity against Gram-positive organisms in vitro and it is worth of more investigation.
