Preliminary Study of the Cerebral Myelin Injury in Rats with Acute Necrotizing Pancreatitis

Abstract:Objective To investigate the changes of cerebrospinal fluid (CSF) and serum myelin basic protein (MBP) level in an experimental rat model with acute necrotizing pancreatitis (ANP). Methods The model with ANP was induced in SD rats, acute edematous pancreatitis (AEP) and sham-operated rats were used as controls. CSF and blood samples were obtained 3, 6, 9, 12 and 24 hours after the introduction of experimental models, and were analyzed for MBP concentration by using an ELISA kit. The cerebral white matter was examined by transmission electron microscope. Results The kinetic CSF MBP concentration of ANP rats showed a rapid uprise at 3 h (P0.01), and remained at a high level at 6 and 9 h after introduction(P0.05), thus showing a significant difference compared with the levels observed in the AEP or sham-operated rats; then followed by a slow decline after 12 h (P0.05). The concentrations of serum MBP in the ANP, AEP and sham-operated rats did not show any significant differences and remained at lower levels. In the ANP group, slight or focal chaos, such as vacuolation and debunching, was observed in the myelin sheaths by electron microscopy at 3 h and 9 h. Conclusions The experimental results provide evidence that demyelination occursing in the early stage of ANP. High concentration of MBP in the CSF reflects the demyelination.
