Effects of Porcine Reproductive and Respiratory Syndrome Virus and Porcine Circovirus Type 2 Co-Infection on Numbers of Native Immune Cells in Peripheral Blood of Piglets

Abstract:Six-week-old healthy piglets were infected with porcine reproductive and respiratory syndrome virus(PRRSV) and porcine circovirus type 2(PCV2) respectively and co-infected with PRRSV+PCV2 in order to explore the effects on native immunity of piglets.The effector cells of native immunity in peripheral blood including white blood cells(WBC),granulocytes,monocytes,NK cells and γδ T cells were analyzed using SWC3a single-color flow cytometry(FCM),CD3/CD4/CD8 triple-color FCM and hemogram before infection and on day 3,7 and 10 post-inoculation.The number of WBC,granulocytes,monocytes,NK cells and γδ T cells dramatically decreased in peripheral blood of the inoculated piglets on day 3 and 7 post-infection with PRRSV alone,PCV2 alone and PRRSV+PCV2,and those of PRRSV+PCV2 co-infection exacerbated more significantly than PRRSV or PCV2 infection alone.The results showed that there was a synergistic effect of PRRSV+PCV2 co-infection on the depletion of effector cells of native immunity in peripheral blood of piglets,indicating that PRRSV+PCV2 co-infection could cause more severe immuno-suppressive effects on native immunity in piglets.
