534 Effect of HIV Integrase Inhibitor on HIV Protease Inhibitor-Induced ER Stress and Disregulation of Lipid Metabolism in Hepatocytes

Abstract:age, light and electron microscope examination were performed, as well as rhodanine copper staining and copper quantification by flamed atomic absorption spectroscopy.Hepatic Sadenosylmethionine (SAM), SAH, and serum HCY were measured by HPLC.Hepatic SAH hydrolase and GRP78 mRNA and protein levels were also measured.RESULTS: Hepatic Cu was significantly higher at both ages in the tx-j mice compared to control mice.Consistent with the hypothesis, the SAM:SAH ratio was significantly lower by 1.8 fold (p=0.01) in the tx-j mice at 16 weeks.At 20 weeks, the tx-j mice showed mild microvesicular steatosis and greater hepatic triglyceride than controls (34.4±4.8 vs 27.4±4.1 mg/g, p= 0.03).SAH hydrolase mRNA expression and protein levels were significantly reduced in the tx-j mice at both time points, whereas Cu concentration correlated negatively with both SAH hydrolase mRNA (p= 0.01; r= -0.61) and protein levels (p= 0.01; r= -0.63).With progression of the hepatic histological damage at 20 wk, both plasma HCY and GRP78 expression in liver were increased in the tx-j mice (p<0.01, and p<0.02, respectively), and hepatic triglyceride was correlated with plasma HCY level (p= 0.005; r= 0.66).CONCLUSIONS: Reduced hepatic SAH hydrolase expression was associated with increased SAH and subsequently by increased plasma HCY and hepatic GRP78 expression that was that was paralleled by progression of hepatic damage and steatosis.These findings are consistent with the hypothesis that aberrant methionine metabolism plays an important role in the pathogenesis of hepatic damage in WD.
