The Morphological Changes of the Model with EAE Pre-inj eced the Gap Junction Blocker into the Lateral Cerebroventricle

Abstract:Objective To investigate the changes of OX_(42) and glial fibrillary acidic protein (GFAP) expression and incidence of the experimental autoimmune encephalomyelitis (EAE) after injecting carbenoxolone (CBX) , the gap junction (GJ) blocker, into the lateral cerebroventricle of guinea pigs. Methods 12 guinea pigs were subgrouped randomly by 2 groups. The treatment group was pre-injected CBX into the guinea pig lateral cerebroventricle and the control group were injected physiological saline. Animals of the EAE models were created by foot sole injectings with myelin basic protein. We observed the changes of OX_(12) and GFAP expression and incidence of EAE. Results The EAE incidence apparently decreased in treatment group and EAE symptoms became alleviated. We observed that the number of GFAP expression was 504. 00 ± 55. 88 in treatment group, while in the control group was 497. 00 ± 49. 69. The difference of GFAP-Li expression between two groups was not found (P>0. 05). The number of OX_(42) expression was 120.00±10.64 in the treatment group, which significantly decreased compared with that in the control group (242.00 ± 35.35, P<0.01). Conclusions In guinea pigs pre-treated by CBX, the EAE incidence was significantly decreased and EAE symptoms became alleviated, which the decreasing expression of OX_(42)-Li in microglias could be the chief reason for alleviation of EAE symptoms. So it indicated and suggested that blocking the GJ might be a new route for treating EAE.
