Abstract:Objective To detect the expression of cytokeratin 20 (CK20) in lymph nodes of patients with endometrial carcinoma (EC) with nest RT-PCR method, thus to know something about the micrometastasis (MM). Methods Nest RT-PCR method was used to determine the expression of CK-20 mRNA in the pelvic lymph nodes of 18 patients who were surgically staged and treated for endometrial carcinoma in Peking University People's Hospital from March 2003 to February 2004. The specificity of the mRNA CK-20 marker was examined in lymph nodes obtained from five healthy patients. The Ishikawa cell line and EC tissue were used as positive control. Result All the Ishikawa cell and EC tissue were positive expression of CK20 mRNA, while all the normal lymph nodes were negative expression. For the normal lymph nodes diagnosed by traditional histopathologic method of endometrial carcinoma of stage Ⅰ and stage Ⅱ, there were about 31% positive expression of CK-20 mRNA. Conclusion CK20 is a good marker to detect micrometastasis of endometrial carcinoma.There are about 31% cases of endometrial carcinoma in stage Ⅰ and stage Ⅱ with can be detected of micrometastasis by Nest RT-PCR of CK-20 mRNA, which maybe related to the relapse of endometrial carcinoma in early stage.
