Abstract:The aggregation behaviors and phase separation of binary nanoparticles immersed in semiflexible polymer brushes are explored using molecular dynamics simulations. The conformations of the binary nanoparticle-brush mixtures are very sensitive to the size ratio q = sigma(l)/sigma(s) where sigma(l) and sigma(s) are the diameters for large and small nanoparticles, respectively, and the attractive interactions epsilon(M) between the nanoparticles and polymer brushes. The phase separation of the binary nanoparticles is observed at a relatively strong attractive interaction under the effects of inter-nanoparticle depletion attractions provided by polymer brushes. Furthermore, the crystallization of small nanoparticles occurs at a strong interaction and a hexagonal close-packed-like (hcp-like) ordered structure is formed. The present results may promise a new approach for controlling the self-assembling behavior of nanoparticles and constructing a better macroscopic performance of the material.

Abstract:The surface-induced phase transitions in dense nanoparticle arrays of lamella-forming diblock copolymers are investigated by using the real-space self-consistent field theory. The dense nanoparticle array provides a distinct and strong confinement environment where there are incomplete confinements in three spatial directions. Several complicated phases with cubic symmetries, such as the monocontinuous and bicontinuous phases, are identified in the dense nanoparticle arrays. Through adjusting the strength of surface preference, the disorder phases are induced into the complicated order phases in the nanoparticle arrays with small periods while the order–order transitions occurs in those with large periods. Investigations on the free and entropic energies indicate that these surface-induced phase transitions are of first-order and the stabilities of order phases are intimately correlated to the strength of surface preference. Our results enrich the knowledge about the phase behaviors of macromolecules in confined systems, which may be helpful to fabricate the novel nanomaterials based on the block copolymers.

Abstract:The crystallization-driven self-assembly of polymers based on semicrystalline poly(epsilon-caprolactone) cores is currently an area of high interest on account of their well-known biocompatibility and biodegradability, yet a comprehensive understanding of coil-crystalline-coil type triblock copolymer assembly behavior with respect to this core chemistry is yet to be realized. Herein, we demonstrate the simple preparation of well-defined tuneable 1D and 2D structures based on poly(epsilon-caprolactone) (PCL) triblock copolymers of different block ratios synthesized by ring-opening polymerization (ROP) and reversible addition-fragmentation chain transfer (RAFT) polymerization. In this report, the assembly of PCL-based amphiphiles in various solvents was investigated to tune the morphology and size of the assemblies with well-defined 2D platelets and long cylinders produced when using long soluble coronal blocks or under good solvent conditions. By contrast, truncated short fibers were obtained for less soluble PCL-containing block copolymers or under poor solubility conditions for the core block as a consequence of the increasing amount of nuclei formed in the crystallization process. Furthermore, the length of PCL-based 1D nanostructures could be controlled by tuning self-assembly conditions where the micelles' lengths varied from 93 to 1200 nm with narrow dispersities. This easy assembly methodology greatly simplifies the lengthy procedure required to prepare biodegradable 1D and 2D nanostructures from PCL with tuneable sizes, which demonstrate great potential as drug-delivery vehicles in the realm of biomedicine.
Surface-field-induced Microstructures of Asymmetric Diblock Copolymer Nanoparticles

Abstract:The microstructures of asymmetric diblock copolymer nanoparticles were investigated using real-space self-consistent field theory. The nanoparticle boundary provides a spherical confinement in which the highly deformable copolymers are forced into a closed and crowded space. The surface-field-induced effects on the microstructures were examined systematically, and a rich variety of novel structures, such as mixtures of distorted cylinders and spherical lamellae, was observed in the copolymer nanoparticles. Investigations of the free energies indicate that the surface-field-induced structural transitions are of the first order. The formation mechanism of the confinement-induced structures was reasonably elucidated, based on the interactions between the polymer chains and confinement boundaries, and on the spherical symmetry from confinements. Our theoretical results are in good agreement with available simulation and experimental observations. The asymmetric diblock copolymer nanoparticles show a rich variety of novel structures, such as mixtures of distorted cylinders and spherical lamellae, due to the surface-field-induced effects from the nanoparticle surfaces.
Poly(d,l-lactide-co-glycolide) Dispersions Containing Pluronics: from Particle Preparation to Temperature-Triggered Aggregation

Abstract:In this work the preparation mechanism, properties and temperature-triggered aggregation of poly(D, L-lactide- co-glycolide) (PLGA) dispersions are investigated. The dispersions were prepared by interfacial deposition in aqueous solution containing Pluronic L62 (EO(6)PO(30)EO(6)) or F127NF (EO(101)PO(56)EO(101)), where EO and PO are ethylene oxide and propylene oxide, respectively. PLGA dispersions were also prepared in the absence of added Pluronic for comparison. The PLGA particles were characterized using SEM, photon correlation spectroscopy and electrophoretic mobility measurements. It was found that the hydrodynamic diameter (d) increased with PLGA concentration used in the organic solvent phase ( C PLGA(o) ). The value for d was proportional to C(PLGA)(o) (1/3). The value for d increased upon addition of 0.04 M NaNO(3) which demonstrated the importance of electrostatic interactions during particle formation. Electrophoretic mobility measurements were conducted as a function of pH and the data used to estimate the Pluronic layer thicknesses on the PLGA particles. The layer thickness was greatest for the PLGA particles prepared in the presence of Pluronic F127NF. PLGA dispersions containing Pluronic L62 exhibited temperature-triggered aggregation in the presence of 0.15 M NaNO(3). It was found that the critical temperature for dispersion aggregation (T(crit)) was comparable to the cloud point temperature ( T(cp)) for the parent Pluronic L62 solution. Conditions were established for achieving temperature-triggered aggregation at body temperature for PLGA particle/Pluronic L62 dispersions under physiological ionic strength and pH conditions. The PLGA/Pluronic L62 mixtures studied may have potential for use as injectable biodegradable implants for controlled release applications.

Abstract:Understanding the micellization of amphiphilic triblock copolymers, especially Pluronics can play a persuasive role in engineering "smart" formulations for drug delivery applications. Their underlying self-assembly in the presence of designer solvents such as ionic liquids (ILs) provides combinatorial benefits of unique munificent properties of ILs and copolymers. The complex molecular interactions in the Pluronic copolymers/ILs mixed system influence the aggregation mechanism of copolymers depending on various aspects with no standardized factors to govern the structure-property relationship, which led to the practical applications. Here, we summarized recent progress in understanding the micellization process of IL-Pluronic mixed systems. Special emphasis was given to pure Pluronic systems (i.e., PEO-PPO-PEO) without any structural modifications, such as copolymerization with other functional groups, and ILs having cholinium and imidazolium groups. We expect that the correlation between existing/developing experimental and theoretical studies will provide the necessary basis and impetus for successful utilization in drug delivery applications.

Abstract:Drug-polymer solid dispersion has been demonstrated as a feasible approach to formulate poorly water-soluble drugs in the amorphous form, for the enhancement of dissolution rate and bioperformance. The solubility (for crystalline drug) and miscibility (for amorphous drug) in the polymer are directly related to the stabilization of amorphous drug against crystallization. Therefore, it is important for pharmaceutical scientists to rationally assess solubility and miscibility in order to select the optimal formulation (e.g., polymer type, drug loading, etc.) and recommend storage conditions, with respect to maximizing the physical stability. This commentary attempts to discuss the concepts and implications of the drug-polymer solubility and miscibility on the stabilization of solid dispersions, review recent literatures, and propose some practical strategies for the evaluation and development of such systems utilizing a working diagram.

Abstract:Using microwave irradiation, we were able to prepare a concentrated (up to 40 wt.%) uniform polystyrene nanoparticles (down to similar to 60 nm) in the presence of 1.82 wt.% of anionic surfactant (sodium dodecylsulfate). Both the nanoparticles and the polystyrene chains inside the nanoparticles were characterized by a combination of static and dynamic laser light scattering (LLS). We also found that various surfactant-free polymeric nanoparticles could be made by microphase inversion; namely, the addition of a polymer solution dropwise into an excess of water or the temperature-induced phase transition. We proposed a stabilization mechanism and experimentally demonstrated that for a given stable dispersion the average surface area (s) occupied per stabilizer is a constant. On the basis of this mechanism, we were able to propose a structural model to control and predict the resultant particle size from the macroscopic monomer/stabilizer weight ratio. We also successfully used the nanoparticles in the studies of enzymatic biodegradation and controllable chemical releasing.

Abstract:Age-long ambition of medical scientists has always been advancement in healthcare and therapeutic medicine. Biomedical research indeed claims paramount importance in nanomedicine and drug delivery, and the development of biocompatible storage structures for delivering drugs stands at the heart of emerging scientific works. The delivery of drugs into the human body is nevertheless a nontrivial and challenging task, and it is often addressed by using amphiphilic compounds as nanosized delivery vehicles. Pluronics belong to a peculiar class of biocompatible and thermosensitive nonionic amphiphilic copolymers, and their self-assemblies are employed as drug delivery excipients because of their unique properties. We herein report on the encapsulation of diclofenac sodium within Pluronic F68 self-assemblies in water, underpinning the impact of the drug on the rheological and microstructural evolution of pluronic-based systems. The self-assembly and thermoresponsive micellization were studied through isothermal steady rheological experiments at different temperatures on samples containing 45 wt % Pluronic F68 and different amounts of diclofenac sodium. The adoption of scattering techniques, small-angle X-ray scattering (SAXS) and small-angle neutron scattering (SANS), allowed for the description of the system features at the nanometer length scale, providing information about the characteristic size of each part of the micellar structures as a function of temperature and drug concentration. Diclofenac sodium is not a good fellow for Pluronic F68. The triblock copolymer aids the encapsulation of the drug, highly improving its water solubility, whereas diclofenac sodium somehow hinders Pluronic self-assembly. By using a simple empirical model and no fitting parameters, the steady viscosity can be predicted, although qualitatively, through the volume fraction of the micelles extracted through scattering techniques and compared to the rheological one. A tunable control of the viscous behavior of such biomedical systems may be achieved through the suitable choice of their composition.

Abstract:Understanding the interplay between kinetics and thermodynamics of polymer-mediated liquid-liquid phase separation is crucial for designing and implementing an amorphous solid dispersion formulation strategy for poorly water-soluble drugs. This work investigates the phase behaviors of a poorly water-soluble model drug, celecoxib (CXB), in a supersaturated aqueous solution with and without polymeric additives (PVP, PVPVA, HPMCAS, and HPMCP). Drug-polymer-water ternary phase diagrams were also constructed to estimate the thermodynamic behaviors of the mixtures at room temperature. The liquid-liquid phase separation onset point for CXB was detected using an inline UV/vis spectrometer equipped with a fiber optic probe. Varying CXB concentrations were achieved using an accurate syringe pump throughout this study. The appearance of the transient nanodroplets was verified by cryo-EM and total internal reflection fluoresence microscopic techniques. The impacts of various factors, such as polymer composition, drug stock solution pumping rates, and the types of drug-polymer interactions, are tested against the onset points of the CXB liquid-liquid phase separation (LLPS). It was found that the types of drug-polymer interactions, i.e., hydrogen bonding and hydrophobic interactions, are vital to the position and shapes of LLPS in the supersaturation drug solution. A relation between the behaviors of LLPS and its location in the CXB-polymer-water ternary phase diagram was drawn from the findings.

Abstract:This study aimed to explore the link between block copolymers’ interfacial properties and nanoscale carrier formation and found out the influence of length ratio on these characters to optimize drug delivery system. A library of diblock copolymers of PEG-PCL and triblock copolymers with additional PEI (PEG-PCL-PEI) were synthesized. Subsequently, a systematic isothermal investigation was performed to explore molecular arrangements of copolymers at air/water interface. Then, structural properties and drug encapsulation in self-assembly were investigated with DLS, SLS and TEM. We found the additional hydrogen bond in the PEG-PCL-PEI contributes to film stability upon the hydrophobic interaction compared with PEG-PCL. PEG-PCL-PEI assemble into smaller micelle-like (such as PEG-PCL4006-PEI) or particle-like structure (such as PEG-PCL8636-PEI) determined by their hydrophilic and hydrophobic block ratio. The distinct structural architectures of copolymer are consistent between interface and self-assembly. Despite the disparity of constituent ratio, we discovered the arrangement of both chains guarantees balanced hydrophilic–hydrophobic ratio in self-assembly to form stable construction. Meanwhile, the structural differences were found to have significant influence on model drugs incorporation including docetaxel and siRNA. Taken together, these findings indicate the correlation between molecular arrangement and self-assembly and inspire us to tune block compositions to achieve desired nanostructure and drug loading.

Abstract:Pluronic poly(ethylene oxide)-b-poly(propylene oxide)-b-poly(ethylene oxide) (PEO–PPO–PEO) block copolymers are grafted with poly(vinyl pyrrolidone) by free radical polymerization of vinyl pyrrolidone with simultaneous chain transfer to the Pluronic in dioxane. This modified polymer has both thermal responsiveness and remarkable capacity to interact with a wide variety of hydrophilic and hydrophobic pharmaceutical agents which is very attractive for medical applications. The chemical structure of the graft copolymers was characterized by FTIR and 1H NMR spectroscopy. Polymerization conditions such as initiators, feed ratio, and reaction times are studied to obtain the ideal graft copolymer.

Abstract:In recent years, advancements in bioengineering and materials science have witnessed increasing interest in synthetic polymers capable of fulfilling various applications. Owing to their distinctive properties, Pluronics can be used as nano-drug carriers, to deliver poorly water-soluble drugs, and as model systems to study colloidal science by tuning amphiphilic properties. In this work, we investigated the effect of diclofenac sodium on the self-assembly and thermoresponsive crystallization of Pluronic F68 in water solutions, by employing experimental rheology and Nuclear Magnetic Resonance (NMR). We built a complete phase diagram as a function of temperature and concentration for 45 wt% Pluronic F68 with various amounts of diclofenac sodium in water. The morphological transitions were followed as a function of temperature via linear rheology. We extrapolated the transition temperatures - identifying distinct phases - as a function of the drug concentration and proposed an empirical model for their prediction. NMR analysis provided further information on the structural characteristics of the systems, shedding light on the interactions between F68 and diclofenac sodium. Although dealing with a pharmaceutical salt, the study is focused on a colloidal system and its interaction with a binding molecule, that is of general interest for colloidal science.
Surface Enrichment and Depletion of the Active Ingredient in Spray Dried Amorphous Solid Dispersions

Abstract:Purpose To study the effects of physicochemical properties of drug and polymer, as well as the drug-polymer interactions, on the surface composition of SDDs. Methods Ethanol solutions containing a model drug (IMC, NMP or FCZ) and a model polymer (PVPK12, PVPK30 or PVP-VA) were spray dried, and the surface composition of SDDs was analyzed by XPS. The surface tensions of pure components and their solutions were measured using Wilhelmy plate and/or calculated using ACD/Labs. NMR and DLS were used to obtain the diffusion coefficients of IMC, NMP, PVPK12 and PVPK30 in solvents. Flory-Huggins interaction parameters for selected drug-polymer pairs were obtained using a melting point depression method. Results Significant surface enrichment or depletion of the drug was observed in SDDs depending on the particular drug-polymer combination. With PVP as the dispersion polymer, IMC and NMP were surface enriched; whereas FCZ, a hydrophilic drug, was surface depleted. With increasing PVP molecular weight, the surface drug concentration increased, and the effect was greater in the NMP/PVP and FCZ/PVP systems than in the IMC/PVP system where strong drug-polymer interaction existed. Changing the polymer from PVP to PVP-VA reduced the surface concentration of the drug. Conclusions The surface concentration of a SDD can be significantly different from the bulk concentration. The main results of this work are consistent with the notion that the relative surface tensions control surface enrichment or depletion. Besides, the relative diffusion rates of the components and the strength of their interactions may also affect the surface composition of the SDDs.
Drug-polymer-water interaction and its implication for the dissolution performance of amorphous solid dispersions.

Abstract:The in vitro dissolution mechanism of an amorphous solid dispersion (ASD) remains elusive and highly individualized, yet rational design of ASDs with optimal performance and prediction of their in vitro/in vivo performance are very much desirable in the pharmaceutical industry. To this end, we carried out comprehensive investigation of various ASD systems of griseofulvin, felodipine, and ketoconazole, in PVP-VA or HPMC-AS at different drug loading. Physiochemical properties and processes related to drug-polymer-water interaction, including the drug crystallization tendency in aqueous medium, drug-polymer interaction before and after moisture exposure, supersaturation of drug in the presence of polymer, polymer dissolution kinetics, etc., were characterized and correlated with the dissolution performance of ASDs at different dose and different drug/polymer ratio. It was observed that ketoconazole/HPMC-AS ASD outperformed all other ASDs in various dissolution conditions, which was attributed to the drugs low crystallization tendency, the strong ketoconazole/HPMC-AS interaction and the robustness of this interaction against water disruption, the dissolution rate and the availability of HPMC-AS in solution, and the ability of HPMC-AS in maintaining ketoconazole supersaturation. It was demonstrated that all these properties have implications for the dissolution performance of various ASD systems, and further quantification of them could be used as potential predictors for in vitro dissolution of ASDs. For all ASDs investigated, HPMC-AS systems performed better than, or at least comparably with, their PVP-VA counterparts, regardless of the drug loading or dose. This observation cannot be solely attributed to the ability of HPMC-AS in maintaining drug supersaturation. We also conclude that, for fast crystallizers without strong drug-polymer interaction, the only feasible option to improve dissolution might be to lower the dose and the drug loading in the ASD. In this study, we implemented an ASD/water Flory-Huggins parameter plot, which might assist in revealing the physical nature of the drug-polymer interaction. We also introduced supersaturation parameter and dissolution performance parameter as two quantitative measurements to compare the abilities of polymers in maintaining drug supersaturation, and the dissolution performance of various solid dispersions, respectively.

Interfacial Engineering of Block Copolymer Nanostructures: Morphology and Solvent Stability

Abstract:Interfacial engineering is a critical pathway for modulating the self-assembled nanostructures of block copolymers (BCPs) during solvent exchange. Herein, we demonstrated the generation of different stacked lamellae of polystyrene-block-poly(2-vinyl pyridine) (PS-b-P2VP) nanostructures during solvent exchange by using phosphotungstic acid (PTA) or PTA/NaCl aqueous solution as the nonsolvent. The participation of PTA in the confined microphase separation of PS-b-P2VP in droplets increases the volume fraction of P2VP and decreases the tension at the oil/water interface. Moreover, the addition of NaCl to the PTA solution can further increase the surface coverage of P2VP/PTA on droplets. All factors impact the morphology of assembled BCP nanostructures. In the presence of PTA, ellipsoidal particles composed of alternatively stacked lamellae of PS and P2VP were formed (named BP), whereas, in the coexistence of PTA and NaCl, they changed to stacked disks with PS-core-P2VP-shell (called BPN). The different structures of assembled particles induce their different stabilities in solvents and different dissociation conditions as well. The dissociation of BP particles was easy because PS chains were only entangled together which can be swollen in toluene or chloroform. However, the dissociation of BPN was hard, requiring an organic base in hot ethanol. The structural difference in BP and BPN particles further extended to their dissociated disks, which makes the cargo (like R6G) loaded on these disks to show a different stability in acetone. This study demonstrated that a subtle structural change can greatly affect their properties.

Elucidation of Molecular Interactions Between Drug-Polymer in Amorphous Solid Dispersion by a Computational Approach Using Molecular Dynamics Simulations

Abstract:Introduction: Amorphous drug dispersion is frequently used to enhance the solubility and dissolution of poorly water-soluble drugs, thereby improving their oral bioavailability. The dispersion of these drugs into polymer matrix can inhibit their recrystallization. The inter-molecular interactions between drug and polymer plays a role in the improvement of the dissolution rate, solubility, and physical stability of drug. Aim: This study aims to investigate the formation and interactions of ritonavir (RTV)/poloxamer (PLX) amorphous formulation using a computational approach via molecular dynamics (MD) simulations, which mimicked solvent evaporation and melt-quenching method. Methods: TheRoot Mean Square Deviation (RMSD) value, Root Mean Square Fluctuation (RMSF), Radial Distribution Function (RDF), Radius of Gyration (Rg), Solvent Accessible Surface Area (SASA), and hydrogen bond interactions were analyzed to determine interaction mechanisms between RTV and PLX in amorphous solid dispersion. Results: The pi-alkyl bonds between RTV and PLX were formed after simulations of solvent evaporation, while the hydrogen bond interactions of RTV-PLX was observed during melt method simulations. These results indicate the successful formulation of amorphous solid dispersion (ASD) from RTV and PLX. The RMSD values obtained from the solvent evaporation, melt-cooling-A, melt-cooling-B, and melt-cooling-C methods were 3.33 Å, 1.97 Å, 1.30 Å, and 1.29 Å, respectively, while the average RMSF values were 2.65 Å, 1.04 Å, 1.05 Å, and 1.07 Å, respectively. This indicates that the suppression of translational motion of RTV from the melt method can be stronger than solvent evaporation caused by the intermolecular interactions of RTV-PLX. Conclusion: MD simulations helped in understanding the formation and interaction mechanisms of ASD formulations that were difficult to detect by experimental approaches.
