Abstract:OBJECTIVE:To observe the expression of Coxsackie B virus-adenovirus receptor (CAR) in two prostate cancer cell lines with different metastatic potentials.METHODS:The expressions of CAR in two prostate cancer cell lines (Du145 and LNCaP) with different metastatic potentials were detected by Western blotting. The Transwell polycarbonate filter was used to analyze the invasive potency.RESULTS:As one of the adhesion associated proteins, CAR highly expressed in the LNCaP cell line, which is well known with a low metastatic potential, and lowly expressed in Du145 with a high metastatic potential (P < 0.01). The invasive potency of Du145 was significantly higher than that of LNCaP (P < 0.05).CONCLUSION:There was a difference in the metastatic phenotypes of CAR among cell lines with different metastatic potentials. The expressions of CAR proteins may play an important role in repressing the metastasis of prostate cancer.
Overexpression of Coxsackie and Adenovirus Receptor Inhibit Growth of Human Bladder Cancer Cell in Vitro and in Vivo.

Abstract:AIM:To study the effect of the overexpression of coxsackie and the adenovirus receptor (CAR) on the growth of the human bladder cancer cell in vitro and in vivo.METHODS:A retroviral vector pLXSN-CAR expressing CAR was constructed and confirmed by restriction enzyme mapping. The pLXSN-CAR vector and control vector pLXSN were transfected into the PT67 packaging cell line to generate retrovirus with high titer. The CAR-negative T24 cell was infected with the pLXSN-CAR and the pLXSN retrovirus, respectively. The positive clone cells were selected with G418 for 2 weeks. The expression level of the CAR protein was detected by Western blot assay. T24 cell growth in vitro was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Anchorage-independent growth was measured by soft-agar colony formation assay. In vivo cell growth was determined by a nude mice xenograft model.RESULTS:The pLXSN-CAR vector containing full-length CAR cDNA was successfully constructed. Western blot analysis showed that a 46 kDa specific band was found in pLXSN-CA-transfected T24 cells. MTT assay identified the growth inhibition of T24/pLXSN-CAR cells. The cell colony forming ability of T24/pLXSN-CAR cells was significantly lower than that of T24/pLXSN and parental T24 cells. There was a reduction in the tumor size in the T24/pLXSN-CAR group as compared with that of the T24/pLXSN group and parental T24 group.CONCLUSION:The overexpression of CAR in T24 bladder cancer cells can inhibit cell growth both in vitro and in vivo.

Abstract:Coxsackievirus and adenovirus receptor (CAR) is a transmembrane cell-cell adhesion receptor that forms homodimers across junctions and plays a key role in mediating epithelial barrier integrity. CAR can also heterodimerise with receptors on the surface of leukocytes and thus plays an additional role in mediating immune cell transmigration across epithelial tissues. Given the importance of both biological processes in cancer, CAR is emerging as a potential mediator of tumorigenesis as well as a target on cancer cells for viral therapy delivery. However, the emerging, often conflicting, evidence suggests that CAR function is tightly regulated and that contributions to disease progression are likely to be context specific. Here, we summarise reported roles for CAR in the context of cancer and draw on observations in other disease settings to offer a perspective on the potential relevance of this receptor as a therapeutic target for solid tumours.

Abstract:Objective:To investigate the relationship between adenovirus type 5 infection efficiency and expression of Coxsackie virus adenovirus receptor(CAR)and integrin in different tumor cell lines,providing a basis for further study of adenovirus gene therapy.Methods:The expression levels of CAR in different cell lines were examined by flow cytometry.The cells were infected with Ad5-CMV-GFP and Ad5-CMV-luc carrying report genes,tested the infection efficiency and determined their expression levels by flow cytometry and Luciferase Assay.The variation of adenovirus infection efficiency was evaluated and compared when the CAR expression was enhanced on cell surface with plasmid expressing CAR or blocked by antibodies of CAR and integrin.Results:The highest CAR expression was found in SMMC-7721 and A549 cell lines,the lowest in K562 cells.The results of fluorescence microscope and flow cytometry suggested that Ad5 had the highest efficiency in infecting SMMC-7721 and A549 cell lines,and the lowest in infecting K562 cells.Conclusion:The infection efficiency of adenovirus is determined by the levels of CAR and integrin on cancer cells.
Relationship between coxsackie adenovirus receptor expression levels with ad5 transduction efficiency in tumor cells

Abstract:Objective: To investigate the relationship between coxsackie adenovirus receptor (CAR) expression and the efficiency of adenovirus gene transfer, in an effort to provide evidence for the clinical application of adenovirus-related bio-preparations. Methods:The esophageal cancer cell lines (KYSE510, KYSE150, EC9706), cervical cancer cell line (HeLa), ovary cancer cell line (SKOV3), hepatoma cell line (HepG2), and lung cancer cell line (A549) were infected with Ad-GFP (100 MOI, 200 MOI) labeled by immunofluorescence; 48 h later we measured the Ad-GFP transduction efficiencies in the above cell lines by flow cytometric analysis. CAR expression levels in vitro were assayed by Western blotting. HeLa (3×10 6 ), A549 (3×10 6 ), SKOV3 (3×10 6 ), and EC9706 (2×10 6 ) cells were implanted subcutaneously into nude mice. When the tumor sizes reached 5-7 mm in diameter, the xenografts were injected with Ad-GFP (1×10 9 PFU) twice with a 48 h interval. After another 48 h the mice were killed and the GFP expression were observed by fluorescence microscopy to determine the transduction efficiency. CAR expression was detected by immunohistochemistry in the xenograft tissues. Results: We found that Ad transduction efficiencies varied greatly in different tissues, even among the cells of the same tissue origin. The transduction efficiencies in A549 (92.67%), HeLa (89.31%), HepG2 (84.98%) and KYSE150 (74.59%) were higher than those in SKOV3 (30.06%), KYSE510 (27.40%), and EC9706 (18.93%). Consistently, GFP expression was much higher in exografts sections resulting from HeLa and A549 than those from SKOV3 and EC9706. Increased CAR expression was predictive for more efficient gene transfer in vitro and in vivo.Conclusion:CAR expression level is closely correlated to Ad transduction efficiency, which suggests that measurement of CAR expression in tumor tissues is useful for individualized adenovirus based gene therapy in clinic.

Abstract:Objective To determine the expression of E-cadherin and CAR in bladder cancer cell lines and explore their significance and relation.Methods Expression of E-cadherin and CAR in bladder cancer cell lines was detected by Western blot.Results E-cadherin and CAR expressions were high in RT4 and 5637 cell,but low in 253J.E-cadherin was not detected in J82 and T24 cell.Expression of CAR was low in J82 cell,and no expression in T24 cell.Conclusion The expressions of E-cadherin and CAR were coincident in bladder cancer cell lines,indicating they may cooperate in the invasion and metastasis of bladder cancer.
Expression and Significance of Coxsackie-Adenovirus Receptor in Myocardia With Dilated Cardiomyopathy

Abstract:Objective:To clarify the expression and significance of coxsackie-adenovirus receptor (CAR) in myocardia with dilated car- diomyopathy (DCM). Methods:Myocardium specimens derived from 20 patients with DCM underwent orthotopic cardiac transplantation and 5 nor- mal myocardium specimens were collected at autopsy from sudden deaths.In the myocardia,coxsackievirus B (CVB)and adeno- virus (ADV) genes were detected by polymerase chain reaction (PCR),and the expression levels of CAR mRNA and protein were examined by reverse transcription (RT)-PCR and immunohistochemistry,respectively. Results:Viral genomes could be amplified from the myocardia of 12 DCM patients (CVB=7,ADV=5).Viral genomes failed to be detected from myocardia of 5 controls.Therefore,the present experiment had four groups:eoxsackievirus,adenovi- rus,the other and the control.There were no CAR mRNA and protein expression in the other group or normol control group. However,the expression levels of CAR mRNA and protein were remarkedly elevated in coxsackievirus and adenovirus groups, There were no differences between coxsackievirus group and adenovirus group. Conclusion:Upregulated CAR expression can increase the susceptibility of CVB or ADV,which makes viral myoearditis pro- gressing to dilated cardiomyopathy.

Abstract:The coxsackie and adenovirus receptor (CAR) is a transmembrane receptor that plays a key role in controlling adhesion between adjacent epithelial cells. CAR is highly expressed in epithelial cells and was originally identified as a primary receptor for adenovirus cell binding. However, studies over the last 10 years have demonstrated that CAR plays a key role in co-ordinating cell-cell adhesion under homeostatic conditions including neuronal and cardiac development and cell junction stability; it has also been implicated in pathological states such as cancer growth and leukocyte transmigration during inflammation. Here we provide an overview of the functions of CAR as an adhesion molecule and highlight the emerging important role for CAR in controlling both recruitment of immune cells and in tumorigenesis.

Abstract:Breast cancer is the second leading cause of death in women, with invasive ductal carcinoma (IDC) and invasive lobular carcinoma (ILC) as the two most common forms of invasive breast cancer. While estrogen receptor positive (ER+) IDC and ILC are treated similarly, the multifocality of ILC presents challenges in detection and treatment, worsening long-term clinical outcomes in patients. With increasing documentation of chemoresistance in ILC, additional treatment options are needed. Oncolytic adenoviral therapy may be a promising option, but cancer cells must express the coxsackievirus & adenovirus receptor (CAR) for adenoviral therapy to be effective. The present study aims to evaluate the extent to which CAR expression is observed in ILC in comparison to IDC, and how the levels of CAR expression correlate with adenovirus transduction efficiency. The effect of liposome encapsulation on transduction efficiency is also assessed.

Abstract:Abstract BackgroundLung adenocarcinoma, the main subtype of non-small cell lung cancer, leading one of the most aggressive and fatal causes of malignant deaths around the world. Scavenger receptor class A member 5 (SCARA5), a newly discovered tumor suppressor gene, belonged to the SR family. The present study’s object was to explore the clinical impacts of SCARA5 in lung adenocarcinoma treatment. MethodsSCARA5 expressions in 120 paired lung adenocarcinoma patients’ tissues and cell lines were detected by real-time quantitative polymerase chain reaction (RT-qPCR) and immunohistochemistry (IHC). CCK-8, EdU, flow cytometry, and western blot assays further demonstrated the significance of SCARA5 on A549 cell growth. Then, the relationships between the expression level of SCARA5 and pathological factors were analyzed. Finally, the receiver operating characteristic (ROC) curve and overall survival analysis was carried out to verify the diagnostic and prognostic significance of SCARA5 in lung adenocarcinoma. ResultsSCARA5 was prominently decreased in lung adenocarcinoma cells and tissues compared with human bronchial epithelial cells and para-carcinoma non-tumor tissues. Meanwhile, SCARA5 expression was strongly correlated with smoking (P = 0.0011), TNM stage (P < 0.0001) and lymph node metastasis (P = 0.0005). Furthermore, SCARA5 may be a crucial biomarker for lung adenocarcinoma diagnosis with an area under the curve (AUC) of 0.9102 while SCARA5 could significantly reduce overall survival (OS; P = 0.0006). In vitro experiments, we found that SCARA5 overexpressing could significantly hinder A549 cell proliferation but facilitate apoptosis through the AKT signaling pathway. ConclusionsSCARA5 might be an important diagnostic and prognostic biomarker for patients with lung adenocarcinoma.
Expression of SRF, E-cadherin and β-catenin in hepatocellular carcinoma and its clinical significances

Abstract:Objective: To investigate the expression of serum response factor(SRF),E-cadherin and βcatenin in hepatocellular carcinoma and its relation to clinicopathological characterisitics of the tumor and prognosis.Methods: Tissue microarray and immunohistochemical study(EnVision method) for SRF,E-cadherin and βcatenin was performed on tissue microarray which consisted of 182 cases of hepatocellular carcinoma,92 cases of adjacent non-neoplastic liver tissues.Results: In the hepatocellular carcinoma group 75.8% tumor samples expressed SRF,and the levels is significantly higher than that in surrounding liver tissue(21.7%,P<0.05).The expression of SRF was positively correlated with the TNM stage,tumor metastasis and recurrence(P<0.05).In the hepatocellular carcinoma group,aberrant expression of E-cadherin and β-catenin was 59.3% and 58.8% respectively,and these levels were higher than those in the adjacent non-neoplastic liver tissue group(30.4% and 26.1% P<0.05).Aberrant expression of E-cadherin andβ-catenin was related to vascular cancer embolus,the TNM stage,tumor metastasis and recurrence(P<0.05).The expression of SRF correlated with aberrant expression of E-cadherin and β-catenin in hepatocellular carcinoma.Conclusion: In hepatocellular carcinoma,the overexpression of SRF and the downregulated expression of E-cadherin and β-catenin may be important mechanisms for promoting the invasion and metastasis of hepatocellular carcinoma and may also play a key role in the occurrence and development of hepatocellular carcinoma.
The clinical and biological significance of MICA in clear cell renal cell carcinoma patients

Abstract:Major histocompatibility complex class I-related chains A (MICA), a ligand of Natural killer group 2, member D (NKG2D) receptor, is broadly upregulated in epithelial originated tumor cells. MICA plays a critical role in the immune surveillance against tumor cells and is associated with the prognosis of several malignancies. The aim of this study is to evaluate the clinical and biological significance of MICA in clear cell renal cell carcinoma (ccRCC). The expression of MICA was analyzed by quantitative real-time PCR (qRT-PCR) and immunohistochemistry (IHC). Both MICA mRNA and protein levels were upregulated in ccRCC tissues, compared with normal tissues. IHC staining revealed a homogenous pattern of MICA staining within each tumor, which combined both membrane staining and granular cytoplasmic staining. Furthermore, high MICA expression was associated with lymph node metastasis and advanced clinical stage and predicted poor prognosis in patients with ccRCC. Gene set enrichment analysis (GSEA) was performed using RNA-sequencing data from The Cancer Genome Atlas Research Network (TCGA) to elucidate the biological role of MICA in ccRCC and revealed that MICA was significantly associated with the epithelial-to-mesenchymal transition (EMT) gene set, which was further confirmed by qRT-PCR. Our findings contribute to the studies on biomarkers of kidney cancers and the mechanism of renal cancer progression driven by EMT pathway.

Abstract:Objective To investigate the roles of cysteine-X-cysteine chemokine receptor 4(CXCR4)in the genesis and development of clear cell renal cell cancer.Methods The expression of CXCR4 was tested by immunohistochemical method in 53 cases of clear cell renal cell cancer specimens(observed group) and 20 cases of peri-cancerous renal specimens(control group),and the relationship between the expression of CXCR4 and clinicopathological parameter of clear cell renal cell cancer were analyzed.Results The positive expression rate of CXCR4 in observed group and control group was 62.2% and 30.0% respectively,P0.05.The expression of CXCR4 was relevant to lymph node status,clinical stage and histologic grade,but was not correlated with tumor size,the age and gender of patients.Conclusion High expression of CXCR4 possibly may improve the genesis and development of clear cell renal cell cancer,and can be used as an important prognostic index of clear cell renal cell cancer.

Abstract:OBJECTIVE: To explore the expression and clinical significance of chemokine receptor (CXCR4) in clear cell renal cell carcinoma (CCRCC). METHODS: The expression of CXCR4 was checked in 42 cases of renal carcinoma and 10 cases of normal renal tissues on the protein level by the immunohistochemical methed. The expression of CXCR4 mRNA was checked at transcription level by reverse transcription PCR (RT-PCR). The correlations between CXCR4 and the clinical stage, pathological grade, lymphatic metastasis respectively were analyzed. RESULTS: The immunohistochemistry analysis showed that the expression of CX-CR4 protein in the CCRCC tissues was 78.5%, and there was not CXCR4 protein detected in the nomal renal tissues. There was highly a significant difference (X 2 = 18.249, P = 0.000). Out of 42 cases, 35 cases presented high expression levels of CXCR4 mRNA (83.3%). CXCR4 had close correlations with the clinical stage, pathological grade and lymphatic metastasis (P<0.05). CXCR4 mRNA had close correlations with the pathological grade and lymphatic metastasis (P<0.05). CONCLUSIONS: The expreessions of CXCR4 mRNA and protein possibly play important roles in the genesis and development of CCRCC. CXCR4 could be an important prognostic index of CCRCC.

Abstract:Coxsackie and adenovirus receptor (CAR) is essential for adenovirus infection to target cells, and its constitutive expression in various cancerous and normal tissues has been reported. Recently, the biological role of CAR in human cancers of several different origins has been investigated with respect to tumor progression, metastasis and tumorigenesis. However, its biological function in tumor cells remains controversial. Here we report the critical role of CAR in growth regulation of oral squamous cell carcinomas (SCCs) in vitro and in vivo via the specific interaction with Rho-associated protein kinase (ROCK). Loss of endogenous CAR expression by knockdown using specific small interfering RNA (siRNA) against CAR facilitates growth suppression of SCC cells due to cell dissociation, followed by apoptosis. The consequent morphological reaction was reminiscent of anoikis, rather than epithelial–mesenchymal transition, and the dissociation of oral SCC cells was triggered not by lack of contact with extracellular matrix, but by loss of cell-to-cell contact caused by abnormal translocation of E-cadherin from surface membrane to cytoplasm. Immunoprecipitation assays of the CAR-transfected oral SCC cell line, HSC-2, with or without ROCK inhibitor (Y-27632) revealed that CAR directly associates with ROCKI and ROCKII, which results in inhibition of ROCK activity and contributes to maintenance of cell-to-cell adhesion for their growth and survival. Based on these findings, in vivo behavior of CAR-downregulated HSC-2 cells from siRNA knockdown was compared with that of normally CAR-expressing cells in intraperitoneally xenografted mouse models. The mice engrafted with CAR siRNA-pretreated HSC-2 cells showed poor formation of metastatic foci in contrast to those implanted with the control siRNA-pretreated cells. Thus, CAR substantially has an impact on growth and survival of oral SCC cells as a negative regulator of ROCK in vitro and in vivo.

Abstract:The coxsackie and adenovirus receptor (CAR) is a member of the junctional adhesion molecule (JAM) family of adhesion receptors and is localised to epithelial cell tight and adherens junctions. CAR has been shown to be highly expressed in lung cancer where it is proposed to promote tumor growth and regulate epithelial mesenchymal transition (EMT), however the potential role of CAR in lung cancer metastasis remains poorly understood. To better understand the role of this receptor in tumor progression, we manipulated CAR expression in both epithelial-like and mesenchymal-like lung cancer cells. In both cases, CAR overexpression promoted tumor growth in vivo in immunocompetent mice and increased cell adhesion in the lung after intravenous injection without altering the EMT properties of each cell line. Overexpression of WTCAR resulted in increased invasion in 3D models and enhanced β1 integrin activity in both cell lines, and this was dependent on phosphorylation of the CAR cytoplasmic tail. Furthermore, phosphorylation of CAR was enhanced by substrate stiffness in vitro, and CAR expression increased at the boundary of solid tumors in vivo. Moreover, CAR formed a complex with the focal adhesion proteins Src, Focal Adhesion Kinase (FAK) and paxillin and promoted activation of the Guanine Triphosphate (GTP)-ase Ras-related Protein 1 (Rap1), which in turn mediated enhanced integrin activation. Taken together, our data demonstrate that CAR contributes to lung cancer metastasis via promotion of cell-matrix adhesion, providing new insight into co-operation between cell-cell and cell-matrix proteins that regulate different steps of tumorigenesis.

Abstract:Background: Long noncoding RNAs (lncRNAs) are an important class of pervasive genes involved in a variety of biological functions. They are aberrantly expressed in many types of cancers. In this study, we described lncRNAs profiles in 6 pairs of human renal clear cell carcinoma (RCCC) and the corresponding adjacent nontumorous tissues (NT) by microarray.Methodology/Principal Findings: With abundant and varied probes accounting 33,045 LncRNAs in our microarray, the number of lncRNAs that expressed at a certain level could be detected is 17157. From the data we found there were thousands of lncRNAs that differentially expressed (>= 2 fold-change) in RCCC tissues compared with NT and 916 lncRNAs differentially expressed in five or more of six RCCC samples. Compared with NT, many lncRNAs were significantly up-regulated or down-regulated in RCCC. Our data showed that down-regulated lncRNAs were more common than up-regulated ones. ENST00000456816, X91348, BC029135, NR_024418 were evaluated by qPCR in sixty-three pairs of RCCC and NT samples. The four lncRNAs were aberrantly expressed in RCCC compared with matched histologically normal renal tissues.Conclusions/Significance: Our study is the first one to determine genome-wide lncRNAs expression patterns in RCCC by microarray. The results displayed that clusters of lncRNAs were aberrantly expressed in RCCC compared with NT samples, which revealed that lncRNAs differentially expressed in tumor tissues and normal tissues may exert a partial or key role in tumor development. Taken together, this study may provide potential targets for future treatment of RCCC and novel insights into cancer biology.
Expression and Significance of NFκB and CXCR4 in Renal Cell Carcinoma

Abstract:Objective To detect the expression of NFκB and CXCR4 in renal cell carcinoma(RCC) and discuss the significance of the two cytokines for the development and progression of RCC.Methods Immunohistochemistry(IMC) was employed to inspect the expression of p65/NFκB and CXCR4 protein in 43 cases of renal cell carcinoma(RCC) and 21 adjacent renal tissues.The correlation between the expression of the two cytokines,and the clinic/pathologic parameter,were analysed.Results NFκB(p65) was positvely expressed in 33(76.7%) RCCs and 10(47.6%) adjacent renal tissues;CXCR4 was positvely expressed in 29(67.4%) RCCs and 7(33.3%) adjacent renal tissues.The positive expression of NFκB(p65) and CXCR4 was obviously diferent between RCC and adjacent renal tissues.NFκB(p65) and CXCR4 were expressed in all RCCs in Ⅲ-Ⅳ clinic-stage(100%),which was obviously higher than in the Ⅰ-Ⅱ clinic-stage(66.7% and 53.3%),but they did not correlate with pathological grades,the age and gender of patients.Furthermore,a correlation existed between NFκB(p65) and CXCR4 in the RCCs(r=0.44).Conclusion NFκB and CXCR4 closely correlated with the development and progression of RCC and there might existe a regulating correlation between them.Therefore,they could become a potential therapeutic and preventive targetets and metastasis of RCC.
Expression and Clinical Significance of RCDG1 in Renal Cell Carcinoma: A Novel Renal Cancer-Associated Gene

Abstract:Recently identified molecular tumor markers have numerous potential applications in the diagnosis, therapy and prognostic prediction of renal cell carcinoma (RCC). Through bioinformatics‑based screening approaches together with validation of western blot and immunohistochemical data, the present study identified a novel renal cancer‑associated gene, preliminarily named Renal Cancer Differentiation Gene 1 (RCDG1), originally known as chromosome 4 open reading frame 46 (C4orf46). RCDG1 expression was evaluated by western blot analysis of RCC and adjacent normal tissues, renal cancer cell lines and normal kidney HEK293T cells. Additionally, RCDG1 expression was assessed in 124 RCC paraffin sections, including 92 paired adjacent normal tissues, by immunohistochemistry. The results showed that RCDG1 was significantly downregulated in RCC tissues as compared with normal adjacent tissues (P<0.001), and the expression of RCDG1 in clear cell (cc) RCC tissues was significantly lower as compared with that of non‑ccRCC tissues (P=0.005). Furthermore, statistical analysis revealed RCDG1 expression was negatively correlated with the Fuhrman grade in ccRCC (P=0.008). A reduction in RCDG1 expression may be associated with the oncogenesis of RCC and the differentiation of ccRCC. Further studies may provide more information about the function of RCDG1 gene in RCC.

Abstract:Objective To investigate the role of coxsackievirus and adenovirus receptor (CAR) in the development of viral myocarditis. Methods Thirty mice were divided randomly into experimental group and control group. The mice were killed and the hearts were removed after being infected 10 days. The expression of CAR in the myocardium were examined by Western blotting and immunohistochemistry. In addition, the levels of CAR antigen were semi- quantitatively analyzed, and the relationship between the CAR expression levels and the myocardial lesions was investigated. Results Western blotting showed that CAR expression levels were significantly higher in experimental group than those in control group. Immunohistochemistry indicated that the expression levels of CAR distinctly increased in experimental group as compared with control group, and its index of CAR antigen positive signal was signi- ficiantly positively correlated with myocardial histopathological score. Conclusion The increase of CAR expression may play an important role in development of viral myocarditis.
