Correlation between the TNM Staging and Level of Circulating DNA in Colorectal Cancer: Clinical Study

Abstract:Objective To investigate the correlation between the TNM staging and level of circulating DNA in colorectal cancer.Methods A total of 42 patients with colorectal cancer,who were admitted to our hospital from March 2010 to September 2011,were enrolled into this study.In all the cases,we determined the levels of circulating DNA and CEA preoperatively and TNM staging postoperatively.Meanwhile,the level of circulating DNA was detected in 14 healthy adults as a control group.Results The mean level of circulating DNA was(22.14±16.16) μg/L in healthy adults,which was significantly lower than that detected in the stages Ⅰ,Ⅱ,Ⅲ,and Ⅳ colorectal cancer [(79.85±53.08) μg/L,(77.21±40.46) μg/L,(85.66±39.87) μg/L,and(145.52±51.72) μg/L,respectively,all P<0.05].In the cases of colorectal cancer,patients in stage Ⅳ showed significantly higher level of circulating DNA than other stages(all P<0.05).No significant difference was detected among the stages Ⅰ,Ⅱ and Ⅲ(P>0.05),nor among different age groups,sex groups,or patients with the tumor at different positions(P>0.05).No linear correlation existed between the levels of CEA and circulating DNA(r=0.153,P=0.333).Conclusions Detection of circulating DNA has advantages of minimal invasion,easy access to specimen,and repeatability.It can be a valuable tumor marker in early diagnosis of colorectal cancer.
