Effect of Berberine on Cognitive Impairment and Hippocampal Lesion in Diabetic Rats

Abstract:Objective To observe the effect of berberine on cognitive deficits and hippocampal tissue lesion in diabetic rats,and to evaluate its preventive effects on the brain of rat model.Methods Male SD rats,except for the blank group,were fed with high sugar and high fat diet for two months,and were given intraperitoneal injection of STZ 40mg/kg for the establishment of diabetes model.After the successful modeling,the diabetic rats were randomized into model group,and low-,middle-and high-dose berberine groups.After medication for one month,the Moriss water maze was used for behavior test,and then the rats were executed for blood and brain sampling.Blood glucose was detected,superoxide dismutase(SOD),malondialdehyde(MDA),Na+-K+-ATP enzyme,and acetylcholinesterase AchE)in the brain homogenate from some rats were examined.The brain of the rest rats were used for preparing hippocampal tissue pathological sections,and then the sections were stained respectively by HE and immunohistochemical method.Results Compared with those in the model group,body weight was increased and blood glucose was reduced(P < 0.05 or P < 0.01),escape latency was shortened,the percentage of staying the original quadrant and the frequencies of crossing the original platform position were increased(P < 0.05 or P < 0.01),SOD and Na+-K+-ATP enzyme activities were significantly increased,MDA level and AchE activity were significantly reduced(P < 0.05 or P < 0.01),and insulin growth factor(IGF-1) and Bcl-2 expression was increased while Bax expression was decreased in hippocampal CA1 area in berberine groups.Conclusions Berberine shows certain cerebral protection by lowering blood glucose,counteracting oxidation and apoptosis,and decreasing cholinesterase activity,thus to improve cognitive disorder and hippocampal tissue lesions in diabetes rats.
