Abstract:Statins, the 3-hydroxy-3-methylglutaryl-cooenzyme A (HMG-CoA) reductase inhibitors, have been shown to inhibit the migration of macrophages and smooth muscle cell proliferation leading to an antiproliferative effect. Although this beneficial effect of statins has been suggested to be independent of lipid lowering properties, the possible mechanisms responsible for this action is largely unknown. Gap junctions, which serve as channels for direct intercellular exchange of ions, secondary messengers, and small signaling molecules, play an important role in tissue homeostasis and regulation of growth, differentiation, and development. This study was designed to test the hypothesis that expression of the component proteins of gap junctions, connexins 40 and 43 (Cx40 and Cx43), is upregulated in arteries subjected to balloon injury and that this upregulation can be suppressed by statin therapy. Male New Zealand white rabbits were subjected to injury in which an angioplasty catheter was introduced into the right iliac artery from the femoral artery under fluoroscopic guidance. Five groups of rabbits (n = 6 to 7) were treated for 2 weeks with one of the following: balloon injury (BL); BL+lovastatin (BL+L, 10 mg/kg/day); BL+fluvastatin (BL+F, 10 mg/kg/day); sham operation (Sham); and control (Con). Immunohistochemistry studies showed that Cx40 and Cx43 were expressed in normal smooth muscle cells (SMC) throughout the media. Reverse transcription-polymerase chain reaction and Western blot analysis showed that Cx40 and Cx43 mRNA and protein expression was elevated after injury (P < .001 for both proteins and both assays), and these elevations were suppressed by lovastatin and fluvastatin to a similar degree (P < .05 for both drugs and both assays). Immunostaining of Cx40 and Cx43 was consistently enhanced in the neointimal area after injury and lovastatin and fluvastatin reduced staining of these proteins in the lessened neointimal layer. Transmission electron microscopy revealed that there were abundant gap junctions between neointimal SMC as well as fewer and smaller gap junctions after statin treatment. Therefore, balloon injury causes upregulation of Cx40 and Cx43 in neointimal SMC. Lovastatin and fluvastatin suppress upregulated Cx40 and Cx43 expression and reduce neointimal proliferation, suggesting that Cx40 and Cx43 may play a role in statin-induced antiproliferative effect.
Effects of Losartan on Expression of Connexins at the Early Stage of Atherosclerosis in Rabbits.

Abstract:AIM:to investigate effects of Losartan on expression of connexin 40 and 43 (Cx40 and Cx43), in arteries at the early stage of atherosclerosis in a rabbit model.METHODS:A total of 28 male New Zealand white rabbits were divided into following groups: control group, high fat diet group, and Losartan group (10 mg/kg/day). Losartan was administrated in food for two weeks. Iliac arteries were obtained for immunohistochemistry, transmission electron microscopy, Western blot, and reverse transcriptase-polymerase chain reaction (RT-PCR).RESULTS:Transmission electron microscopy revealed abundant gap junctions between neointimal smooth muscle cells (SMCs), which were markedly reduced by treatment. RT-PCR and Western blot assay showed that the mRNA and protein expression of Cx40 and Cx43 were elevated in the neointimal area at the early stage of atherosclerosis. The mRNA and protein expression of Cx43 were significantly down-regulated by losartan treatment but those of Cx40 were not markedly changed.CONCLUSION:Cx40 and Cx43 in the neointimal SMCs were up-regulated at the early stage of atherosclerosis. Losartan (an angiotensin-converting enzyme inhibitor) could reduce neointima proliferation and down-regulate the elevated protein expression of Cx43, suggesting the rennin-angiotensin system (RAS) plays an important role in the remodeling of gap junction between ventricular myocytes under pathological conditions.
Simvastatin Reduces Connexin43 Expression and Inhibits Rabbit Atherosclerotic Lesion Formation

Abstract:AIM: To demonstrate that simvastatin can influence atherosclerotic plaque formation and stability by decreasing the expression of gap junction protein connexin43 (Cx43) in atherosclerotic lesions. METHODS: The role of Cx43 in atherogenesis was examined by a pharmacological approach. The rabbit model of atherogenesis was established by a high-cholesterol diet. The expression of connexin 43 and macrophages in vivo was determined by immunohistochemistry and the expression of connexin 43 in cultured smooth muscle cells was determined with Western blot. The quantity of connexin43 expression was analyzed with computer images. RESULTS: We observed that HMG-CoA reductase inhibitors, or statins, lipid-lowering drugs well known for their pleiotropic antiatherogenic effects, reduced Cx43 expression in primary human vascular cells in vitro. Atheroma of rabbits orally treated with simvastatin contained fewer inflammatory cells and exhibited thicker fibrous caps than controls, which was associated with reduced Cx43 expression in lesions of statin-treated rabbits. CONCLUSION: These data indicate the critical role of Cx43-mediated gap junctional communication in atherosclerotic plaque formation. Reduced Cx43-mediated intercellular communication leads to changes in atherogenesis. These findings show that Cx43-mediated intercellular communication can be used as a new potential therapeutic target in atherogenesis.
Effects of losartan and ramipril on the connexin expression in a rabbit balloon injury model

Abstract:Intercellular gap junction (GJ) plays a pivotal role in the proliferation and transformation of vascular smooth muscle cells (VSMCs). Atorvastatin Prevents Connexin43 Remodeling in Hypertrophied Left Ventricular Myocardium of Spontaneously Hypertensive Rats.

Abstract:BACKGROUND:Connexin43 (Cx43) is the predominant gap junction protein in heart and is involved in the control of cell-to-cell communication to modulate the contractility and the electrical coupling of cardiac myocytes. Left ventricular (LV) hypertrophy is accompanied by changes of Cx43 expression. Recent studies have demonstrated that statins reduced cardiac hypertrophy. However, it is unknown whether statins can affect Cx43 expression in hypertrophied left ventricular myocardium. This study was designed to assess the effects of atorvastatin on LV hypertrophy and Cx43 expression in spontaneously hypertensive rats (SHR).METHODS:Nine-week old SHRs were randomly divided into two groups. Some received atorvastatin at 30 mg/kg by oral gavage once daily for 8 weeks (SHR-A); others received vehicle. Age-matched Wistar-Kyoto rats (WKY) received atorvastatin or vehicle for 8 weeks were used as controls. At the end of the experiment, we investigated LV hypertrophy and the expression of Cx43 in LV myocardium in four groups. Cx43 expression was investigated by the methods of Western blotting, immunohistochemistry, and transmission electron microscope. LV hypertrophy was accessed by pathological analysis and plasma brain natriuretic peptide (BNP) level.RESULTS:LV hypertrophy was prominent in untreated SHR. In SHR, LV myocardium Cx43 level was upregulated, and the distribution of Cx43 was displaced from their usual locations to other sites at various distances away from the intercalated disks. After atorvastatin treatment, myocardium Cx43 level was reduced in SHR-A, and the distribution of Cx43 gap junction became much regular and confined to intercalated disk. Statins also prevented LV hypertrophy in SHR.CONCLUSIONS:These results provide novel in vivo evidence for the key role of Cx43 gap junctions in LV hypertrophy and the possible mechanism in anti-hypertrophic effect of statins. Atorvastatin treatment may have beneficial effects on LV hypertrophy in spontaneously hypertensive rats.
Effects of Angiotensin II on Connexin 43 of VSMCs in Arteriosclerosis

Abstract:Objective: To observe the effect of angiotensin II (Ang II) on expression of gap junction channel protein connexin 43 (Cx43) in the proliferation process of vascular smooth muscle cells (VSMCs) during the early stage of arteriosclerosis. Methods: Thirty-two adult male rabbits were randomly divided into 4 groups. Rabbits in Group A were fed common diet while others in Groups B, C, and D were fed high-cholesterol diet. Losartan (10 mg/(kg·d)) and ramipril (0.5 mg/(kg·d)) were added in the diet of Groups C and D, respectively. The animals were sacrificed after 8 weeks and abdominal aortas were removed and dissected. The expression of Cx43 was assayed using RT-PCR and Western Blotting analysis. Results: Cx43 was increased markedly in both protein and mRNA level in Groups B, C, and D fed high-cholesterol diet compared with that in control group (P<0.01). Cx43 level in losartan or ramipril treated groups was higher than that in control group (P<0.01, P<0.05), but lower than that in high-cholesterol diet groups (P<0.05, P<0.01). Conclusion: Cx43 level was upregulated in VSMCs during early atherosclerosis. Losartan and ramipril can inhibit the expression of Cx43.

Abstract:Background: Gap junctions, which serve as intercellular channels that allow the passage of ions and other small molecules between neighboring cells, play an important role in vital functions, including the regulation of cell growth, differentiation, and development. Statins, the 3-hydroxy-3-methylglutaryl-coenzymeA (HMG-CoA) reductase inhibitors, have been shown to inhibit the migration and proliferation of smooth muscle cells (SMCs) leading to an antiproliferative effect. Recent studies have shown that statins can reduce gap junction protein connexin43 (Cx43) expression both in vivo and in vitro. However, little work has been done on the effects of statins on gap junctional intercellular communication (GJIC). We hypothesized in this study that lovastatin inhibits vascular smooth muscle cells (VSMCs) migration through the inhibition of the GJIC. Methods: Rat aortic SMCs (RASMCs) were exposed to lovastatin. Vascular smooth muscle cells migration was then assessed with a Transwell migration assay. Gap junctional intercellular communication was determined by using fluorescence recovery after photobleaching (FRAP) analysis, which was performed with a laser-scanning confocal microscope. Results: The migration of the cultured RASMCs were detected by Trans. well system. Cell migration was dose-dependently inhibited with lovastatin. Compared with that in the control (110 +/- 26), the number of migrated SMCs was significantly reduced to 72 +/- 24 (P < .05), 62 +/- 18 (P < .01), and 58 +/- 19 (P < .01) at the concentration of 0.4, 2, and 10 umol/L, per field. The rate of fluorescence recovery (R) at 5 minutes after photobleaching was adopted as the functional index of GJIC. The R- value of cells exposed to lovastatin 10 umol/L for 48 hours was 24.38% +/- 4.84%, whereas the cells in the control group had an R- value of 36.11% +/- 10.53%, demonstrating that the GJIC of RASMCs was significantly inhibited by lovastatin (P < .01). Smaller concentrations of lovastatin 0.08 umol/L did not change gap junction coupling (P > .05). Conclusions: These results suggest that lovastatin inhibits migration in a dose-dependent manner by attenuating JIC. Suppression of gap junction function could add another explanation of statin-induced antiproliferative effect.
Losartan Reduced Connexin43 Expression in Left Ventricular Myocardium of Spontaneously Hypertensive Rats

Abstract:Objective To assess the effect of angiotensin II type 1 (AT 1 ) receptor antagonist losartan on myocardium connexin43 (Cx43) gap junction (GJ) expression in spontaneously hypertensive rats (SHRs) and investigate possible mechanisms. Methods Sixteen 9-week-old male SHRs and 8 age-matched male Wistar-Kyoto (WKY) rats were included in this study. SHRs were randomly divided into two groups to receive losartan at 30 mg/(kg·d) by oral gavage once daily for 8 weeks (SHR-L) or vehicle (0.9% saline) to act as controls (SHR-V); WKY rats receiving vehicle for 8 weeks served as normotensive controls. At the end of the experiment, rats were sacrificed and the hearts were removed. Expressions of Cx43 and nuclear factor-kappaB p65 (NF-κB p65) proteins in all three groups were observed and further investigations on the effect of angiotensin II type 1 receptor antagonist losartan (30 mg/(kg·d), 8 weeks) on Cx43 expression were conducted with Western blot and immunohistochemistry. NF-κB p65 protein in nuclear extracts was determined by Western blot. Results Left ventricular (LV) hypertrophy was prominent in SHRs, Cx43 and NF-κB p65 protein expressions were obviously upregulated and Cx43 distribution was dispersed over the cell surface. Treatment with losarton reduced the over-expressions of Cx43 and NF-κB p65 in LV myocardium. The distribution of Cx43 gap junction also became much regular and confined to intercalated disk after losartan treatment. Conclusion Cx43 level was upregulated in LV myocardium of SHR during early stage of hypertrophy. Angiotensin II type 1 receptor antagonist losartan prevented Cx43 gap junction remodeling in hypertrophied left ventricles, possibly through the NF-κB pathway.
The Connexin43 expression in the role of Ginkg biloba inhibiting Formation of atherosclerotic plaques in rabbit

Abstract:Objective To study the effects of Ginkgo biloba on the blood fat and plaque and the expression of Connexin43 in artherosclerosis rabbits.Methods A total of forthy male rabbits were randomly divided into 5 group:rabbilts in control group were given normal diet,rabbits in other groups were given cholesterol diet.Rabbits in Simvastatin group were fed by Simvastatin,and Rabbits in low or high Ginkgo biloba group were fed by Ginkgo biloba.Serum lipid level,ratio of plaque area to aorta area and expression of Connexin43 were measured.Results Serum lipid level,ratio of plaque area to aorta area and expression of Connexin43 were reduced in rabbits in Ginkgo biloba group than in cholesterol group.Conclusion Our study show that Ginkgo biloba may lower Serum lipid level and reduce the degree of artherosclerosis in artherosclerosis rabbits,which were achieve by reducing expression of Connexin43 probably.
Inhibition of Atherosclerosis Progression, Intimal Hyperplasia, and Oxidative Stress by Simvastatin and Ivabradine May Reduce Thoracic Aorta’s Stiffness in Hypercholesterolemic Rabbits

Abstract:Aims: This study aims to evaluate atherosclerosis, oxidative stress, and arterial stiffness attenuation by simvastatin and ivabradine in hyperlipidemic rabbits. Methods and Results: Forty rabbits were randomly divided into 4 groups: atherogenic diet (group C), atherogenic diet plus simvastatin (group S), atherogenic diet plus ivabradine (group I), and atherogenic diet plus simvastatin and ivabradine (group S + I). After 9 weeks, rabbits were euthanized and descending aortas excised for mechanical testing. Atherogenic diet induced the development of significant atherosclerotic lesions in group C animals but in none of groups S, I, and S + I. RAM-11 and HHF-35–positive cells were significantly reduced in groups S, I, and S + I compared with group C ( P < .001). A significant neointimal hyperplasia and intima–media ratio reduction was demonstrated in groups S ( P = .015 and P < .001), I ( P = .021 and P < .001), and S + I ( P = .019 and P < .001) compared with group C. Protein nitrotyrosine levels were significantly decreased in group S compared with group C ( P = .009), and reactive oxygen species levels were decreased in group I compared with group C ( P = .011). Aortic stiffness was significantly reduced in groups S, I, and S + I compared with group C ( P = .003, P = .011, and P = .029). Conclusion: Simvastatin and ivabradine significantly inhibited intimal hyperplasia and oxidative stress contributing to aortic stiffness reduction in hyperlipidemic rabbits.

Abstract:Statins reduce atherosclerosis, but it is controversial whether they suppress abdominal aortic aneurysm (AAA) expansion. We hypothesized that statins (rosuvastatin and atorvastatin) would attenuate angiotensin II (AngII)-induced atherosclerosis and AAA.Sixty apoE-/- male mice fed a normal diet were administered with either rosuvastatin (10mg/kg/day) or atorvastatin (20mg/kg/day) through drinking water for 1 week prior to initiating 28-day AngII infusion (1000 ng/kg/min). Statins administration led to therapeutic serum concentrations of drugs. Administration of either rosuvastatin or atorvastatin exerted no significant effect on AngII-induced expansion of suprarenal diameter or area. However, atorvastatin significantly reduced AngII-augmented atherosclerotic lesion areas in intimas of both aortic arches and cross-sections of aortic roots (P<0.001). Atherosclerosis was attenuated independent of reductions in serum total cholesterol concentrations. Although serum MCP-1 and MIF concentrations were not changed by either statins, atorvastatin administration increased PPAR-α and -γ mRNA abundances and decreased NF-κB p50, p65, MCP-1 and TNF-α mRNA abundances in atherosclerotic lesions.This study demonstrated both statins failed to suppress AngII-induced AAA. In contrast, atorvastatin reduced AngII-induced atherosclerosis associated with no change in serum inflammatory markers but a shift to upregulation of anti-inflammatory status in lesions.
Simvastatin Reduces Expression and Activity of Lipoprotein-Associated Phospholipase A(2) in the Aorta of Hypercholesterolaemic Atherosclerotic Rabbits.

Abstract:Lipoprotein-associated phospholipase A(2) (Lp-PLA(2)) contributes to atherosclerotic plaque instability and subsequent sudden coronary death. Statins are associated with decreased stroke risk and may improve stability of atherosclerotic plaques. The present study investigated the effect of simvastatin on expression of Lp-PLA(2) levels in atherosclerotic plaques and on Lp-PLA(2) activity in atherosclerotic aortas. Rabbits were a fed chow (control group) or a high-cholesterol diet (atherosclerosis group) for 12 weeks. An additional group on the high-cholesterol diet received simvastatin (5 mg/kg per day) for the last 4 weeks (simvastatin group). Lp-PLA(2) activity in plasma and atherosclerotic aortas was significantly higher in the atherosclerosis group than in the control group and, consistent with this, abundant Lp-PLA(2) protein was detected in plaques in the atherosclerosis group. Simvastatin significantly reduced Lp-PLA(2) activity in plasma and aorta tissue, and reduced Lp-PLA(2) protein level in atherosclerotic plaques. Whereas there was no significant difference in total atherosclerotic lesion area between simvastatin and atherosclerosis groups, simvastatin significantly reduced macrophage content, lipid retention and the intima/media ratio but increased the content of smooth muscle cells in atherosclerotic lesions. Thus, statin treatment markedly reduced Lp-PLA(2) in both plasma and atherosclerotic plaques. This was associated with attenuation of the local inflammatory response and improved plaque stability.

Abstract:Statins have shown pleiotropic effects, many of them independent of their impact on lipids. Angiogenesis can be beneficial in the acute myocardial infarction to improve circulation. However, it also can be harmful due to worsening of atherosclerosis. Here, we established a new minimal invasive rabbit model to study ischemic myocardium and atherosclerosis together to mimic clinical scenario. We demonstrated that simvastatin has the effect of pro-angiogenesis and further improve cardiac function in ischemic myocardium, as well as the effect of anti-angiogenesis to improve atherosclerosis in aorta vessels.

Abstract:Objective To study the effect of atorvastatin on matrix metalloproteinase 9(MMP-9) expression in the upstream shoulder(proximal end) of aortic atherosclerotic plaques in rabbits.Methods Thirty male New Zealand rabbits were randomized into normal control group(n=9),high-lipid diet group(n=11) and atorvastatin group(n=10).An aortic atherosclerotic model was induced by high-lipid diet in the last two groups.From the ninth week,the high-lipid diet group rabbits were administered starch 1.5 mg/kg body weight per day,and the rabbits of atorvastatin group were administered atorvastatin 1.5 mg/kg body weight per day.After the rabbits were killed,the gross specimen of aorta was observed,and the rabbits whose intimal hyperplasia complicated with the formation of atherosclerotic plaque were regarded as the successful induction of the model.The percentage of macrophage and smooth muscle cell positive area in the upstream shoulder of plaques and the expression of MMP-9 were detected by immunohistochemistry methods.Results The formation of neointima was seen in aorta in the high-lipid diet and atorvastation groups,the ratios of intima-media thickness were 1.41±0.34 and 0.63±0.12,respectively.The distribution sites of macrophages in the plaques were in concordance with the region of positive expression of MMP-9,and they were all assembled at the upstream shoulder of the plaques.As compared with the high-lipid diet group,atorvastatin reduced aggregation of macrophages significantly(the percentages of positive areas were 26.5%±4.3% and 12.4%±1.5%,respectively) and expression of MMP-9(the values of optical density were 0.081±0.014 and 0.022±0.004,respectively) in the upstream shoulders of the aortic plaques.There were significant differences between the two groups(P<0.01).The percentages of positive areas of smooth muscle cells in the high-lipid diet and atorvastatin groups were 47.2%±12.3% and 50.4%±10.8%,respectively.There were no significant differences between the two groups(P>0.05).Conclusions The macrophages in the atherosclerotic plaque and the expressions of MMP-9 are mainly in the upstream shoulder.Atorvastatin may significantly reduce the aggregation of macrophages and the expression of MMP-9 in this area.

Abstract:Objective: The 5-lipoxygenase catalyzed formation of leukotriene lipid mediators is a mediator for inflammatory response in arteries. The present study investigated the relationship between atorvastatin and the 5-lipoxygenase pathway in an atherosclerotic rabbit model. Methods: Thirty male New Zealand White Rabbits were randomized into negative control, positive control and atorvastatin groups. At week 4, the rabbits were subjected to carotid balloon-dilation injury or carotid balloon-dilation injury, followed by treatment with atorvastatin. At week 12, all the animals were sacrificed. Plasma lipids, LTD4, and 15-epi-lipoxin A(4) were measured using the enzymatic endpoint method and ELISA, respectively. RT-PCR was performed to detect the gene expression of 5-lipoxygenase-activating protein and cysLT1R in rabbit carotid arteries. Finally, histological analysis was used to evaluate the pathophysiological changes of rabbit carotid arteries. Results: The results showed atorvastatin markedly lowered serum lipids and LTD4 levels compared with the control group. Similarly, mRNA expression of 5-lipoxygenase-activating protein and cysLT1R was significantly inhibited by atorvastatin. Decreased carotid plaque instability was evident in atorvastatin-treated animals, as demonstrated by a thickened elastic layer, less neointima hyperplasia and macrophage proliferation. Conclusions: Atorvastatin may stabilize carotid plaque by regulating the 5-lipoxygenase pathway in atherosclerotic rabbits and delay the progression of atherosclerosis by exerting anti-inflammatory effects. Abstract:Objective: To study the serum lipid (TC, TG, LDL-C) and the role of type I collagen, NF-κB, type I collagen mRNA and NF-κB mRNA levels and their mechanisms in the occurrence and progress of atherosclerosis in order to provide theoretical gist for its prevention. Methods: Blood vessel wall pathological and the serum lipid changes, the expressions of type I collagen, NF-κB, type I collagen mRNA and NF-κB mRNA levels were observed in dietary induced atherosclerosis rabbit model by morphology study, immunohistochemistry, in situ hybridization technique and color image analyzer (CMAIS). Results: The model of AS was established successfully after rabbits were fed with cholesterol, who developed the fatty streak lesion in the aortic intra-wall. In the rabbit model, TC and LDL were respectively increased 43 times and 37 times and there were no difference in TG and body weight. Type I collagen, NF-κB, type I collagen mRNA and NF-κB mRNA were 0.27 ± 0.02, 0.19 ± 0.01 and 0.30 ± 0.03, 0.35 ± 0.03, respectively, which were higher than those of control group(0.08 ± 0.01, 0.09 ± 0.01 and 0.11 ± 0.01, 0.10 ± 0.09, respectively) (P < 0.05). Conclusion: We understand further that hyperlipidemia is one of the uppermost causes of AS and provide theoretical gist for preventing AS. In other words, elevation of type I collagen in atherosclerotic lesions can stimulate NF-κB activity , which is one of the mechanisms causing AS.

Abstract:AIM:To evaluate the inhibitory effect of simvastatin via investigating the overall expression level of proteins in the artery of atherosclerotic rabbit.METHODS:Experimental model was established by feeding the rabbits a high fat diet (cholesterol 0.5 g.kg-1.d-1, lard 0.5 mL.kg-1.d-1) for 8 weeks. Then simvastatin (5 mg.kg-1) were fed for 4 weeks to the rabbits. The overall protein levels were measured using two-dimensional gel electrophoresis and a PDQUEST data processing.RESULTS:Twenty nine protein spots showed significant quantitative changes in comparison with the normal and the diseased rabbits. Furthermore, after the diseased rabbit having taken simvastatin, an obvious decay of symptom of fatty liver was observed, and the intensity of most spots has not been back-regulated.CONCLUSION:Simvastatin facilitates the metabolism of fat in the blood, but the lesion of the internal wall of the atherosclerotic artery cannot be restored.
Effects of Simvastatin in A Rabbit Model of Atherosclerosis: Role of Ho-1/Co-Cgmp Pathway and Related Anti-Oxidative Enzyme Expression

Abstract:Objectives Clinical and experimental observations indicated that HMG-CoA reductase inhibitors (statins) have pleiotropic effects. The present study was designed to elucidate the status of cyclic adenosine monophosphate (c-GMP), Cyclooxygenase-2 (COX-2), Carbon Monoxide Haemoglobin (COHb) and haem oxygenase-1 (HO-1) to the antoxidative effects of Simvastatin in the hypercholesterolemic rabbit. Methods Twenty four male Japanese white (JW) rabbits weighing approximately 2.0 kg were used in this study. The animals were randomised into three groups of eight animals each: one control group and other two groups which were maintained on high-cholesterol diet (HCD) for 24 weeks. After 8 weeks on this diet, the animals were divided into two groups: non-treated group and simvastatin group; the animals were euthanised with an overdose of sodium pentobarbital at the end 24th week. The aorta were excised, placed in ice-cold sterile saline, and cleaned of connective tissue. Blood was taken through cardiac puncture and plasma was used for biochemical assay of lipid peroxides. Levels of HbCO, COX-2 and cGMP were measured by Enzyme-Linked Immunosorbent Assay (ELISA) kits. HO-1mRNA in arterial were analysed using Real Time Quantitative PCR methods, and expression of MMP-9 protein was measured by immunohisto-chemical assay, serum malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were also measured. Results Twenty-four weeks of atherogenic diet significantly increased the levels of MDA cytotoxic (19.72±2.60 vs 14.24±1.87 ng/ml control group), and SOD (1.31±0.45 vs 7.92±1. μg/ml control), cGMP (18.66±1.47 vs 12.54±1.45 nmol/l control group) decreased in serum; HO-1mRNA expression of non-treated group is no different than the control group (1.75±1.02 vs 1.34±0.63), but is showing the tendency to increase (3.22±1.67vs 1.75±1.02 non-treated group, p=0.062) after treatment with Simvastatin. SOD, NO and cGMP were as well as increased (p<0.01 or p<0.05), but MMP-9 and HbCO (56.12±11.67 vs 101.0±41.55 µg/ml non-treated group) decreased significantly than the non-treated group (p<0.01 or p<0.05) after treatment with Simvastatin. Simvastatin partially restored the HO-1, SOD and NO, decreased MDA and HbCO levels, but to COX-2(67.69±6.39 vs 68.90±9.04 U/l non-treated group), no differences among groups were observed. Conclusions Atherosclerosis induced an oxidative damage and simvastatin had significant anti-atherogenic and anti-oxidation effects in an animal model with rabbits fed a hypercholesterolemic diet, maybe it mediated by HO-1, restoring the SOD and NO levels and reducing cytotoxic MDA levels.

Abstract:Objective To investigate the effect of simvastatin on expression of tissue inhibitor of matrix metalloproteinase 1(TIMP 1) in iliac artery of atherosclerotic rabbits Methods Eighteen male New Zealand white rabbits were studied The rabbits were fed on a normal diet ( n =5) or a cholesterol diet (cholesterol, 1 g/d; lard, 10 g/d; n =13) for 6 weeks; cholesterol fed rabbits underwent balloon injury of iliac arteries 2 weeks after initiation of the diet, 6 of whom were fed with simvastatin (15 mg/d per rabbit) after injury Four weeks after balloon injury, untreated and balloon injury iliac arteries were harvested for immunocytochemical staining, Western blot, and semi quantitative reverse transcription polymerase chain reaction (RT PCR) studies, respectively Results Simvastatin could remarkably lower plasma total cholesterol and inhibit the formation of atherosclerosis The relative values of arterial TIMP 1 mRNA in 3 groups were 1 57±0 11,2 72±0 10,3 33±0 28, respectively, with significant difference among them ( P 0 05) Western blot results were as follows: 2 72±1 16, 7 51±1 44, 16 90±1 45, respectively The expression of TIMP 1 antigen in simvastatin group increased significantly compared with baseline group( P 0 000 1) Conclusion Simvastatin can enhance expression of arterial TIMP 1 and inhibit the formation of atherosclerosis in iliac artery of atherosclerotic rabbits

Abstract:Objective To explore the effect of pravastatin on apoptosis in vulnerable plaques in abdominal aorta in model rabbits. Methods 30 New Zealand rabbits randomly divided into two groups were made as atherosclerosis plaques by a diet of high cholesterol and balloon-induced abdominal aorta wall injury. The rabbits were given a diet of high cholesterol as control group, the others were given a diet of high cholesterol and pravastatin as pravastatin group. The intima-media thickness (IMT) and the atherosclerosis plaques were measured with ultrasound imaging. The rabbits were killed 4 months after feeding to obtain the vulnerary plaques to detect the plaque apoptosis by TUNEL.Results The abdominal aorta IMT gradually increased, but that in pravastatin group were significantly smaller than that in control 1 month after operation (P<0.01). The apoptosis expressions in control group were mainly in lipid-rich core and fibrous cap, higher than that in other two areas (P<0.01). The apoptosis expressions focused on lipid-rich core, higher than that in other three areas (P<0.01), and that in fibrous cap in pravastatin group were lower than that in control (P<0.01).Conclusions IMT could be significantly related to degree of atherosclerosis, pravastatin could level off plaque through decreasing apoptosis in fibrous cap.
