EXPRESSION OF PTEN GENE IN HUMAN HEPATOCELLULAR CARCINOMA AND ITS CLINICAL SIGNIFICANCE

Abstract:Objective: To study PTEN expression in primary human hepatocellular carcinoma (HCC) and morbid tissues adjacent to carcinomas and its clinical significance. Methods: RT-PCR and immunohistochemistry method were used to detect PTEN-mRNA expression in HCC, morbid tissues adjacent to carcinomas, and healthy tissues, and PTEN-protein expression in HCC and morbid tissues adjacent to carcinomas. Result: PTEN-mRNA expression in HCC, morbid tissues adjacent to carcinomas, and healthy tissues decreased in turn, showing remarkable statistical significance(P<0.01); PTEN-protein expression appeared, however, significantly lower than morbid tissues adjacent to carcinomas(P=0.038). Analysis of its association with some clinicopathological features revealed that PTEN gene expression correlated well with Edmondson’s grade, portal tumor thrombus, and pathological grading(P=0.031, P=0.001, P=0.025). PTEN expression was higher for Edmondson’s grade I~Ⅱ than for grade Ⅲ; it was higher for none portal tumor thrombus than for portal tumor thrombus; it was higher for pathological grade I~Ⅱ than for grade Ⅲ~Ⅳ.Conclusion: PTEN gene mutation or loss of its expression can occur in progression of HCC. Loss of PTEN expression has close relations with the degree of malignancy of HCC, disease progression, and invasion and metastasis of HCC. The lower the PTEN gene expresses, the later stage the HCC patients are at, and the higher the degree of malignancy of liver cancers appears, the easier the invasion and metastasis tends to happen,the poorer post-prognosis will be.
