Abstract:Inter-simple sequence repeat (ISSR) marker technology has become a hotspot in DNA molecular technology in recent years, and plays an important role in genetic research. However, this technology is still at an initial stage in animal research, compared with its applications in plant research. This paper introduced the principles and main methods of ISSR marker technology, and summarized its applications in analyzing the genetic structure of animals, such as genetic diversity, resources identification, and genetic relationship, etc. The application prospects of this technology were also discussed, aimed to provide reference for the further applications of this technology in animal research.

Abstract:Cloning could not only be used to protect endangered animals,but also be applied for therapeutic cloning when combined with stem cell engineering technology.Thus it has great scientific significance and practical value.As pig somatic cloning has great potential in the aspect of human organ transplantation,it now becomes a research focus.However,there are still many difficulties,especially too many steps and low cloning efficiency.In this paper,the preparation of nuclear donor cells and oocytes,microinjection,activation of reconstruction embryos and several key steps influencing pig somatic cloning technology were reviewed.

Abstract:Exogenous genescan be introduced into the host genome chromosome, stably integrated and transmitted to future gen- erations by transgenic cloning technology. Transgenic cloning technology has been extensively applied to theoretical research of the gene expression and regulation, animal models establishment of human genetic diseases, pharmaceutical protein production, disease resistance breeding and the human organ transplant research. Transgenic animal research and application in the 21st century have become the most popular technology in the life sciences. In particular, as a bioreactor it can provide medical value of human organs. its economic value and social benefits will be incalculable. In this paper, we have discussed main technologies of transgenic animal cloning, such as mi- croinjection technology, nuclear transfer technology, gene targeting and eukaryotic expression vector of BAC,and its application in xeno-transplantation and building bioreactor. The advantages and disadvantages of transgenic technologies were discussed in details here, This paper was aimed to supply a basis for the transgenic animal cloning researches.

Abstract:Nuclear transfer and therapeutic cloning have widespread and attractive prospects in animal agriculture and biomedical applications. We reviewed that the quality of oocytes and nuclear reprogramming of somatic donor cells were the main reasons of the common abnormalities in cloned animals and the low efficiency of cloning and showed the problems and outlets in therapeutic cloning, such as some basic problems in nuclear transfer affected clinical applications of therapeutic cloning. Study on isolation and culture of nuclear transfer embryonic stem (ntES) cells and specific differentiation of ntES cells into important functional cells should be emphasized and could enhance the efficiency. Adult stem cells could help to cure some great diseases, but could not replace therapeutic cloning. Ethics also impeded the development of therapeutic cloning. It is necessary to improve many techniques and reinforce the research of some basic theories, then somatic nuclear transfer and therapeutic cloning may apply to agriculture reproduction and benefit to human life better.

Abstract:Somatic cell nuclear transfer offers exciting opportunities in many areas and brings tremendous changes to agriculture, conservation and therapeutics. However, the fundamental research of nuclear transfer including nuclear reprogramming is not fully understood. Some inappropriate epigenetic modifications have been reported in the clone animals, including abnormal regulation of DNA methyltransferase expression, improper expression of imprinted genes,reactivation of the inactive X chromosome. The low efficiency of nuclear transfer hinders the further progress in some degree, we describe some recent advances in applications, methods and main problems limiting efficiency.

Abstract:Somatic cell nuclear transfer (SCNT) shows great value in the generation of transgenic animals, protection of endangered animals, and stem cell therapy. The combination of SCNT and gene editing has produced a variety of genetically modified animals for life science and medical research. Rabbits have unique advantages as transgenic bioreactors and human disease models; however, the low SCNT efficiency severely impedes the application of this technology. The difficulty in SCNT may be attributable to the abnormal reprogramming of somatic cells in rabbits. This review focuses on the abnormal reprogramming of cloned mammalian embryos and evaluates the progress and prospects of rabbit somatic cell cloning.

Abstract:Both 'Human Genome Project(HGP)' and animal cloning from somatic cells will be listed as two important milestones in the history of sciences.The success of HGP started the new era of genomics with two major perceptions that life is in sequence and life is digital.Based on this belief,BGI has been focusing on sequencing and bioinformatics,aiming at analyzing genomes of everybody in the world and every(living)thing on Earth via vast collaborations.Since the birth of Dolly the sheep,successes have been made in cloning other mammals.With the main principle of reprogramming,somatic cell nuclear transfer(SCNT) is becoming the bond among genetic munipulation,stem cell and synthetic biology,making the 20th Century The Century of Biology.Based on the information obtained from genomics and improved technology of handmade cloning,BGI has established an animal cloning platform,which focuses on: 1)generating animal models for human diseases as a strong tool for studying the mechanism and developing therapy methods of human diseases;2)livestock improvement for making quality better.
