Abstract:An exhaustive literature survey on the secondary metabolites of widely distributed Prunella vulgaris L. species has been carried out. Triterpenoids, sterols, phenylpropanoids, flavonoids, coumarins, fatty acids, volatile oils and carbohydrates along with some other compounds, have been reported from this species. Many of these compounds have been found to possess significant biological properties, including antihypertensive activity, anticancer activity, anti-lipidemic activity, antibacterial and antivirus activities, anti-inflammatory activities, hypoglycemic effects, heptaprotective effects, antineoplastic activities, etc. P. vulgaris might offer a rich natural source with relative cheap price for developing new anti-neoplastic drugs.

Abstract:Prunella vulgaris L. (PVL) is dried fruit spike of Lamiacea plant Prunella vulgaris L., which is a perennial herb with medicinal and edible homology used for thousands of years. PVL is bitter, acrid, cold, and belongs to the liver and gallbladder meridians. It clears the liver and dissipate fire, improve vision, disperse swelling, and has satisfactory clinical therapeutic effects on many diseases such as photophobia, dizziness, scrofula, goiter, breast cancer. The collection of information and data related to PVL comes from literatures retrieved and collated from various online scientific databases (such as CNKI, VIP, PubMed, Web of Science, Research Gate, Science Database), ancient books of traditional chinese medicine (Encyclopedia of Traditional Chinese Medicine, Classics of Traditional Chinese Medicine, Dictionary of Traditional Chinese Medicine), and Doctoral and Master's Dissertations. Currently, the major chemical constituents isolated and identified from PVL are triterpenoids, steroids, flavonoids, phenylpropanoids, organic acids, volatile oils and polysaccharides. Modern pharmacological studies have shown that PVL has a wide range of pharmacological activities, including anti-inflammatory, anti-tumor, antibacterial and antiviral effects, as well as immune regulation, antihypertensive, hypoglycemic, lipid-lowering, antioxidant, free radical scavenging, liver protection, sedative and hypnotic effects. This paper reviewes the botany, ethnopharmacology, traditional application, phytochemistry, analytical methods, quality control, pharmacological effects of PVL. It can be used not only as medicine, but also gradually integrated into the "medicine and food homology" and "Chinese medicine health" boom. More importantly, it has great potential for drug resources development. This paper deeply discusses the shortcomings of current PVL research, and proposes corresponding solutions, in order to find a breakthrough point for PVL research in the future. At the same time, it is necessary to further strengthen the research on its medicinal chemistry, mechanism of action and clinical application efficacy in the future, and strive to extract, purify and synthesize effective components with high efficiency and low toxicity, so as to improve the safety and rationality of clinical medication.

Abstract:Cancer is one of the most serious threats to public health around the world. Efforts in developing new therapies and prevention strategies are clearly insufficient in the face of a dramatically rising disease burden worldwide. New evaluation of alternate strategies, including those based on traditional medicine, is increasingly needed. These therapeutic or prevention approaches could prove complementary to current medical practice or could potentially aid in the development of new classes of pharmaceutical drugs with anti-cancer properties. Prunella vulgaris, a perennial herb, is a representative Chinese herb that has been put into practice to treat various types of diseases, including cancer. The triterpenoid, flavonoids, and phenylpropanoids in P. vulgaris have shown a collective therapeutic effect against cancer mediated through multiple pathways. This review discusses the chemical constituents of P. vulgaris, summarizes all of the known formulas that contain P. vulgaris, and also discusses in vitro, in vivo, and clinical studies on the anti-tumor properties of P. vulgaris. The aim is to bring better insights regarding P. vulgaris as an effective complementary method for treating cancer. Highlighted throughout this review is the necessity for additional prospective clinical trials on anti-tumor properties of P. vulgaris.

Abstract:OBJECTIVE:To investigate the herbal medicine of Prunella vulgaris by textual researches, and provide a theoretical basis for clinical medication, exploitation and protection of wild P. vulgaris resources.METHOD:Textual research on medicinal works of past dynasties and field work were adopted.RESULT:The natural distributions of P. vulgaris were concentrated distribution in Sichuan province, Huaihe river basin, and Middle-Lower Yangtze river valley in Chinese history. The indications of P. vulgaris in ancient and modern times were basically identical. While there were difference between the medicinal parts, harvest period and processing methods existed difference between ancient and modern.CONCLUSION:Three periods that whole grass of P. vulgaris as medicinal parts (from late Ming dynasty to late Qing dynasty and early stage of Republic of China), both whole grass and spicas as medicinal parts (from mid-term Republic of China to 1963), and the semi-maturity or maturity of spicas as medicinal parts (from 1963 to today) existed. The processing method for medicinal parts of P. vulgaris adopted sun drying and shady drying in ancient China, but only the sun drying was only used in modern times.

Abstract:OBJECTIVE:To investigate the original plant and dietotherapy history of Prunella vulgaris by textual researches, and provide a theoretical basis for clinical medication, exploitation and protection of wild resources. METHOD:The ancient literatures in the graphic, geographical distribution and morphological description of P. vulgaris were used in this studied. RESULT:The varieties of P. vulgaris for therapeutic were no confusion. However, there has existed confusion between P. vulgaris and its confusable species (Ajuga ciliate, A. decumbens and Lagopsis supina) with appellations, morphological description and attached drawing of original plant. CONCLUSION:The original plant is originated from two species (P. vulgaris, P. asiatica). P. hispida was used as a certified medicinal material in Yunnan province from Ming Dynasty to modern time. The dietotherapy history of P. vulgaris in China can be traced back to Ming Dynasty or before.

Abstract:Objective To determine the main chemical components of the Prunella vulgaris L.collected from nine different places and carry out the principal component analysis and cluster analysis. Methods The polysaccharides and total flavonoids of Prunella vulgaris L. were determined by spectrophotometer and the rosmarinic acid was determined by HPLC.Results The content of polysaccharides,total flavonoids and rosmarinic acid in the Prunella vulgaris L.collected from nine different places was 6.81% ~ 12.49%,3.29% ~ 5.82%,and 1.0%-3.3% respectively.The polysaccharides proved to be the main chemical component of Prunella vulgaris L. The results of clustering analysis showed that the Prunella vulgaris L.with geographical proximity could be clustered together.Conclusion There are significant differences among the Prunella vulgaris L.collected from nine different places in the main chemical components,so it is suggested that the content of Prunella vulgaris L.should be considered in the quality the quality evaluation of Prunella vulgaris L.

Abstract:Objective To investigate the variation of chemical characteristics with environmental factors and establish a relationship between morphological characters and chemical composition of Prunella vulgaris collected in different areas of China. Methods Twelve phenotypic traits and three chemical compositions were assessed in 28 populations of P. vulgaris collected from different locations in China. Results The variability ranges observed at phenotypic and chemical levels were polymorphic. According to the morphological traits, 28 populations of P. vulgaris could be grouped into six clusters, and two morpho-types could be clearly distinguished. Perceptible differences could be discerned in the plant height, leaf length, corolla length, calyx length, fruiting spikes length, and maturity period. Based on three kinds of components including ursolic acid, total flavonoids, and total polysaccharides, all populations could be identified as four types. Cluster IV showing high content of ursolic acid, total flavonoids, and total polysaccharides could be utilized to develop superior derivatives. Conclusion The variation of chemical characteristics is influenced by the genetic and environmental factors, such as soil, climate, longitude, and altitude. It provides a solid basis for efficiently evaluating qualities and establishing good agricultural practices for P. vulgaris.

Abstract:Prunellae Spica is the dried spica of Prunella vulgaris belonging to Labiatae and it is widely used in pharmaceutical and general health fields. As a traditional Chinese medicine cultivated on a large scale, it produces a large amount of non-medicinal parts, which are discarded because they are not effectively used. To analyze the chemical constituents in the different samples from spica, seed, stem, and leaf of P. vulgaris, and explore the application value and development prospect of these parts, this study used ultrahigh performance liquid chromatography-tandem quadrupoles time of flight mass spectrometry(UPLC-Q-TOF-MS/MS) to detect chemical constituents in different parts of P. vulgaris. As a result, 117 compounds were detected. Among them, 87 compounds were identified, including 32 phenolic acids, 8 flavonoids, and 45 triterpenoid saponins. Some new triterpenoid saponins containing the sugar chain with 4-6 sugar units were found. Further, multivariate statistical analysis was conducted on BPI chromatographic peaks of multiple batches of different parts, and the results showed that spica had the most abundant chemical constituents, including salviaflaside and linolenic acid highly contained in the seed and phenolic acids, flavonoids, and triterpenoid saponins in the stem and leaf. In general, the constituents in the spica were composed of those in the seed, stem, and leaf. UPLC was used to determine the content of 6 phenolic acids(danshensu, protocatechuic acid, protocatechuic aldehyde, caffeic acid, salviaflaside, and rosmarinic acid) in different parts. The content of other phenolic acids in the seed was generally lower than that in the spica except that of salviaflaside. The content of salviaflaside in the spica was higher than that in the stem and leaf, but the content of other phenolic acids in the spica was not significantly different from that in the stem. The content of protocatechuic aldehyde and caffeic acid in the spica was lower than that in the leaf. DPPH free radical scavenging method was used to detect the antioxidant activity of four parts, and there was no significant difference in the antioxidant activity between the spica and the stem and leaf, but that was significantly higher than the seed. Moreover, the antioxidant activity of these parts was correlated with the content of total phenolic acids. Based on the above findings, the stem and leaf of P. vulgaris have potential application value. Considering the traditional medication rule, it is feasible to use the whole plant as a medicine. Alternatively, salviaflaside, occurring in the seed, can be used as a marker compound for the quality evaluation of Prunellae Spica, if only using spica as the medicinal part of P. vulgaris, as described in the Chinese Pharmacopoeia(2020 edition).

Abstract:Water-soluble polysaccharides from Prunella vulgaris Linn (P. vulgaris) were fractionated using DEAE-Sepharose fast-flow column to obtain several eluents of water (PV-P1), 0.1M NaCl (PV-P2) and 0.2M NaCl (PV-P3). Structural analyses showed that PV-P1 had a higher molecular weight and degree of branching as compared to PV-P2 and PV-P3. Tertiary structure analyses indicated that PV-P1, PV-P2 and PV-P3 did not have triple-helical conformation. PV-P2 and PV-P3 showed stronger antioxidant activities than PV-P1, as measured radical scavenging capacities. PV-P1 showed stronger immunomodulatory activities than PV-P2 and PV-P3 in term of stimulation of the production of pro-inflammatory cytokines, including nitric oxide (NO), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6) in murine macrophage RAW 264.7 cells. PV-P1, PV-P2 and PV-P3 did not exhibit cytotoxicities against RAW 264.7 at the concentrations tested. These results suggest that P. vulgaris polysaccharides could be explored as potential antioxidant and immunomodulatory agents for the complementary medicine or functional foods.

Abstract:Prunella vulgaris L. (Labiatae) contains a variety of structurally diverse natural products, primarily rosmarinic acid (RA), ursolic acid (UA) and oleanolic acid (OA), which possess a wide array of biological properties. In the present study, P. vulgaris was harvested at three developmental stages (vegetative, full-flowering and mature-fruiting stages), dissected into stem and leaf tissues and assayed for chemical contents using high performance liquid chromatography. Significant changes in the concentrations of the major secondary metabolites (RA, UA and OA) were observed at the different development stages. The highest concentrations of RA, UA and OA were found at the full-flowering stage (15.83 mg/g dry weight (DW) RA, 1.77 mg/g DW UA and 0.65 mg/g DW OA). Among the different aerial parts of the plant, the concentrations of RA, UA and OA were higher in the leaves than in the stems at the different developmental stages. These results suggest that the full-flowering stage is characterized by the highest concentrations of bioactive compounds. Therefore, this stage may be the optimum point for harvesting P. vulgaris plants. In additional, the leaves of P. vulgaris demonstrated higher RA, UA and OA concentrations than the stems, suggesting higher utilization potential.
Accumulation of Total Flavonoids at Reproductive Stage and Antioxidant Activity of Prunella Vulgaris Linn.

Abstract:The research was aimed to investigate the accumulation of total flavonoids in different organs and antioxidant activities in different extracts of Prunella vulgaris Linn.,which would provide theoretical basis for further research.The total flavonoids contents in different organs during different periods were measured at reproductive stage.Total flavonoids content was investigated by spectrometric method.The powder of P.vulgaris was extracted with 70% ethanol(in water,v/v) using ultrasonic extraction,and then the filtrate was extracted followed with ethyl acetate,n-butanol and water at equal volume.The extracts of different solvents could be gotten.The antioxidant activities of different extracts were evaluated and BHT was used as a positive control.The results showed that:the total flavonoids content of roots,steams and leaves of P.vulgaris increased with plant growth and reached the peak at flower-perished period,while that of the spikes reached the peak at flowers-blooming period.The total flavonoids content of leaves was the highest one among all the organs.Scavenging abilities on DPPH· of the extracts and BHT were in descending order:ethyl acetaten-butanolBHTwater.Scavenging abilities on ·OH of the extracts and BHT were in descending order:BHTethyl acetaten-butanolwater.There was strong correlation between antioxidant activity and total flavonoids content in different extracts of P.vulgaris.The flavonoids of P.vulgaris have great antioxidant activity,and they can be obtained by processing the leaves or the whole herb which were collected in flower-perished period or spike-dry period.
[Research progress in chemical constituents in plants of Polygonatum and their pharmacological effects]

Abstract:Polygonatum plants are perennial plants of Liliaceae. There are about 60 species reported at home and abroad,32 species in China,mainly distributed in the north temperate zone. Their main chemical components are steroidal saponins,polysaccharides,flavonoids,alkaloids,etc. They have anti-aging,anti-tumor,immunomodulation,antibacterial,antiviral,hypoglycemic and blood lipid effects. With the development of health industry,Polygonati Rhizome used as medicine and food has attracted great attention in recent years,and has become a research hotspot. However,the material basis of its efficacy is unclear and the product quality is uneven,which seriously limited the rapid upgrading of the industry. This review summarizes Polygonatum plants system classification,the chemical composition and pharmacological activity to provide theoretical basis for the development and utilization of Polygonatum plants.

Abstract:Angelicae Pubescentis Radix (APR) is a widely used antirheumatic Chinese medicinal herb known as "Duhuo" in China. It has the effects of dispelling wind and removing dampness, diffusing impediment, and relieving pain, and is mainly indicated for rheumatic arthritis with pain in the lower back and knees, and headache. To the best of our knowledge, an attempt is made to provide an up-to-date review on these aspects based on published materials, including ancient and modern books; Master's and doctoral theses; monographs on medicinal plants; the pharmacopoeia of different countries, websites for publication of patent and electronic databases, such as SCI finder, PubMed, Web of Science, ACS, Science Direct, Wiley, Springer, Taylor, CNKI, and Google Scholar. APR, which has a good clinical effect, has been used for traditional Chinese medicine more than 2000 years. Since 1957, a variety of chemical constituents have been reported from the medicinal plants of this herb, mostly coumarins and volatile oil. In the past 30 years, numerous studies have shown that the extracts and compounds isolated from APR showed effective analgesic and anti-inflammatory actions, also showing well effects on central nervous system, effects on cardiovascular system and deworming activity. In addition, we also present and discuss the botany, traditional medicinal use, pharmacokinetics, toxicity, quality control, future trends and prospects of APR. All this information suggest that future research of APR should be supplemented in the area of pharmacology and toxicology to provide further insight on the clinical use and quality control.
