Differentiation and therapeutic effects of C17.2 neural stem cells after transplanted into hippocampus of Aβ_(1-40)-injuried rats

Abstract:Objective To explore the differentiation and therapeutic effects of immortalized C17.2 neural stem cells in the rat model of Alzheimer’s disease (AD). Methods Pre-aggregated Aβ1-40 was microinjected into the dentate gyrus of rat hippocampus to mimic AD, and C17.2 neural stem cells were transplanted into the vicinity of Aβ1-40 2 weeks later. The survival and differentiation of grafted cells were observed with immunofluorescence staining. Learning and memory abilities of animals were evaluated by Morris water maze test. Results The grafted cells survived and migrated extensively. Most C17.2 cells differentiated into GFAP-positive glia and some into NF200-positive neurons with long processes into the host brain. Learning and memory of AD rats were significantly improved after C17.2 neural stem cell transplantation. Conclusion The satisfactory survival, differentiation and improvement on learning and memory indicate the therapeutic potential of immortalized neural stem cells for neuronal replacement.
