Abstract:Enhanced biological phosphorus removal(EBPR) is one of the most economical and sustainable process options for phosphate removal.However,deterioration or even complete failure of EBPR could happen because of the proliferation of glycogen-accumulating organisms(GAO).The anaerobic and aerobic GAO metabolic mechanisms were reviewed,and especially the microbial characteristics of two groups of GAO(Competibacter and Defluviicoccus vanus related organisms) were introduced,and the key factors were analyzed,such as carbon source,pH and temperature,that affect the growth of GAO.Based on the review,some future research directions were suggested.

Abstract:The overgrowth of the glycogen accumulating organisms(GAOs) has become one of the main reasons causing the unsteady operation of enhanced phosphorus removal(EBPR) processes.Therefore,the metabolic mechanism of GAOs as well as their microbial morphology and community is introduced for better understanding the exact microbial community of EBPR.GAOs can use synthetic glycogen as the energy source to assimilate volatile fatty acids(VAFs) anaerobically,resulting in a competition with phosphorus accumulation organisms(PAOs) for VAFs.Also,they will become the dominate population under un-identified conditions by outcompeting PAOs,ultimately leading to the failure of EBPR.The studies show that the influent organic composition,P/C,pH,temperature and nitrites are the key factors impacting the competition between PAOs and GAOs.Within the certain ranges of the above operational parameters,the accumulation of GAOs occurs and the PAOs activities will be completely inhibited.

Abstract:Glycogen-accumulating organisms (GAOs) are recognized as competitors of phosphorus-accumulating organisms (PAOs) in enhanced biological phosphorus removal (EBPR) systems. However, different types of GAOs exhibit unique metabolic traits in different environments. To characterize the metabolic activity of GAOs, two distinct modes were constructed. Long-term operation with the mode of anaerobic post-drainage demonstrated the successful establishment of a rapid and enduring GAOs system dominated by Candidatus Contendobacter community. Acetate was completely consumed within 60 min of the anaerobic phase. Concurrently, glycogen levels decreased by 0.062 g Glycogen/g SS, while intracellular carbon storage of poly-hydroxybutyrate (PHB) and poly-hydroxyvalerate (PHV) increased by 0.027 g PHB/g SS and 0.007 g PHV/g SS, respectively. Various concentrations of phosphorus loads illustrated the resistance of the system to high phosphorus loads, while maintaining glycogen accumulating metabolism (GAM). Under complete phosphorus limitation, no correlation between glycogen and acetate was observed during the initial 140 d. The addition of 1 mg/L phosphate to the influent preserved the typical GAM characteristics. Moreover, the pathway involved in glycogen conversion to trehalose may be pivotal in enhancing anaerobic glycogen degradation. The complete tricarboxylic acid cycle ensured efficient energy production, thereby sustaining the foundational metabolism. Notably, sucC and mdh were critical regulators of energy flux. This study provides a distinctive metabolic profile of a robust and resilient GAOs model with Ca. Contendobacter .

Abstract:Glycogen accumulating organisms (GAOs) are closely related to the deterioration of enhanced biological phosphorus removal systems. However, the metabolic mechanisms that drive GAOs remain unclear. Here, the two-thirds supernatant of a reactor were decanted following the anaerobic period to enrich GAOs. Long-term monitoring demonstrated that the system was stable and exhibited typical characteristics of GAOs metabolism. Acetate was completely consumed after 60 min of the anaerobic phase. The level of glycogen decreased from 0.20 to 0.14 g/gSS during the anaerobic phase, whereas the level of glycogen significantly increased to 0.21g/gSS at the end of the aerobic period. Moreover, there was almost no phosphate release and absorption in the complete periods, thus confirming the successful construction of a GAOs enrichment system. Microbial community analysis demonstrated that Ca. Contendobacter was among the core functional genera and showed the highest activity among all of the communities. Furthermore, our study is the first to identify the involvement of the ethyl-malonyl-CoA pathway in the synthesis of polyhydroxyvalerate via croR, ccr, ecm, mcd, mch and mcl genes. The Embden-Meyerhof-Parnas (EMP) pathway was preferentially used via glgP. Furthermore, the glyoxylate cycle was the main source of ATP under anaerobic conditions, whereas the tricarboxylic acid cycle provided ATP under aerobic conditions. aceA and mdh appeared to be major modulators of the glyoxylate pathway for controlling energy flow. Collectively, our findings not only revealed the crucial metabolic mechanisms in a GAOs enrichment system but also provided insights into the potential application of Ca. Contendobacter for wastewater treatment.

Abstract:A large number of phosphorus accumulative organisms(PAOs) and ordinary heterotrophic organisms(OHOs) exist in biological phosphorus removal system.There may exist substrate competition between the above organisms.In this paper,competition between(OHOs) and PAOs on carbon under anoxic condition is investigated.Although kinetic performance of OHOs and PAOs appears irrelevant,inhibition of acetic acid on anoxic polyphosphate storage is found according to batch test designed.The phenomenon is inconsistent with common understanding of denitrifying phorsphorus removal.According to the study,a part of ASM2D model describes the reaction kinetics has been improved.The measured data show that the improved model can better describe biochemical activity of PAOs under anoxic conditions.

Abstract:Stable glycogen accumulating organisms (GAOs) granules were cultivated in three sequencing batchreactors(SBR), fed with acetate, glucose and glucose-acetate as organic substrate respectively, by means of speciaalternating anaerobic-aerobic operation mode, i.e. withdrawal after anaerobic period and sludge aeration during aerobicperiod to suppress the activity of phosphate accumulating organisms(PAOs). Analyzing the variation of organicsphosphate, intracellular glycogen and PHB in typical cycle verified that the sort of organic substrate had marked influenceon the energy utilization mode, organics uptake rate and intracellular storage polymer sort of GAOs. Moreover, the initiaintracellular glycogen content was the key factor influencing the organics uptake amount and uptake rate. whenintracellular glycogen content was below 0.05g/g SS, anaerobic organics uptake efficiencies appeared to be dependent onglycogen content, and organics uptake efficiencies tended to be stable as glycogen content was higher than 0.05g/g SS. Inaddition, anaerobic organics uptake rate was in great difference with different glycogen content, during fast uptake periodin initial anaerobic stage, anaerobic organics uptake rate was increased with intracellular glycogen content increased.

Abstract:The proliferation of glycogen accumulating organisms(GAOs)has been regarded as one of the main reasons causing the deterioration of enhanced biological phosphorus removal(EBPR)systems.To better understand the competition mechanism between the phosphorus accumulating organisms(PAOs)and GAOs,the glycogen metabolism of PAOs is presented as well as the metabolism of GAOs under the anaerobic and aerobic phases,based on the biological phosphorus metabolism of Activated Sludge Model No.2.A number of factors,such as substrates,pH,temperature and sludge retention time(SRT),were found to have significant impact on the stoichiometry and dynamic coefficients of GAO and PAO metabolism,which subsequently determine the outcome of the competition between PAOs and GAOs.With the presented metabolism models,a more exact competition mechanism between PAOs and GAOs can be evaluated when EBPR processes operate under more extensive conditions,e.g.influent C/P,pH,SRT and temperature.Then,an effective strategy can be created and adopted to maintain the steady operation of the EBPR systems once the accumulation of GAOs occurs.Most research so far has focused on the anaerobic competition metabolism of PAOs and GAOs,so the aerobic/anoxic competition metabolisms for these two organism groups may be considered subjects for further research.

Abstract:In this paper, a study was conducted on the effect of polyhydroxyalkanoates (PHA) and glycogen transformations on biologic nitrogen and phosphorus removal in low dissolved oxygen (DO) systems. Two laboratory-scale sequencing batch reactors (SBR1 and SBR2) were operating with anaerobic/aerobic (low DO, 0.15–0.45 mg·L −1 ) configurations, which cultured a propionic to acetic acid ratio (molar carbon ratio) of 1.0 and 2.0, respectively. Fewer poly-3-hydroxybutyrate (PHB), total PHA, and glycogen transformations were observed with the increase of propionic/acetic acid, along with more poly-3-hydroxyvalerate (PHV) and poly-3-hydroxy-2-methyvalerate (PH2MV) shifts. The total nitrogen (TN) removal efficiency was 68% and 82% in SBR1 and SBR2, respectively. In the two SBRs, the soluble ortho-phosphate (SOP) removal efficiency was 94% and 99%, and the average sludge polyphosphate (poly-P) content (g·g-MLVSS −1 ) was 8.3% and 10.2%, respectively. Thus, the propionic to acetic acid ratio of the influent greatly influenced the PHA form and quantity, glycogen transformation, and poly-P contained in activated sludge and further determined TN and SOP removal efficiency. Moreover, significant correlations between the SOP removal rate and the (PHV + PH2MV)/PHA ratio were observed ( R 2 >0.99). Accordingly, PHA and glycogen transformations should be taken into account as key components for optimizing anaerobic/aerobic (low DO) biologic nitrogen and phosphorus removal systems.

Abstract:The mechanisms of biochemical reactions involved in the enhanced biological phosphorus removal (EBPR), using three different substrates (acetate, propionate and glucose) as the carbon source, were presented focusing on: the energy and reducing power of phosphate accumulating organisms (PAOs), glycogen accumulating organisms (GAOs) and latic acid producing organisms (LPOs) under anaerobic and aerobic conditions, formation and composition of PHA, metabolic pathways of glycogen and lactic acid (L-type). Although stable and good EBPR performance may result by proper control of the substrate conditions, the present metabolism models can not represent all biochemical processes equally as well. Further research in pure culture of PAOs and relative activity of enzyme in the biochemical metabolism were recommended.

Abstract:A high phosphate (P) recovery concentration was achieved in pilot-scale biofilm sequencing batch reactor (BSBR) with a low carbon source (C) cost. Especially, a high-abundance glycogen-accumulating organisms (GAOs) (13.93–31.72%) was detected that was accompanied by a high P recovery concentration of BSBR. High-abundance GAOs obtain additional C through various C compensation pathways (split tricarboxylic acid cycle (TCA cycle), glyoxylate shunt and gluconeogenesis), thus reducing the need to compete with polyphosphate-accumulating organisms (PAOs) for C and weakening the adverse effects on P recovery by PAO cells. Under the action of N -acyl homoserine lactones (AHLs)-mediated quorum sensing (QS), GAOs promoted the secretion of a large amount of extracellular polymeric substances (EPS), which helped to realize the P recovery of EPS-dominated biofilms (68.02%–96.89%). This study provides a low-carbon technology for the recovery of high concentration P from municipal wastewater, and improves the ecological theory of P recovery in collaboration with GAOs and PAOs.

Abstract:Glycogen accumulating organisms (GAO) compete the carbon substrates with phosphorus accumulating organisms (PAO), which potentially lead to the upsets and deterioration of the EBPR process performance. To optimize the EBPR activated sludge system effective and robust, the integrated affecting factors consisting of temperature, carbon sources, pH, other minor factors and their interaction effect should be focused on the competition of PAO and GAO in EBPR systems. To improve the EBPR system stable and reliable, the control approaches should be matched to favor development of PAO and to suppress GAO. Hence, the factors affecting the competition of PAO and GAO in EBPR activated sludge system and relevant implementation of control strategies are discussed in this paper.

Abstract:Anaerobic HRT(AHRT) is a key factor of anaerobic-aerobic(A/O) unit for phosphorus(P) removal.In most cases,AHRT is determined empirically by designers.In this paper,the statistics of P removal,and changes of polyhydroxyalkanoates(PHAs) and glycogen content in P accumulating organisms(PAOs) under different AHRT were carried out on the basis of stable operation of lab continuous flow A/O P removal system to investigate the enhanced biological P removal metabolic process by a batch test.Anaerobic decomposition of PHAs and replenishment of glycogen were found during the test.To explain the above phenomenon,a new metabolic mode of PAOs was proposed.

Abstract:Lower temperature or polyphosphate limitation is the favorable condition to enrich polyphosphate-accumulating organisms (PAOs) or glycogen accumulating organisms (GAOs) in biological phosphorus removal process. In this study,a sludge highly enriched in PAOs was used to investigate the metabolic pathways and carbon transformation in batch tests under the two conditions for elucidating the contradiction. The results showed that the PAOs are able to take up acetate and store it as polyhydroxyalkanoates (PHA) in anaerobic phase although the poly-P pool is emptied through washing with negligible P release. The stoichiometric ratios of the PHB and PHV production and glycogen degradation to the acetate uptake combined with the Nile blue staining indicated that the PAOs possess the anaerobic metabolism of GAOs using glycogen as the sole energy source,but no transformation of PAOs to GAOs is observed. After reconverting to the normal operation,phosphorus uptake and release are recovered the same as the parent reactor.

Abstract:Glycogen‐accumulating organisms (GAO) have the potential to directly compete with polyphosphate‐accumulating organisms (PAO) in EBPR systems as both are able to take up VFA anaerobically and grow on the intracellular storage products aerobically. Under anaerobic conditions GAO hydrolyse glycogen to gain energy and reducing equivalents to take up VFA and to synthesise polyhydroxyalkanoate (PHA). In the subsequent aerobic stage, PHA is being oxidised to gain energy for glycogen replenishment (from PHA) and for cell growth.
