Abstract:A series of (E)-3-Aryl-2-(3,4,5-trimethoxybenzoyl) acrylonitrile derivatives (5a-5i) were designed, synthesized and characterized by H-1 NMR and C-13 NMR spectroscopy. These newly synthesized compounds were evaluated for their in vitro anticancer activity against seven human cancer cell lines (HepG2, A549, MCF-7, A2780S, SKOV3, HT29 and CHL-1) which got an insight in structure-activity relationship (SAR). Among the series, compounds 5c, 5g and 5i displayed potent anticancer activity against different human cancer cell lines. In particular, 5g with a methoxyl group at the para position of the right phenyl ring exhibited the most potent anticancer activity against HepG2, A549, MCF-7, A2780S, SKOV3, HT29 and CHL-1 cell lines (IC50 = 1.20, 2.09, 1.84, 1.88, 1.27, 1.28 and 1.22 mu M, respectively). Furthermore, molecular docking studies supported the biological assay data and suggested that 5g may be a potential antitubulin agent. These results indicated that this class of compounds had potential for further development as anticancer agents.
