Effects of Hyperbaric Oxygenation on Concentration of Tumor Necrosis Factor-α in Rats CNS after Unsafe Fast Decompression

Abstract:Objective To investigate the changes of activity of microglial cell and content of tumor necrosis factor-α(TNF-α) in rat’s central nerve system(CNS) after being injured by unsafe fast decompression, and the effects of hyperbaric oxygenation. Methods Unsafe fast decompression animal model was constructed by exposing male SD rats to 1.0MPa for 5.5min and then fast decompression in 50s. HBO inducted 6h later. TNF-α immunopositive cells were shown by immunohistochemistry. Concentrations of TNF-α in brain and spinal cord were determined by ELISA. Results There was no TNF-α positive cell in control group and safe decompression group. Positive cells could be seen in 6h after fast decompression and summated in 24h-48h (P0.01). HBO treatment decreased the amount of TNF-α positive cell in CNS (P0.05,P0.01). The positive cells were mainly distributed in spinal cord gray matter, cerebral cortex and hippocampus. Concentrations of TNF-α were increased significantly after fast decompression and summated in 48h (P0.01). The concentration of TNF-α in spinal cord was higher than that of brain (P0.01). HBO treatment decreased the content of TNF-α in cerebrum and spinal cord(P0.05 or P0.01). Conclusion CNS injury by unsafe fast decompression activated the synthesis and secretion of TNF-α and increased the content of TNF-α in central nerve tissue. HBO therapy was supposed to restrain microglial reaction, reduce content of TNF-α in central nerve tissue, which may contribut to the mechanism of its therapeutic effectiveness.
