The effects of premature light exposure on the development of new retinal vessels in a mouse model of oxygen-induced retinopathy

Abstract:Objective To investigate the effects of premature light exposure on retinopathy which was induced by oxygen in a mouse model.Methods Forty-eight healthy C57BL/6J mice were randomly divided into 4 groups:12 mice in the control group,12 mice were raised in a box with 75% oxygen from P7(postnatal day 7) to P12,the right eyelids of another 12 mice were surgically opened on P4 to be exposed to light prematurely,and the eyelids of the last 12 mice were opened on P4 and then raised in a box with 75% oxygen from P7 to P12.The eyeballs of all of the mice were removed on P27.The development of the retinal vessels was observed by ADPase histochemisty and the proliferative neovascular response was quantified by counting the nuclei of new vessels extending from the retina into the vitreous in serial sagittal cross sections.Results New retinal vessels could not be observed in the control group and in the mice with prematurely opened eyelids,but could be clearly observed in the oxygen-induced retinopathy group.There were fewer nuclei in new vessels extending from the retina into the vitreous in mice raised in high-concentration oxygen with prematurely opened eyelids than in those without premature lidopening.Conclusion The retina of mice raised in high-concentration oxygen engendered neovascularization.Premature light exposure can weaken the effect of a high concentration of oxygen.
