Effects of Inhibiting the Expression of Gene Periodl on Morphine-induced Reward and Expression of DBI mRNA in Mice

Abstract:Objective:To investigate the morphine conditioned place preference (CPP) and changes of morphine induced expres- sion of diazepam binding inhibitor mRNA in mice following inhibition of Periodl (per1) expression by using the deoxyribozyme 164 (DRz164) targeting porl mRNA.Method:Male mice were randomly divided into three groups:A,lipofectamine+saline intracere- broventricularly (i.c.v.) injected with vehicle (containing lipofectamine and saline);B,lipofectamine+morphin;C,Lipofectaminet DRzI64 + morphine i.c.v.injected with DRz164 2 d before conditioning.During the first 3 sessions of conditioning,the mice were i. c.v.injected with vehicle or DRzI64 before subcutaneous (sc) injection of morphine.The control group was given i.c.v.injection vehicle and s.c.injection saline instead of morphine.Mice were sacrificed after CPP was tested,and then DBI mRNA in cortex was examined by in situ hybridization.Result:Pretreated with DRz164,mice did not acquire morphine CPP;Pretreatment with DRz164 significantly attenuated the expression of DBI mRNA in cortex in mice,it decreased by 65.41%.Conclusion:It indicated that perl played an important role in morphine reward using a CPP procedure,as well as it may be associated with the expression of DBI mRNA in mice cortex.
