Characterization and Identification of Triterpenoid Saponins in Crude Extracts from Clematis Spp. by High-Performance Liquid Chromatography/Electrospray Ionization with Multi-Stage Tandem Mass Spectrometry

Abstract:High-performance liquid chromatography coupled with electrospray ionization multi-stage tandem mass spectrometry (HPLC/ESI-MS11) was applied to characterize and identify triterpenoid saponins in crude extracts from nine species of Clematis L. After separation on a Zorbax SB-C-18 column, negative ESI-MS experiments were performed. The quasi-molecular ions [M-H](-), [M+Cl](-) and [M+HCOO](-) were observed in the full-scan MS spectra of all compounds. The MSn (n = 2-4) data of the [M-H](-) ions provided structural information on the sugar sequence of the oligosaccharide chains and on the aglycone of the saponins. In addition, the fragmentation mechanisms could be deduced from the fragment ions. As a result, eight saponins were unambiguously identified in C. ganpiniana by comparison with reference compounds. In addition, a new compound was tentatively identified as 3-O-ribopyranosyl -> rhamnopyranosyl -> (glucopyranosyl) -> arabinopyranosyl hederagenin 28-O-rhamnopyranosyl -, glucopyranosyl -> glucopyranosyl ester (peak 1), and another one was tentatively deduced to be 3-O-glucopyranosyl -> ribopyranosyl -> rhamnopyranosyl -> arabinopyranosyl hederagenin 28-O-rhamnopyranosyl up arrow glucopyranosyl -> glucopyranosyl ester (peak 5) from the genus Clematis L. for the first time. By ESI-MSn, non-isomeric saponins could be discriminated rapidly. It is of interest that cleavage preferentially occurrs at the ester bond at C-28 and the charge is easy to transfer onto the oligosaccharide chain when the ester bond of a monodesmosidic saponin like HNH cleaves. Abstract:Four new triterpene saponins, clematomandshurica saponins A-D (1-4), together with three known saponins (5-7) have been isolated from the roots and rhizomes of Clematis mandshurica. Abstract:OBJECTIVE To study the constituents in the whole plants of Clematis puberula Hook.f.& Thoms.var.ganpiniana(Lévl.& Van.) W.T.Wang.METHODS Column chromatography(including D101 macroporous resin,silica gel and Sephadex LH-20) and reversed-phase preparative HPLC were used to separate constituents whose structures were elucidated by ESI-MS and NMR spectral data.RESULTS Seven triterpenoid saponins were isolated,along with one lignan,and elucidated as HN saponin H(Ⅰ),huzhangoside B(Ⅱ),hederacholichiside F(Ⅲ),clematichinenoside B(Ⅳ),huzhangoside D(Ⅴ),clematichinenoside C(Ⅵ),prosapogenin CP11(Ⅶ) and clemastanin B(Ⅷ).CONCLUSION All these compounds were isolated from Clematis puberula Hook.f.& Thoms.var.ganpiniana(Lévl.& Van.) W.T.Wang for the first time,with compound Ⅲ isolated for the first time from the genus Clematis and compound Ⅶ as a genuine saponin from plants.

The structure of the new saponin was elucidated as 3-O-beta-[(O-alpha-L-rhamnopyranosyl-(1 --> 6)-O-beta-D-glucopyranosyl-(1 --> 4)-O-beta-D-glucopyranosyl-(1 --> 4)-O-beta-D-ribopyranosyl-(1 --> 3)-O-alpha-L-rhamnopyranosyl-(1 --> 2)-alpha-L-arabinopyranosyl)oxy]olean-12-en-21alpha-hydroxy-28-oic acid-O-alpha-L-rhamnopyranosyl-(1 --> 4)-O-beta-D-glucopyranosyl-(1 --> 6)-beta-D-glucopyranosyl ester (1) by spectral analysis and chemical methods. The effects of two major saponins (clematichinenosides AR and AR(2)) on the secretion of TNF-alpha in murine peritoneal macrophages induced by lipopolysaccharides were further investigated. The result indicated that a majority of triterpenoid saponins of this herb may be useful in the exploration of lead compounds for the treatment of some autoimmune diseases.

Abstract:Selected species of the genus Clematis (Ranunculaceae) have been screened for occurrence of triterpenoid saponins by qualitative HPLC-MSn analysis of root and rhizome materials from 18 Clematis samples as well as the whole plant materials of Clematis puberula var. ganpiniana and Clematis terniflora. The HPLC-MSn analysis allowing the detection of 17 oleanolic acid or hederagenin saponins was carried out in the negative selected ion monitoring (SIM) mode. Triterpenoid saponin profiles of these taxa were used for phylogenetic studies, and results are presented as a dendrogram. Huzhangoside B could be unambiguously identified in all analyzed Clematis taxa, as well as in the investigated Ranunculus taxa. The distribution and chemotaxonomic importance of the triterpenoid saponin profile within this genus are discussed.

Abstract:A new indole alkaloidal glucoside together with three known compounds aurantiamide acetate (2), eleutheroside E (3) and 1-O-caffeoyl-β-D-glucopyranoside (4) has been isolated from ethanol extract of the aerial parts of Clematis terniflora DC. On the basis of their spectroscopic and chemical evidence, the new compound was elucidated as (6-O-β-D-glucopyranosyl-1H-indol-3-yl) carboxylic acid methyl ester (1). Compounds 1 and 3 showed significant cytotoxicity against human ECA-109.

Abstract:Seven new pentacyclic triterpenoid saponins, named dianversicosides A-G (1-7), together with nine known compounds, were isolated from the aerial parts of Dianthus versicolor. The structures of 1-7 were elucidated on the basis of spectroscopic data and chemical evidence. The absolute configuration of the 3-hydroxyl-3-methylglutaryl (HMG) group in 1-4 was ascertained by chemical analysis combined with a chiral HPLC method. Cytotoxic activities of the isolated compounds were evaluated against a small panel of cancer and other cell lines.

Abstract:Three new triterpenoid saponins, heracleifolianosides A-C (1-3), together with seven known compounds (4-10), were isolated from the whole plants of Clematis heracleifolia. Moreover, three new secondary saponins (1a, 2a and 3a), two known secondary metabolites (5a and 7a) were obtained by alkaline hydrolysis. Their structures were elucidated by extensive spectroscopic analysis and chemical evidences. The cytotoxicity of eight native saponins and five prosapogenins against human breast tumor MDA-MB-231 and gastric carcinoma SGC-7901 cell lines were evaluated by MTT method. Remarkably, the prosapogenin monodesmosidic saponin 7a showed significant cytotoxicity against MDA-MB-231 or SGC-7901 cell lines with IC<sub>50</sub> values in the range of 6.05-6.32 μmol/L. It is suggested that it might be a feasible way to change the inactive bisdesmosic triterpenoid saponins to active monodesmosic saponins by a simple procedure of alkaline hydrolysis.

Abstract:Phytochemical investigation of the whole plants of Clematis tangutica led to the isolation of three new triterpenoid saponins (1–3), together with four known saponins (4–7). Their structures were determined by extensive spectral analysis and chemical evidences. Compounds 1–7 were evaluated for their cardioprotective activities in cardiomyocytes anoxia/reoxygenation (A/R) model. The results showed that those saponins exhibited cardioprotective effects by decreasing the levels of creatine kinase-MB (CK-MB) and lactate dehydrogenase (LDH).

Compounds 1–3 possess a novel 12β,30-dihydroxy-olean-28,13β-lactone aglycone and a 12β-hydroxy substituent that is rarely found in this kind of triterpenoid saponin.

Abstract:Gynostemma pentaphyllum (Thunb.) Makino (Cucurbitaceae family) is a perennial creeping plant with a common Chinese name of "south ginseng". To date, more than 250 individual saponins with dammarane-type skeleton have been isolated from G. pentaphyllum. The purpose of this study was the isolation and structural characterization of novel, minor gypenosides from G. pentaphyllum and evaluation of their Sirt1 agonist activity. Individual saponins from G. pentaphyllum were isolated and purified by a variety of chromatography techniques, and their structures were elucidated by means of various spectroscopic analysis and comparision with the reported data. Sirt1 enzyme activity detection kit was used to preliminarily evaluate the Sirt1 agonist activity of thirty three individual saponins purified from G. pentaphyllum. Fourteen new triterpenoid saponins named gypenoside CII-CXV (1–14) along with twenty six known compounds (15–40) were isolated from G. pentaphyllum. Thirty three of all the isolates were screened for Sirt1 agonist activity, and the results showed that three dammarane-type saponins (2, 18, 37) and one cucurbitane-type saponin 33 exhibited satisfactory Sirt1 agonist activity. These findings suggested that G. pentaphyllum was worthy of further investigation to find small molecule Sirt1 agonist and facilitate their utilization as "south ginseng".

Determination of Triterpenoid Saponins in Radix Clematidis and Related Clematis Species by High Performance Liquid Chromatography-Mass Spectrometry

Abstract:A simple and rapid gradient high performance liquid chromatography coupled with mass spectrometric method has been developed and validated for simultaneous determination of four triterpenoid saponins,i.e.,clematichinenoside B,huzhangoside D,clematichinenoside C and huzhangoside B in Radix clematidis and related Clematis species.The separation was performed on a Zorbax SB-C18 column with gradient elution using acetonitrile and 0.05%(V/V) aqueous formic acid as mobile phase.The flow rate was 0.6 mL/min and the column temperature was 25℃.Electrospray ionisation(ESI) mass spectra were acquired using negative ionization and selective ion monitoring.Under the optimized conditions,the analytes were well separated in 25 min.The results showed that the samples contains a wide range(0.0652-41.7mg/g) of the four analytes.The saponin content in Radix clematidis(Tianmushan,Zhejiang) was the highest(41.7 mg/g),which was about 7 times that in Radix clematidis(commodity)(5.80 mg/g).This might be due to different harvest times and places,suggesting that the source of Radix clematidis should be restricted.This method has good linearity,high sensitivity,precision,accuracy,and consumes short time,which is helpful in controlling the quality of traditional Chinese medicine derived from the genus of Clematis.This study may provide some scientific basis for development of new medicinal resources from Clematis species.
